Abstract

Oral galenical formulation comprises (a) miniaturised granules prod. under high or very high compression, having a central core convered by several layers contg. >=1 active ingredient (I) plus an active excipient contg. a physiologically active neutralisation agent for pH control. Layers contg. (I) are sepd. from each other by layers of excipient which determin the slow penetration of digestive liq. Similar miniaturised granules contg. >=1 (I) coated with excipients or a very thin layer of lipid (representing 5-30% relative to (I)) are also claimed. The compsn. may contain 0-100% of each type of granule depending on the required absorption pattern, and pref. the granules have been compresed under >=10 tonnes pressure and have granule dia. 0.2-2 mm. The pref. excipients are PVP, cellulose, ethylcellulose, methacrylates, acetophthalates and gum lac. The release of (I) can be programmed and regulated as needed and absorption controlled regardless of whether (I) is normally slow or rapidly absorbed. Also therapeutic effect is improved, and the size of the formulation and the amt. of lipid needed for potentiation are both reduced.
