Abstract
Compounds of the formula Ar-A-NH-C(: NH)NH2 and pharmaceutically acceptable salts thereof, wherein A is <FORM:1040542/C2/1> or (CH2)n, R1 being H or C1- 5 alkyl R2 and R3, which may be the same or different, being H, OH or C1- 5 alkyl, and n being 3-6, and Ar is a phenyl residue optionally substituted by one or more OH, C1- 5-alkyl or -alkoxy, or trifluoromethyl groups or halogen atoms, with the proviso that, when R1 is H, Ar is not an unsubstituted or 2-monosubstituted phenyl radical or a 4 - hydroxy-, 4 - methoxy- or 3,4 - dihydroxyphenyl radical, and, when R1 is methyl or when R2 and R3 are both H, Ar is not an unsubstituted phenyl radical, are prepared by reacting an amine of the formula Ar-A-NH2 with a salt of an S-methylisothiourea or with a cyanamide, dicyanamide, another S-alkylisothiourea, an O-alkylisourea, or thiourea, and a desulphurizing agent. 2,6-Dichlorophenethylamine is prepared by reduction of 2,6-dichloronitrovinylstyrene. Pharmaceutical compositions comprise the guanidines (I) and a carrier therefor. The compositions, which exhibit diuretic, natriuretic, hypotensive and hypoglycemic activity, are administered enterally or parenterally.
