Abstract
1,172,919. Pancreatic heparinoids. MICRON DI BIANCHINI DOTTOR PIETRO & C.- S.N.C. 19 July, 1967 [22 July, 1966], No. 33071/67. Heading A5B. Pancreatic heparinoids are prepared by:- (a) subjecting mammal or fish pancreas to proteolysis by adding to the bulk or mass an aqueous alkaline solution of pH 7-8.5 comprising at least one antibiotic acting as bacterial floro inhibitor which is penicillin in an amount of 10,000-50,000 units of activity per kg of pancreas, or a tetracycline or chloramphenicol in an amount of 5-50 mg/kg of pancreas; (b) maintaining the suspension at 20-50‹C for 12- 48 hours out of contact with air; (c) heating at 70-100‹C for 10-45 minutes (d) separating the proteinous parts from the suspension; (e) subjecting the liquid obtained by separation; after adjusting it to a pH of 5-6.5 with an acid, to a treatment with a polar organic solvent in an amount of 2-5 vols/vol of liquid obtained and (f) allowing it to settle and collecting a hypophase having a high content of low molecular weight heparinoid. The alkaline solution of stage (a) may contain trypsin, chymotrypsin or papain. The proteolysis may be carried out in the presence of a reducing agent such as sodium hypophosphate or cysteine.
