La présente invention concerne le domaine des solvants eutectiques naturels, permettant la solubilisation et l’extraction de composés d’origine naturelle (par exemple d’origine végétale), ainsi que l’obtention d’extraits (par exemple d’extraits végétaux) à partir de matière(s) première(s) d’origine naturelle (par exemple à partir de matière(s) première(s) d’origine végétale, fongique, animale et/ou microbienne).The present invention relates to the field of natural eutectic solvents, allowing the solubilization and the extraction of compounds of natural origin (for example of vegetable origin), as well as the obtaining of extracts (for example of vegetable extracts) at from raw material(s) of natural origin (for example from raw material(s) of plant, fungal, animal and/or microbial origin).

Dans le domaine de l’extraction à l’échelle industrielle (mise en œuvre, par exemple, dans les industries chimique, alimentaire, pharmaceutique, de cosmétique et parfumerie, phytopharmaceutique), et en particulier de l’extraction végétale, des solvants organiques issus de la pétrochimie (dits « solvants d’origine pétrochimique » ou « Composés Organiques Volatiles (COV) ») sont largement utilisés pour l'extraction de composés, principes actifs ou substances à partir de matière(s) première(s) d’origine végétale. Selon la nature des composés, principes actifs ou substances ciblés, des solvants organiques, tels que les alcools, l'hexane et l'acétate d'éthyle, sont généralement utilisés dans les procédés d’extraction. En particulier en raison des problématiques susvisées, des réglementations nationales et supranationales contraignantes (telles que la directive 1999/13/CE, relative aux émissions de COV, la directive 67/548/CEE, relative aux substances dangereuses, la directive 1999/45/CE, relative aux produits dangereux et, depuis le 1erjuin 2007, le règlement européen REACH (« Registration, Evaluation, Autorisation and Restriction of Chemicals », en langue anglaise)) sont venues encadrer, de manière stricte, l’utilisation des solvants d’origine pétrochimique, conduisant au besoin de substituer lesdits solvants d’origine pétrochimique par des solvants d’extraction alternatifs renouvelables et faiblement toxiques (voire dépourvus de toxicité), communément dénommés « solvants verts ». De surcroît, dans certains domaines, comme par exemple celui des mélanges liquides conditionnés en ampoules ou unidoses, la réglementation s’avère encore plus contraignante concernant le solvant utilisé qui fait partie du produit final consommé. Par ailleurs, en ce qui concerne les produits sous forme solide/sèche issus du séchage d’extraits végétaux, l’ajout d’agent(s) technique(s) s’avère généralement nécessaire en fin de procédé, notamment pour faciliter le séchage de l’extrait et/ou suite audit séchage, avant de réaliser l’étape - ou les étapes - de formulation à proprement parler. Par exemple, la préparation de comprimés à partir de produits sous forme solide issus du séchage d’extraits végétaux nécessite traditionnellement l’ajout d’agent(s) technique(s) - et plus précisément d’agent(s) de compression - avant de réaliser l’étape de compression. Les agents techniques (par exemple un ou plusieurs agents de compression) ainsi utilisés sont plus ou moins naturels, ce qui, le cas échéant, peut donner lieu à des problèmes de compatibilité avec les réglementations idoines en matière de produits biologiques. En outre, l’ajout de tel(s) agent(s) technique(s) avant l’/les étape(s) de formulation tend à complexifier le procédé de formulation dans son ensemble, ce qui est notamment susceptible d’engendrer un surcoût non négligeable. Furthermore, with regard to products in solid/dry form resulting from the drying of plant extracts, the addition of technical agent(s) is generally necessary at the end of the process, in particular to facilitate drying. of the extract and/or following said drying, before carrying out the stage - or stages - of formulation strictly speaking. For example, the preparation of tablets from products in solid form resulting from the drying of plant extracts traditionally requires the addition of technical agent(s) - and more precisely of compression agent(s) - before to perform the compression step. The technical agents (for example one or more compression agents) thus used are more or less natural, which, if necessary, can give rise to problems of compatibility with the appropriate regulations in terms of biological products. In addition, the addition of such technical agent(s) before the formulation step(s) tends to complicate the formulation process as a whole, which is in particular likely to generate a significant overhead. Indeed, the person skilled in the art does not have, in the current state of knowledge, an extraction solvent which would integrate at least one technical agent, such as a compression agent, which, once the solvent has dried , would make it possible to obtain a solid dosage form—such as a tablet—having satisfactory pharmaco-technical properties.
Confronté aux problématiques susvisées, l’homme du métier s’est naturellement tourné vers l’utilisation de l’eau comme solvant pour réaliser l’extraction de composés d’origine naturelle (et en particulier de composés bioactifs). En effet, l’eau demeure le solvant le plus sûr (à savoir le moins toxique) et le moins onéreux. Néanmoins, son caractère très polaire limite son pouvoir de solubilisation aux composés naturels polaires (hydrophiles), excluant de facto les composés naturels peu polaires (également dénommés « composés à polarité intermédiaire ») et apolaires. De surcroît, si l’on s’intéresse à l’extraction de composés bioactifs à partir de matière(s) première(s) végétale(s) (matériel végétal), l’homme du métier sait que l’efficacité du recours à l’eau comme solvant d’extraction sera inéluctablement affectée par la mauvaise diffusion de l’eau dans le tissu végétal. Par exemple, dans le cas de plantes contenant des lipides ou des molécules apolaires ou à polarité intermédiaire, l’eau ne peut que difficilement - voire très difficilement - mouiller la surface de la plante et pénétrer au sein du tissu végétal, en raison de la non-miscibilité et la non solvatation des molécules apolaires présentes en grande quantité dans la plante. Enfin, le fait d’utiliser l’eau comme solvant d’extraction ne permettrait pas de disposer d’un réel support pour réaliser la compression de la matière sèche obtenue par séchage de l’extrait correspondant, dans une optique de production de comprimé.Faced with the above-mentioned problems, those skilled in the art naturally turned to the use of water as a solvent to carry out the extraction of compounds of natural origin (and in particular of bioactive compounds). For example, in the case of plants containing lipids or molecules that are apolar or of intermediate polarity, it is difficult - or even very difficult - for water to wet the surface of the plant and penetrate within the plant tissue, due to the non-miscibility and non-solvation of apolar molecules present in large quantities in the plant. Finally, the fact of using water as an extraction solvent would not make it possible to have a real support to carry out the compression of the dry matter obtained by drying of the corresponding extract, with a view to producing tablets.
Dans cette recherche d’alternatives aux solvants d’extraction d’origine pétrochimique, d’autres solvants d’extraction - et catégories de solvants d’extraction - ont été explorés. A cet égard, il est possible de citer, par exemple, la demande PCT publiée sous la référence WO 2018/122514 A1, laquelle divulgue l’utilisation de l’eau de coco (dont le degré Brix est compris entre 4 et 6, à 20°C) comme solvant d’extraction et les extraits obtenus par extraction avec cette eau de coco. Il est également possible de faire état d’une célèbre famille de solvants, à savoir les liquides ioniques (LIs), consistant en des sels liquides se différenciant de l’ensemble des sels fondus par une température de fusion inférieure à 100°C [1]. Un grand nombre d’entre eux sont liquides à température ambiante. Ces liquides ioniques sont constitués d’un cation, le plus souvent organique, associé à un anion organique ou inorganique. Ils ont été découverts pour la première fois par Paul Walden en 1914 dans le cas du nitrate d'éthylammonium, obtenu par neutralisation de l'éthylamine avec de l'acide nitrique concentré. Bien que les LIs présentent des propriétés intéressantes (ininflammabilité, stabilité thermique, faible pression de vapeur, etc.), ils ne peuvent toutefois pas être considérés comme de véritables solvants d’extraction « verts » notamment en raison de leur faible biocompatibilité et biodégradabilité, ainsi que de leur toxicité [2] [3].In this search for alternatives to extraction solvents of petrochemical origin, other extraction solvents - and categories of extraction solvents - have been explored. In this regard, it is possible to cite, for example, the PCT application published under the reference WO 2018/122514 A1, which discloses the use of coconut water (whose Brix degree is between 4 and 6, at 20°C) as extraction solvent and the extracts obtained by extraction with this coconut water. It is also possible to mention a famous family of solvents, namely ionic liquids (ILs), consisting of liquid salts differing from all molten salts by a melting temperature below 100°C [1 ]. Many of them are liquid at room temperature. These ionic liquids consist of a cation, most often organic, associated with an organic or inorganic anion. They were first discovered by Paul Walden in 1914 in the case of ethylammonium nitrate, obtained by neutralization of ethylamine with concentrated nitric acid. Although LIs have interesting properties (non-flammability, thermal stability, low vapor pressure, etc.), they cannot however be considered as true "green" extraction solvents, in particular because of their low biocompatibility and biodegradability, as well as their toxicity [2] [3].
Depuis le début des années 2000, des solvants eutectiques profonds (SEPs ; « Deep Eutectic Solvents » en langue anglaise, dont l’acronyme est DESs) ont été développés. Généralement définis comme une sous-catégorie des LIs ([4] et [5]), ces SEPs consistent en des mélanges d'au moins deux espèces qui s'associent entre elles par des liaisons intermoléculaires non-covalentes (en particulier par des liaisons hydrogènes). Ces interactions entraînent une diminution énergétique caractérisée par la température de fusion du mélange. On utilise la dénomination « mélange eutectique » lorsque le point de fusion du mélange (ou point eutectique), à un ratio molaire bien déterminé, est inférieur à celui des deux composés du mélange pris individuellement [6].Since the early 2000s, deep eutectic solvents (SEPs; “Deep Eutectic Solvents” in English, whose acronym is DESs) have been developed. These interactions lead to an energy decrease characterized by the melting point of the mixture. The term “eutectic mixture” is used when the melting point of the mixture (or eutectic point), at a well-determined molar ratio, is lower than that of the two compounds of the mixture taken individually [6].
Les propriétés des SEPs sont similaires à celles des Lis (ou voisines de celles-ci) ; les avantages principaux inhérents aux SEPs étant leur facilité de préparation et le faible coût de leurs constituants. Parmi ces SEPs, on peut citer le chlorure de choline, qui joue le rôle d’accepteur de liaison hydrogène (ALH), et permet de former un mélange eutectique avec la plupart des donneurs de liaison hydrogène (DLH). Ainsi, il est possible de citer un SEP formé par le mélange du chlorure de choline et de l'urée (dans un ratio molaire 1/2), lequel fut historiquement un des premiers SEPs mis en évidence.The properties of SEPs are similar to those of Lis (or close to them); the main advantages inherent in SEPs being their ease of preparation and the low cost of their constituents. Among these SEPs, we can cite choline chloride, which acts as a hydrogen bond acceptor (ALH), and makes it possible to form a eutectic mixture with most hydrogen bond donors (DLH). Thus, it is possible to cite a SEP formed by the mixture of choline chloride and urea (in a 1/2 molar ratio), which was historically one of the first SEPs demonstrated.
Toutefois, et bien que les SEPs aient déjà été testés en vue d’extraire des molécules d’intérêt à partir de matière(s) première(s) végétale(s) (par exemple pour extraire la quercétine et le kaempférol, cf. [7]), il convient de noter que la viscosité de ces SEPs (et en particulier de ceux comprenant du chlorure de choline comme ALH) demeure problématique pour l’extraction naturelle, et en particulier l’extraction végétale, au point que certains systèmes requièrent l’ajout d’additif(s) tel(s) que de l’eau pour être utilisés. A cela s’ajoute l’absence de preuve établie concernant la non-toxicité des SEPs en comparaison aux solvants LIs « traditionnels » dont le profil toxicologique a largement été interrogé par le passé. En outre, les SEPs comprennent des composés ALHs (tels que le chlorure de choline) et des composés DLHs obtenus par chimie de synthèse, ce qui ne permet pas de garantir l’innocuité (ou à tout le moins la compatibilité avec les réglementations idoines en matière de produits biologiques, cf. notamment règlement CE n° 889/2008 du 5 septembre 2008 - portant sur les modalités d'application du règlement (CE) n° 834/2007 du Conseil relatif à la production biologique et à l'étiquetage des produits biologiques en ce qui concerne la production biologique, l'étiquetage et les contrôles - et ses modifications/rectifications, ainsi que le nouveau règlement (UE) 2018/848, entré en vigueur le 1erjanvier 2021), des composés et/ou des extraits obtenus à partir de matière(s) première(s) naturelle(s) (par exemple de matière(s) première(s) végétale(s)) par extraction au moyen de ces SEPs.However, and although SEPs have already been tested with a view to extracting molecules of interest from plant raw material(s) (for example to extract quercetin and kaempferol, cf. [ 7]), it should be noted that the viscosity of these SEPs (and in particular of those comprising choline chloride such as ALH) remains problematic for natural extraction, and in particular plant extraction, to the point that certain systems require adding additive(s) such as water for use. Added to this is the absence of established proof concerning the non-toxicity of SEPs in comparison with “traditional” LI solvents, the toxicological profile of which has been widely questioned in the past. In addition, SEPs include ALHs compounds (such as choline chloride) and DLHs compounds obtained by synthetic chemistry, which does not guarantee safety (or at the very least compatibility with the appropriate regulations in with regard to organic products, see in particular EC regulation no. organic products with regard to organic production, labeling and controls - and its modifications/rectifications, as well as the new Regulation (EU) 2018/848, which entered into force on 1 January 2021), compounds and/or extracts obtained from natural raw material(s) (for example vegetable raw material(s)) by extraction using these SEPs.
C’est pourquoi, ces dernières années, une attention particulière a été accordée aux SEPs constitués uniquement de molécules naturelles. L’acronyme « NADES » (pour « Natural Deep Eutectic Solvents », dont la traduction en langue française est « solvant eutectique profond d’origine naturelle » (SEPNa)) a été proposé dans la demande PCT publiée sous la référence WO 2011/155829 A1). WO 2011/155829 A1 divulgue notamment un procédé d’extraction réalisé à partir d’une matière biologique (par exemple d’origine végétale), mis en œuvre au moyen d’un agent d’extraction consistant en un solvant eutectique profond d'origine naturelle ou un liquide ionique d'origine naturelle. Selon un mode de réalisation, ledit solvant eutectique profond est défini comme comprenant la combinaison d’au moins deux composés, sensiblement sans liaison chimique ou ionique, à savoir :This is why, in recent years, particular attention has been paid to SEPs consisting solely of natural molecules. - un acide organique naturel ou un composé inorganique, et- a natural organic acid or an inorganic compound, and
- un sucre mono- ou dimérique naturel, un polyol, un acide aminé, un di ou trialcanol, ou la choline ou ses dérivés, tels que la phosphatidylcholine.- a natural mono- or dimeric sugar, a polyol, an amino acid, a di or trialkanol, or choline or its derivatives, such as phosphatidylcholine.
Différents NADES et/ou procédés d’extraction incluant ces NADES ont été développés par la suite. A titre d’exemple, l’on peut citer l’enseignement de la demande PCT publiée sous la référence WO 2016/162703 A1. Cette dernière concerne un solvant d'extraction eutectique pour extraire, par exemple, un matériel biologique végétal, animal et/ou procaryote, ledit solvant d’extraction eutectique étant un mélange, décrit comme transparent, stable et fluide, comprenant :Different NADES and/or extraction processes including these NADES have subsequently been developed. Plus récemment encore, de nouveaux SEPs ont été décrits. L’acronyme « SUPRADES » pour « Supramolecular Deep Eutectic Solvents », dont la traduction en langue française est « Solvants eutectiques profonds supramoléculaires » (SEPsSup), a été proposé en 2020 (cf. [8]). Ces SUPRADES sont obtenus en mélangeant un DLH avec un ALH (l’ALH étant une cyclodextrine). Ces mélanges à faible température de fusion (par exemple à température ambiante) sont décrits comme possédant une structure supramoléculaire. D’après les auteurs, les SUPRADES ainsi obtenus permettraient de maintenir les propriétés encapsulantes des cyclodextrines composant ces SEPs. Rapidement ensuite, la même équipe de recherche a découvert de nouveaux SUPRADES élaborés à partir de diverses cyclodextrines modifiées chimiquement (cf. [9]). D’après ces auteurs, de tels SUPRADES permettraient de solubiliser le trans-anéthole d’un facteur plus de 1000 fois supérieur à l’eau. Il est important de retenir que ces SUPRADES sont obtenus en mélangeant uniquement un DLH (par exemple l’acide lévulinique) avec uniquement un ALH (en l’espèce une cyclodextrine). Ces mélanges sont donc constitués de 2 molécules (mélanges binaires) entre lesquelles un nombre bien défini et limité de liaisons hydrogène s’établissent. Enfin, ces SUPRADES basés sur l’utilisation de cyclodextrines, bien qu’étant présentés comme des « solvants verts », sont susceptibles de ne pas être conformes aux réglementations idoines en matière de produits biologiques, dans la mesure où les cyclodextrines elles-mêmes ne sont pas conformes aux réglementations idoines en matière de produits biologiques.Even more recently, new SEPs have been described. According to the authors, the SUPRADES thus obtained would make it possible to maintain the encapsulating properties of the cyclodextrins composing these SEPs. Soon after, the same research team discovered new SUPRADES made from various chemically modified cyclodextrins (cf. [9]). According to these authors, such SUPRADES would make it possible to solubilize trans-anethole by a factor of more than 1000 times greater than water. It is important to remember that these SUPRADES are obtained by mixing only a DLH (for example levulinic acid) with only an ALH (in this case a cyclodextrin). These mixtures therefore consist of 2 molecules (binary mixtures) between which a well-defined and limited number of hydrogen bonds are established. Finally, these SUPRADES based on the use of cyclodextrins, although presented as "green solvents", are likely not to comply with the appropriate regulations for biological products, insofar as the cyclodextrins themselves do not do not comply with the relevant regulations for organic products.
A la lumière de ce qui précède, il existe donc un besoin important de concevoir et mettre au point de nouveaux SEPNa :In light of the foregoing, there is therefore a significant need to design and develop new SEPNas:
