Abstract

1,156,844. Purification of nystatin. E. R. SQUIBB & SONS Inc. 14 July, 1966, No. 31728/66. Heading C2A. Nystatin is recovered from a nystatin fermentation broth by separating the mycelium from the broth, preferably washing with water, and treating the mycelium with an aqueous C 1-8 alkanol and with an acid and an amine, or the corresponding salt, at a substantially neutral pH and recovering the nystatin from the alcoholic extract. The pH is preferably 6À0 to 6À5. The alcoholic extract is concentrated to remove the organic solvent and the nystatin is recrystallized from aqueous acetone or other water-miscible ketone. The acid may be organic, e.g. oxalic, malonic, succinic or citric, but is preferably a mineral acid, e.g. sulphuric acid. The amine may be a primary, secondary or tertiary amine. The preferred solvent is methanol containing the sulphate salt of a tertiary amine such as triethylamine.
