Abstract
1a - N - phthalyl a -amino acid derivatives of mitomycin C of Formula II <FORM:1102259/C2/1> wherein R is H, CH3, <FORM:1102259/C2/2> C6H5CH2 or CH3-S-CH2 -CH2, are prepared by reacting N-phthalylamino acid halides of Formula III <FORM:1102259/C2/3> wherein R is the same as in Formula II and X is halogen, with mitomycin C in the presence of a base and an inert solvent. The derivatives of the following amino acids are disclosed: L-alanine, L-phenylalanine, L-leucine, glycine, L-valine, L-isoleucine and L-methionine, e.g. 1a-N-phthalyl alanyl mitomycin C. The compounds are effective anti-tumour agents. N-Phthalyl a -amino acids are prepared by reacting a -amino acids with phthalic anhydride in the presence of an inert solvent and a base. The a -amino acids are those specified above. The acid may subsequently be treated with a halogenating agent, e.g. PCl5, SOCl2 to obtain the corresponding acid halide.
