Abstract

1,176,854. 2-Amino-3,4-dihydroquinazolines. CHAS. PFIZER & CO., Inc. 20 Nov., 1967 [11 Oct., 1967], No. 52759/67. Heading C2C. The invention comprises compounds of formulµ and their acid addition salts, wherein X and Y are H, OH, halogen, CF 3 , C 1-4 alkyl or C 1-4 alkoxy, or X-Y is benzo, methylenedioxy or ethylenedioxy; R 1 and R 2 are H, C 1-6 alkyl, aryl, aralkyl, alkenyl or dialkylaminoalkyl, or R 1 -R 2 is C 3-7 cycloalkyl; R 3 is H, C 1-4 alkyl, aryl or aralkyl, and R 4 is morpholino, or 4- alkyl, 4-alkenyl or 4-(optionally substituted) phenylpiperazino. These compounds are prepared by (a) (where R 3 = H) reducing the corresponding 4-chloroquinazoline with H 2 /Pd-C, or (b) reducing the corresponding 4(3H)-quinazolinethione with Raney Ni/MeOH. Therapeutic compositions comprise the above compounds which may be administered orally or parenterally for reducing the blood pressure of hypertensive subjects, in combination with pharmaceutically-acceptable carriers. Alternatively, these compounds may be administered above.
