Abstract

Prepn. of hydroxocobalamin and its salts comprises redn. then oxidn. of cyanocobalamin to give hydroxocobalamin hydrochloride, then opt. converting it into the free base or another salt, in which the redn. is performed in absence of oxygen at 25-30 degrees C by rapid addn. of HCl to an aq. mixt. of cyanocobalamin and an alkali borohydride, the reaction mixt. being illuminated by a light source. Process is simple and rapid giving high yields of high purity. Typically cyanocobalamin in water was reduced with KBH4 in NaOH under N2, then 6N HCl was quickly added under N2 while illuminated with four 1000-watt lamps. Finally the N2 was replaced by O2 at room temp. to give hydroxocobalamin hydrochloride.
