Abstract

1,164,644. 2,2 - Ethylene - 3 - oxo - steroids. STERLING DRUG Inc. 17 Oct., 1966 [22 Oct., 1965], No.46383/66. Heading C2U. The invention comprises (1) a process for preparing a 2,2-ethylene-3-oxosteroid (the steroid moiety having from 17 to 23 carbon atoms exclusive of ester radicals and the substituent at the 2-position, and being a member of the estrane, 18-nor-estrane, androstane, etiocholane, cholane, pregnane or allopregnane series) which comprises treating with an acid or heating a corresponding spiro[steroid-2,3<SP>1</SP>(2<SP>1</SP>α)-1<SP>1</SP>-pyrazolin]-3-one or spiro[steroid-2,3<SP>1</SP>(2<SP>1</SP>α)-5<SP>1</SP>-pyrazolin]-3-one; and (2) steroids of the formula (wherein R is C 1-6 alkyl, alkenyl or alkynyl, acetyl, hydroxyacetyl, 1,2-dihydroxyethyl or 1-hydroxyethyl; X is H 2 , H(α- or #-OH) or O; X<SP>11</SP> is H, C 1-4 alkyl or halogen; Y and Y<SP>1</SP> are H or CH 3 ; and Z is H or OH, being #-OH when R is alkyl, alkenyl or alkynyl, and being α-H or α-OH, when R is acetyl, hydroxyacetyl, 1,2-dihydroxyethyl or 1 -hydroxyethyl; and there are optional double bonds in the 4,5- and/or 6,7- positions) and carboxylic esters thereof derived from C 1-12 acids, and 3-enol ethers and 3-enol esters thereof. The starting materials for process (1) above are prepared by the methods disclosed in Specifications 1,164,645, 1,164,646 and 1,164,647. Ketal groups may be protected by ketal formation and released by hydrolysis. Reference has been directed by the Comptroller to Specification 1,015,622.
