Abstract

1282376 Benzofuran derivatives J R GEIGY AG 19 Nov 1970 [20 Nov 1969] 54947/70 Addition to 1266766 Heading C2C Novel 2,3-dihydro-5-(2-methylene-valeryl)- 6,7.dimethyl-benzofuran-2-carboxylic acid is prepared by decomposition of compounds I in which Am is a secondary amino group, to effect cleavage of the base H-Am. The starting material may be prepared thus: 2,3-Dimethylphenol and malic acid are reacted with H 2 SO 4 to give 7,8-dimethylcoumarin, which is then reacted with Br 2 and the bromine derivative decomposed in ethanolic KOH to 6,7- dimethyl-benzofuran-2-carboxylic acid; the latter is then reduced to give the 2,3-dihydro-derivative which is reacted with AlCl 3 and valeryl chloride to give 2,3-dihydro-5-valeryl-6,7-dimethyl-benzofuran-2-carboxylic acid; the latter on reaction with formaldehyde and H-Am yields the starting material which is not isolated. Pharmaceutical compositions comprise the novel compound together with a suitable carrier and are administered orally as diuretics and saluretics.
