1,272,354. Arylsulphonyl ureas and aryl sulphonyl semicarbazides. FARBENFABRIKEN BAYER A.G. 6 Aug., 1970 [6 Aug., 1969; 4 June, 1970], No. 37983/70. Heading C2C. Novel arylsulphonyl ureas and arylsulphonylsemicarbazides of the general Formula I: wherein R is a C 1-10 alkyl or alkylene radical, a cyclo-, bicyclo-, tricyclo- or tetracyclo-alkyl optionally substituted with a C 1-4 , alkyl and which is directly bonded or bonded via an alkylene group to the NH'group, or an alkylene imino group optionally substituted by methyl groups and can be unsaturated or contain an endoalkylene bridge; R<SP>1</SP> and R<SP>11</SP> are the-same or different and may be hydrogen, halogen, alkyl or alkylene, C 1-6 alkoxy, trifluoromethyl, nitro or acylamino; R<SP>111</SP> is hydrogen, methyl or ethyl and X and Y are direct bonds or C 1-6 straight or branched alkylene groups and their salts with alkali or alkaline earth metals may be prepared by (a) reacting an amine RNH 2 with a sulphamide of the Formula (3) with elimination of HA, or with a sulphonyl isocyanate of the Formula (4): (b) reacting a sulphonamide of the general. Formula (5): or an alkali metal salt thereof with an amine derivative BCONHR or an isocyanate RNCO with elimination in the former case of HB or the alkalimetal salt thereof; (c): oxidative-hydrolysis of a compound (8): wherein Z is SH; or (d) acid or alkaline hydrolysis of (8) in which Z is amino, thioalkyi. or, chlorine. The intermediate compound (3) may be prepared by condensation of an acid chloride with an amine to form the amide group. The intermediate compound (5) may be prepared as for (3) above or by diazotization of a compound (9) in the presence of SO 2 Cu(II), halide: to form the sulphohalide followed by reaction with ammonia to form the sulphonamide group. The intermediate (9) may in turn be prepared. by reduction of a nitro. group to the amino group, the nitro compound in turn being prepared by condensation of an acid chloride with an amine to form the amide group. Pharmaceutical compositions of the compounds I show hyperglycemic activity when administered orally or parenterally with the usual excipients.
