1301899 Antibiotic pyracrimycin A GRUPPO LEPETIT SpA 21 Dec 1970 60645/70 Headings C2A and C2C The antibiotic pyracrimycin A which is 1-pyrroline-2-acrylamide; also known as desdanine (see Specification 1,125,723) is produced by aerobically cultivating Streptomyces eridani nov. sp. A.T.C.C. 21619 in aqueous nutrient medium containing assimilable sources of carbon and nitrogen and inorganic salts. Pyracrimycin A is characterized as being a white crystalline substance melting at 215‹ to 216‹ C., molecular weight 138À17, analysis: C = 60À99, H = 7À50, N = 20À20, O (by difference) = 11À31, having a U.V. absorption peak in methanol at 235 mÁ (# = 22,000) and I.R. absorption peaks at 3250, 3050, 1670, 1620, 1590 and 972 cm<SP>-1</SP>. The antibiotic is isolated from the filtered fermentation broth by extracting with a water-immiscible solvent e.g. butanol, concentrating the extract and adding petroleum ether, when pyracrimycin A is precipitated in admixture with an antibiotically inactive metabolite, pyracrimycin B. The mixture is recrystallized from methanol and the solid is extracted with a hot chlorinated hydrocarbon solvent, e.g. chloroform, which dissolves out the inactive component leaving the purified pyracrimycin A. The antibiotic may be reduced to its novel antibiotically active dihydro derivative, 2- pyrrolidinacrylamide of formula by treatment with a metal borohydride (e.g. NaBH 4 ) in ethanol. Pharmaceutical compositions having antibiotic activity against Gram-positive and -negative micro-organisms, comprise pyracrimycin A (i.e. 1 - pyrroline - 2 - acrylamide) or dihydropyracrimycin A (i.e. 2-pyrrolidinacrylamide) together with a pharmaceutically acceptable carrier.
