Claims (9)
- REVENDICATIONS1- Procédé pour induire des recombinaisons méiotiques ciblées dans une cellule eucaryote comprenant - l'introduction dans ladite cellule : a) d'une protéine de fusion comprenant un domaine Cas9 et un domaine Spo 1 1 ou d'un acide nucléique codant ladite protéine de fusion ; et b) d'un ou plusieurs ARN guides ou d'un ou plusieurs acides nucléiques codant lesdits ARN guides, lesdits ARN guides comprenant une structure ARN de fixation au domaine Cas9 de la protéine de fusion et une séquence complémentaire de la région chromosomique ciblée; et - l'induction de l'entrée en prophase 1 de méiose de ladite cellule et dans lequel ladite cellule eucaryote est une cellule de levure, de plante ou de champignon.
- 2. Procédé selon la revendication 1, dans lequel la protéine de fusion comprend en outre une séquence signal de localisation nucléaire.
- 3. Procédé selon l'une quelconque des revendications précédentes, dans lequel le domaine Cas9 est une protéine Cas9 déficiente pour l'activité nucléase.
- 4. Procédé selon l'une quelconque des revendications précédentes, dans lequel l'expression de l'acide nucléique codant ladite protéine de fusion est placée sous le contrôle d'un promoteur constitutif, inductible ou spécifique de la méiose.
- 5. Procédé selon l'une quelconque des revendications précédentes, comprenant en outre l'introduction d'un ou plusieurs ARN guides supplémentaires ciblant une ou plusieurs autres régions chromosomiques, ou acides nucléiques codant lesdits ARN guides supplémentaires.
- 6. Procédé selon l'une quelconque des revendications précédentes, dans lequel la cellule est une levure.
- 7. Procédé selon la revendication 6, dans lequel l'induction de l'entrée en prophase 1 est obtenue par transfert de la levure d'un milieu riche à un milieu de sporulation.. Protéine de fusion telle que définie dans l'une quelconque des revendications précédentes. 9. Acide nucléique codant pour la protéine de fusion selon la revendication
- 8. 10. Cassette d'expression ou vecteur comprenant un acide nucléique selon la revendication
- 9. 11. Vecteur selon la revendication 10, ledit vecteur étant un plasmide comprenant - une origine de réplication bactérienne, de préférence l'origine ColEl. - une cassette d'expression comprenant un acide nucléique tel que défini dans la revendication 9 sous le contrôle d'un promoteur d'expression, de préférence le promoteur ADH1, et un terminateur, de préférence le terminateur ADH1' - un ou plusieurs marqueurs de sélection, de -préférence des marqueurs de résistance tels que le gène de résistance à la kanamycine ou à l'ampicilline, et/ou - une ou plusieurs séquences permettant l'insertion ciblée du vecteur, de la cassette d'expression ou de l'acide nucléique dans le génome d'une cellule hôte, de préférence au niveau du locus TRPI du génome d'une levure. 12. Vecteur selon la revendication 11, comprenant, ou consistant en, une séquence nucléotidique sélectionnée parmi la SEQ ID NO: 1 et la SEQ ID NO :
