Abstract

1,212,304. Polymyxin methane sulphonic acids. PFIZER CORP. 18 Feb., 1969, No. 8845/69. Heading C2A. Polymyxin methane sulphonic acids are prepared by reacting a polymyxin or salt thereof in aqueous solution with formaldehyde to give an N-methylol polymyxin which is reacted with sulphur dioxide to give the required sulphonic acid. An aqueous solution of the preferred polymyxin B or its sulphate is treated with gaseous or aqueous formaldehyde and the mixture is then neutralized to precipitate N-methylol polymyxin B, which is suspended in an inert medium (e.g. water or methanol) and treated, preferably at elevated temperature, with gaseous or aqueous sulphur dioxide. Polymyxin B methane sulphonic acid is formed, and precipitates when the inert medium is methanol or is recovered from aqueous medium by lyophilization. Pharmaceutical compositions having antibiotic activity against Gram negative bacteria comprise a polymyxin methane sulphonic acid together with a parenterally acceptable solvent or carrier.
