Abstract

Microgranules for oral admin. to man or animals liberate naftidofuryl oxalate (I) in a controlled manner. Liberation rate of (I) is as follows: less than 20 percent after 1 hour, 30-50 percent after 4 hours, and over 80 percent after 8 hours. This liberation is independent of pH. The microgranules comprise the following: an inert spherical support 500-600 microns in dia., a deposit of 0.338-0.413 mg of (I) fixed to the support, and a coating over the deposit of (I) contg. a methacrylic acid anionic polymer, the coating being 4-7 wt.% of the uncoated material. (I) is known to have circulatory and metabolic activating activity, and is useful in the treatment of arteriopathies, circulatory disorders, cerebral vascular disorders and circulatory insufficiency. The controlled release of (I) gives reproducible results, liberating large amts. of (I) at a precise area in the stomach or small intestine.
