Abstract

The invention comprises derivatives of 1,2,4-benzothiadiazine-1 1-dioxide of the formulae <FORM:1009013/C2/1> and <FORM:1009013/C2/2> wherein R1 is a phenyl group optionally bearing a halogen or alkyl substituent, or a cycloalkyl group, R2 is an alkyl, cycloalkyl or aralkyl group and B is a hydrogen or chlorine atom. Compounds of Formula I are prepared by reacting an o-sulphamoyl aniline derivative with a carboxylic acid R2.COOH or a reactive derivative thereof such as the halide, preferably in the presence of a strong acid as a condensing agent. Compounds of the Formula II are made by reacting an o-sulphamoyl aniline derivative with an aldehyde R2.CHO or a reactive derivative thereof, or with a compound of the formula <FORM:1009013/C2/3> wherein X and Y are piperidino or morpholino radicals or together with the carbon atom form an imidazoline ring (as in 1,3-diphenyl-2-cyclohexyl imidazoline; or alternatively compounds of the Formula II may be prepared by reduction of the corresponding compounds of the Formula I, e.g. by catalytic hydrogenation, by reaction with a mixed hydride reducing agent such as lithium aluminium hydride or electrolytically. Pharmaceutical preparations having hypnotic and tranquillizing properties comprise the above benzothiadiazines in association with a carrier or excipient. The preparations may take forms adapted to oral, rectal or parenteral administration, e.g. tablets, cachets, capsules, dragees, solutions, suspensions, emulsions or suppositories.
