Abstract

Azapurinones are of formula (I) and salts. In the formula, R8 is 1-6C opt. branched alkoxy. R9 is CN, -NHCR12= CR10R11, -N=N-CHR13R14, -CONR15R16, -NH.C(Y1)NHR15 or (II) R10, R13 and R14 are CN, or 2-6C opt. branched alkanoyl or alkoxycarbonyl R11 is H, CN, 2-6C opt. branched alkanoyl or alkoxycarbonyl R12 is H or, when R11=H it can be opt. branched 1-6C alkyl or 2-6C alkoxycarbonyl; or R11+R12= (CqH2q)- q = 3-6, R15 and R16 are H, opt. branched 1-6C alkyl, Y1 is O or S. (I) are used for treating respiratory disorders caused by reaction of antibodies fixed to tissues with specific antigens, e.g. allergic bronchial asthma. They have weaker emetic effects than known similar cpds. and their acute LD50 values in mice were all >1g/kg orally and 220-470 mg/kg. i.v. The usual adult dose is 0.02-1.6 mg/kg/day by inhalation; or 4-120 mg/kg/day orally.
