Abstract
1,265,515. 19-Norsteroids. F. HOFFMANN.LA ROCHE & CO. A.G. 2 Oct., 1969 [4 Oct., 1968; 28 July, 1969], No. 45275/70. Divided out of 1,265,512. Heading C2U. The invention firstly comprises d-compounds of Formula Ia and also the l- and dl -modifications thereof [wherein V<SP>1</SP> is H or alkali metal, R<SP>1</SP> 2 is H, C 1-7 alkyl, (C 1-7 alkoxy)-(C 1-7 alkyl) or phenyl- (C 1-7 alkyl) and R 4 is H or C 1-7 alkyl] and their preparation from compounds of Formula IV [wherein V is H, alkali metal or C 1-7 alkyl or is phenyl optionally substituted by one or more C 1-7 alkyl, C 1-7 alkoxy, nitro, amino or halo groups] by (i) cyclization and (ii) hydrolysis of any esterified carboxylic group CO 2 V. Steps (i) and (ii) may be performed concurrently or sequentially depending on reaction conditions. Cleavage of any 17-ether group may also sometimes accompany steps (i) and (ii) or may be effected in a separate stage subsequently. The invention secondly comprises a process for the preparation of d-compounds of Formula Ib and also the l- and dlmodifications thereof from compounds of Formula Ia [wherein V<SP>1</SP> has the broader meaning defined for V] by decarboxylation, or directly from compounds of Formula IV by concurrent cyclization and de carboxylation. Cleavage of any 17-ether groupmay accompany these conversions. Optical resolution of racemic 17-hydroxy compounds may be effected at any stage during the above-described reaction sequences by separating the camphorsulphonates or by separating salts with optically active bases of 17-hemiesters. Compounds of Formula IV are prepared as described in Specification 1,265,516.
