Abstract
Vaccines against B. pertussis are prepared from isolated cell wall material from B. pertussis which has been treated in aqueous suspension with a C1-C5 water-miscible monohydric alcohol, aldehyde, or ketone at room temperature or below. The preferred agent is acetone, used at -20 DEG C for 18-24 hours. The bacteria are cultured on a conventional medium, harvested, suspended in cold water, and ruptured by explosive decompression using pressures of 15,000 to 50,000 p.s.i. The cell walls are sedimented in a centrifuge, washed, and treated with the above-mentioned agent, washed again, homogenised and suspended in cold distilled water, to which a preservative, such as thimerosal, benzethonium chloride, benzyl alcohol, or g -picolinium chloride may be added, to provide a vaccine. The vaccine may alternatively be an adjuvant vaccine, prepared by flocculating the distilled-water-suspended cell walls with alum, or by absorbing them on alumina, aluminium phosphate, or hydroxide. Also other antigens, such as diphtheria or tetanus, may be added to give polyvalent vaccines.
