Abstract

1,139,097. 5<SP>1</SP>-Purine nucleotides. KYOWA HAKKO KOGYO CO. Ltd. 16 May, 1966 [18 May, 1965], No. 21559/66. Heading C2C. 5<SP>1</SP>-Purine nucleotides are prepared by culturing a micro-organism in the presenoe of a purine base in an aqueous nutrient medium under aerobic conditions, the micro-organism being of a strain which is not a purine base-requiring mutant strain but has the characteristics of being capable of (1) producing at least one of the compounds ribose-5-phosphate and 5-phosphoribosyl-1-pyrophosphate outside the cell walls thereof and (2) secreting the enzymes 5-phosphoribose pyrophosphokinase and 5<SP>1</SP>-purine nucleotide pyrophosphorylase from the cells thereof and recovering the product. The preferred strains of micro-organism include bacteria such as Bacillus megaterium, Arthrobacter ureafaciens, Micrococcus glutamicus, Corynebacterium rathayi, Brevibacterium ammoniagenes and Micrococcus sodonensis, moulds such as Aspergillus oryzae, Aspergillus sojae, Eremothecium ashbyii and penicillium chrysogenum, yeasts such as Saccharomyces cerevisiae, Torula utilis and Schizosaccharomyces pombe and actinomycetes such as Streptomyces fradiae and Streptomyces griseus. Examples are given for the preparation of 5<SP>1</SP>-inosinic acid, 5<SP>1</SP>-guanylic acid and 5<SP>1</SP>-adenylic acid and sodium salts thereof.
