Abstract

1309832 Substituted cyanamides ASPRONICHOLAS Ltd 19 April 1971 [14 Feb 1970] 7228/70 Heading C2C Novel 1,4-dicyano-piperazines of the general formula wherein R 1 and R 2 are hydrogen or (C 1 -C 4 ) alkyl, may be prepared by a process for producing substituted cyanamides wherein a secondary amine or a salt thereof is reacted with a cyanogen halide in the presence of ammonia or ammonium hydroxide. The reaction is preferably carried out at or below room temperature in an aqueous and/or organic solvent using a stoichiometric excess of the ammonia or ammonium hydroxide. The secondary amine is suitably of the formula wherein R is alkyl, cycloalkyl, hydroxyalkyl, alkoxyalkyl or aralkyl, R 1 is phenyl, alkyl, cycloalkyl, aralkyl or hydroxyalkyl, or R and R 1 together with the adjacent nitrogen atom represent an alicyclic ring optionally interrupted by O or N. The secondary amine is preferably a piperazine giving, as reaction product, the novel compounds. When this reaction product is reacted with cyanamide, preferably in the presence of a solvent and a basic catalyst, a 1,4-bis-cyanoamidino piperazine is obtained, which, when reacted with a substituted aniline or an acid addition salt thereof gives a biguanide of the general formula or an acid addition salt thereof wherein R 3 is halogen, alkyl or alkoxy.
