Abstract

L'invention concerne l'utilisation d'une matrice de collagène réticulée poreuse, principalement composée de collagène de type II et de glycosaminoglycanes, comme support à la culture cellulaire in vitro de chondrocytes destinée à être implantée pour réparer les défauts localisés du cartilage et douée d'une stabilité mécanique et dimensionnelle en culture. Elle est composée de collagène de type II et de glycosaminoglycanes. Elle est réticulée chimiquement. Le procédé de préparation de la matrice comprend une étape de mélange du collagène II et du glycosaminoglycanes, une étape de lyophilisation, une étape de réticulation chimique et une étape de stérilisation ; la réticulation chimique de la composition de collagène II et de glycosaminoglycanes est effectuée à l'aide de N-hydroxysuccinimide (NHS) et d'éthyl-3(3-diméthylaminopropyl) carbodiimide (EDC). Cette matrice sert à la culture de chondrocytes autologues et plus particulièrement à la conservation des chondrocytes dans les pores de la matrice grâce à la taille et à la stabilité de son maillage.
