Abstract

1,146,459. Nicotinic acid derivatives. AMERICAN CYANAMID CO. 7 Nov., 1967 [14 Nov., 1966], No. 50637/67. Addition to 1,070,620. Heading C2C. The invention comprises compounds of the Formula (I), wherein each R, the same or different, represents a trifluoromethyl, or C 1-4 alkyl radical or a halogen atom; n represents 1, 2 or 3; R 5 represents a C 1-4 alkyl radical and R 3 and R 4 represent C 1-4 alkyl or aryl radicals or R 3 and R 4 , taken together with the nitrogen atom to which they are attached, represent a pyrrolidino, piperidino, morpholino, 4-C 1-4 - alkyl - piperazino, 4 - (# - hydroxy - C 1-4 - alkyl)- piperazino or hydroxypiperidino group and their preparation by reacting a nicotinate ester of the Formula (II) with an unsymmetrical hydrazine of the Formula (III) in the presence of excess of the hydrazine as solvent or in the presence of an inert solvent. The invention also comprises N-methyl-4-(α,α,α -trinuoro-mtoluidino) nicotinamide which may be prepared by reacting methyl 4-(α,α,α-trifluoro-m-toluidino) nicotinate with methyl formamide and sodium methoxide. Therapeutic compositions in forms suitable for oral or parenteral application and having antiinflammatory, non-addicting analgesic and diuretic properties comprise compounds of the invention with a carrier.
