Abstract
1,053,307. 1-Hydroxy-1,2-dihydro-1,3,5-triazines. UPJOHN CO. Feb. 26, 1964 [April 16. 1963], No. 8020/64. Heading C2C. The invention comprises compounds of the Formula I and their acid addition salts, wherein R 1 represents a hydrogen atom or a C 1-4 alkyl group, R 2 represents a hydrogen atom or a C 1-4 alkyl or piperidino group and R 3 represents a C 2-8 dialkylamino, dibenzylamino or bis-(2- hydroxypropyl) amino group or an N-aziridinyl, N-azetidinyl, N-pyrrolidinyl, piperidino, N- hexahydroazepinyl, heptamethyleneimino, oc. tamethyleneimino or morpholino group, each of the above heterocyclic radicals having optionally attached thereto 1 to 3 C 1-8 alkyl radicals, and compounds of the Formula IV wherein R 8 represents a C 2-8 alkenyl radical, R 9 represents a C 1-8 alkyl radical or a C 2-8 alkenyl radical and R 6 and R 7 represent C 1-4 alkyl or C 2-4 alkenyl radicals. Compounds of the Formulae I and IV are prepared by mixing triazines of the Formulae II and V respectively with a compound of the Formula III wherein X represents a halogen atom or a C 1-8 alkyl, C 1-8 alkoxy, or nitro group and n represents 0 to 5. Compounds of the Formula I wherein R 3 represents a C 2-16 dialkylamino group are prepared by hydrogenating compounds of the Formula IV in the presence of a hydrogenation catalyst. Compounds of the Formula I wherein R 1 and R 2 represent hydrogen atoms are prepared by reacting a cyanoguanidine of the Formula R 3 -C(: NH)NHCN with cyanogen bromide in the presence of a strong base and reacting the resulting dicyanoguanidine salt with a hydroxylamine acid addition salt. Triazines of the Formulae II and V may be prepared from cyanuric chloride by appropriate replacement of the three chlorine atoms by primary, secondary and tertiary amino groups. Therapeutic compositions with anti-hypertensive, vasodilatory, antisecretory and central nervous system depressant properties in the forms of tablets, capsules, powders, pills, granules, syrups, elixirs, suppositories, solutions, and dispersions, for oral, topical and parenteral application, comprise compounds of the Formula I and acid addition salts thereof together with a pharmaceutically acceptable carrier.
