Abstract

Nitroindanediones of formula (I) and their acceptable salts are new (R1 and R2=H, lower alkyl, alkenyl, alkynyl or alkoxy; or R1+R2 on adjacent C complete a 3-5C alkylene including 1,4-buta-1,3-dienylene. A is a gp. X bond or O. Q=1-9C linear alkylene opt. substd. by one OH on a C atom which is not bonded to the ether O. R3, R4, R5, R6 and R7 are each H, halo, NO2, OH, CN, COOH, NH2 (opt. substd. by 1 or 2 alkyl or 1 or 2 acyl), alkyl alkenyl, alkynyl, alkoxy, alkoxycarbonyl, acyl, acyloxy, phenyl, alkylphenyl, phenoxycarbonyl or benzyloxycarbonyl; or 2 of them on adjacent C complete a 3-5C alkylene including 1,4-buta-1,3-dienylene. All alkyl etc. are 'lower'). Also new are intermediates of formula (IV), (V) and (VIII) (I) directly antagonise the slow-reaction anaphylaxis substance and inhibit release of mediators from cells activated by antibody-antigen complexes. They are useful in treatment and prophylaxis of allergic conditions such as asthma, hay fever and rhinitis. In an example, 2-nitro-5-(3-phenoxypropoxy)indane-1,3-dione was prepd. by treating 3-(itromethylene-5 (and/or 6)-(3-phenoxypropoxy)phthalide with ET3N.
