Abstract
1,151,867. 3 - Oxo - 4,6 - dehydro - 19 - nor - steroids. GLAXO LABORATORIES Ltd. 17 June, 1966 [22 June, 1965], No. 26390/65. Heading C2U. 3 - Oxo - 4,6 - dehydro - 19 - nor - steroids are prepared by treatment of 3-oxo-5( 10),7-dehydrosteroids with an acid or a base. If a base is used a proportion of the novel 3-oxo-4,6-dehydro- 8α,9α,10α-19-nor-steroids are produced in addition to those of normal, i.e. 8#,9α,10#, configuration. They may be converted to the normal isomers by acid treatment. The starting materials may be prepared by reaction of 3 - hydroxy - 2,5(10),7 - dehydro - steroid 3 - ethers or 3 - oxo - 5(10),7 - dehydro - steroid 3-ketals with a weak acid. If a strong acid is used direct conversion to the 3-oxo-4,6-dehydro- 19-nor-steroids is achieved. Other groups in the molecule may be hydrolysed during these processes, e.g. a 17-ketal to a 17-oxo group. The products may be reduced to 3-oxo-4-dehydro- 19-nor-steroids by the action of metal-ammonia or metal-amine reducing systems; the preparation of 19-nor-testosterone by this method is described.
