Abstract

A new antibiotic funginon, effective against gram-positive and -negative microorganisms and ascites tumor is produced by cultivating Aspergillus clavatus TPR-3202 (ATCC. 15550) or a funginon-producing mutant thereof in an aqueous nutrient medium containing assimilable C and N sources and inorganic substances. Funginon is recovered from the medium by adding diatomaceous earth, filtering, adding active C to the filtrate and then extracting the absobed funginon from the active C with 80% aqueous acetone, ethylacetate and ether after distillation at each stage, absorbing funginon on to a column of chromatographic active alumina, followed by elution with ethylacetate, which is distilled off, leaving a residual of pure funginon. Funginon, a yellow oily matter, is soluble in water, methanol, ethanol, acetone, ethyl acetate, butyl acetate, glacial acetic acid, ether, pyridine and dioxane, slightly soluble in chloroform and benzene, and insoluble in carbon tetrachloride, petroleum ether and hexane; it has the elemental analysis: C 53.34%, H 5.33%, O 41.31% (by difference), a mol. weight of 144 and optical rotation [a ]20D= +43.55 DEG (in water); it gives a brownish-black colour but no silver mirror by Tollens test, positive nitro-prusside, Molisch, Benedict, Nylander, diazo, Fehling and Nessler tests and negative FeCl3 and Biuret tests, a maximum U.V. absorption of E1% = 725 at 273-263 mm 1cm in methanol and I.R. absorption maxima when suspended in a KBr disc at 3380, 2930, 1775, 1750, 1680, 1450, 1403, 1375, 1250, 1220, 1173, 1075, 1035, 1005, 877, 829 and 785 cm-1. Funginon is stable at pH 3.0-7.0 but is unstable above pH 7.5 and on heating.
