Abstract

In a process for the industrial-scale prodn. of high-titre interferon from continuous cell strains, the cells are maintained in culture form and suspension form in a single cuvette during the following process steps:- (a) interferon of the type corresp. to the cells is added up to saturation density to the nutrient soln. of a cell strain developed in suspension form, L-glutamine also being added in an amt. sufficient to maintain the cells in an active metabolic state, and the culture is left (with suitable agitation) for a few hrs. at 37 degrees for interferon-stimulation to take place; (b) agitation is stopped and cells are allowed to settle out; (c) the cell precipitate is resuspended in the minimum volume of a highly concd. suspension of an induction virus and the suspension is agitated 1 hr. then centrifuged; (d) the cells are subjected to the action of metabolic inhibitors for several hrs., then the inhibitors are removed by centrifugation; (e) the cells are resuspended in serum-free nutrient soln. and allowed to incubate 15-20 hrs.; and (f) the cells are allowed to settle, and the interferon-contg. supernatant is collected, and concd. and purified by known procedures. Suspension culture procedure enables interferon to be obtd. in yields previously obtainable only by monolayer culturing.
