Abstract
Benzisotheriazolones of formula (I) and their acid addn. salts when a basic N is present, are new. In the formula R1 and R2 are H, halo, 1-6C alkyl alkoxy, haloalkyl or alkanoylamino, NO2 or NH2 or together on adjacent C they are 3-6C alkylene or 1-3C oxyalkyleneoxy. X is 1-12C opt. branched alkylene, R3=H 1-6C alkyl, (opt. substD. by aryl or OH), 2-6C alkenyl, aryl or N-heterocyclyl, R4=1-6C alkyl (opt. substd. by aryl or OH), 2-6C alkenyl or aryl, or if R3 is not = H, also N-heterocyclyl. R3 and R4 are not both H or lower alkyl when R1 and R2 at positions 5 and 7 are H or halo, and X is lower alkylene. They are prepd. e.g. by reacting the corresp. N-H cpd. with Q.X.NR3R4(Q=easily eliminated gp. (I) are thrombocyte aggregation inhibitors so are useful for preventing or treating thrombosis in humans or animals. The pref. dose for adult humans is 10-500 mg orally 1-3 times per day. (I) can also be used to conserve blood in vitro, pref. at 0.01-50 mu g/ml.
