Abstract

<PICT:1041105/C6-C7/1> Apparatus for the anaerobic cultivation of bacteria consists of a main container 1 for the culture and nutrient medium, and a smaller container 2, containing a wad 8 impregnated with alkaline pyrogallol, rigidly carried by a side tube 3 which extends laterally from adjacent to the top of the main container 1 and is plugged with a wad of sterile non-absorbent filter material (cotton wool). The main container is shown supported in a test tube rack 6; however, the container may take the form of a conical flask. The two containers and the side tube are preferably made of glass and the side tube may contain a ground glass fitting so that the containers can be disconnected one from the other. The open tops of the two containers may be sealed with stoppers, bungs or ground glass rotary taps. The apparatus may be employed for the anaerobic cultivation of De-sulphovibrio desulphuricans, a sulphate-reducing organism used in the gas industry to test the potentiality for hydrogen sulphide production in stored gas and distribution systems. Sodium thioglycollate is added to the culture medium to poise the redox potential. It is stated that were residual oxygen to be removed catalytically by sparking, the hydrogen sulphide produced by the bacteria would soon poison the catalyst.
