Abstract

1288843 Enzymes PFIZER CORP 15 April 1970 [12 July 1969] 18070/70 Heading C3H [Also in Divisions C2 and C6] The isolation yield of fermentation product, such as antibiotic or penicillin acylase, from a fermentation broth is increased by introducing protease, cellulase or lysozyme and, optionally, a chelating agent e.g. ethylenediamine tetraacetic acid or tris(hydroxy-ethyl) amino-methane, into the broth before isolating the product. The following enzymes are mentioned: proteases including bromelain, chymotrypsin, ficin, papain, pepsin and trypsin; cellulases; and various lysozymes. The enzyme and chelating agent should each be added in concentrations of at least 10 p.p.m. According to one of the examples, lysozyme is added at a level of 100 p.p.m. to a whole culture fermentation broth of Proteus rettgeri ATCC 9918. Quantitative penicillin acylase present is determined by the addition of benzylpenicillin which is hydrolysed to 6-amino penicillanic acid. The broth is filtered, adjusted to pH 2À5-3À0 with dilute hydrochloric acid and extracted with methyl isobutyl ketone to remove unconverted benzylpenicillin. The pH of the aqueous 6 - aminopenicillanic acid - containing phase is raised to pH 7À5 and phenylacetyl chloride is added. After 50 minutes at room temperature with continuous agitation, the pH is adjusted to 6À5 and the broth is bioassayed for benzylpenicillin. Significantly higher levels of penicillin acylase were obtained with the lysozyme treated broth.
