1282378 Bacterial preparations and control specimens F HOFFMANN-LA ROCHE & CO AG 4 Dec 1970 [5 Dec 1969] 57728/70 Heading C6F Stable dry bacteriological control specimens have a generally disc-shaped configuration, comprise 10<SP>5</SP>-10<SP>8</SP> viable bacteria per disc suspended in nutrient gelatine and materials (specified) which stabilize the viability of the bacteria, and the discs are produced by vacuum desiccating discrete drops of the bacterial suspension on to a non-adhering surface. The discs are stored in small sterile screw-capped vials containing a desiccant e.g. silica gel, CaCl 2 or CaSO 4 . The preferred suspension medium contains 90-97% by weight nutrient gelatine, 0À005-0À015% diethylaminoethyl dextran (or dextran or dextran sulphate) and the remainder monosodium glutamate. The dextran and glutamate may, however, be replaced by glucose, sucrose, lactose, mannitol or polyvinylpyrrolidone. Other suspension media are comprised of 99.1- 99.5% of the nutrient gelatine and, as stabilizers, cysteine, thiourea, glutathione or monothioglycerol. In use, the discs may be added directly to the diagnostic medium whose biochemical properties are to be checked. Alternatively, the specimen micro-organisms may be re-cultivated prior to testing. According to an Example two discs, one with Staphylococcus aureus and the other with Staphylococcus epidermidis were placed in separate test tubes containing some of the rabbit plasma being used as a test medium in a coagulase test procedure. In three hours, the plasma clotted in the tube containing S. aureus while S. epidermidis gave a negative test, thereby confirming that the rabbit plasma was performing properly in the testing. The oxidase test of Pseudomonas aeruginosa and the raffinose test for Enterobacter cloacae are among the many-other tests and bacteria particularly mentioned.
