Abstract

1,154,133. Helicocerin. SANKYO CO. Ltd. 12 Feb., 1968 [10 Feb., 1967], No. 6783/68. Heading C2A. The invention comprises an antibiotic Helicocerin, which is effective against yeasts, fungi and mycobacteria, which is soluble in methanol, ethanol, n-butanol, acetone, chloroform &c., but sparingly soluble in petroleum ether, nhexane and cyclohexane and insoluble in water; elementary analysis 64À36% C, 7À73% H and 6À43% N; empirical formula C 12 H 17 O 3 N; molecular weight 229À2; m.pt. 89-90‹ C.; No U-V spectrum; and IR-spectrum as in the figure. Hellicocerin is produced by the aerobic cultivation of a helicocerin-producing strain of Helicoceras oryzae, e.g. strain NI 4288 (Japanese Federation of Culture Collections), Helicoceras celtidis, Helicoceras plantaginis and Helicoceras nymphaerum. The clarified broth is extracted with a water-immiscible solvent such as methylene chloride, ethyl acetate, benzene, chloroform or carbon tetrachloride. After removing the solvent, the residue is treated with ethyl acetate and the extract is chromatographed on silica gel using ethyl acetate as eluant. After removal of solvent, the residue is dissolved in hot n-hexane and allowed to crystallize therefrom. A pharmaceutical composition comprises the antibiotic, helicocerin, and a carrier.
