Abstract
Bacteria in urine samples are detected and counted by a highly sensitive, rapid, accurate and reproducible procedure in which (a) non-bacterial ATP-contg. cells are disintegrated (e.g. by a pref. anionic or nonionic detergent), (b) the non-bacterial ATP is decomposed (e.g. by an ATPase), (c) the ATP-decomposing agent is removed, (d) the bacterial cells are disintegrated to liberate their ATP, (e) the pH is adjusted to a value (pref. 7.4) favourable for a bioluminescence reaction of ATP with a luciferase-luciferine mixture, (f) a luciferase-luciferine mixture contg. Mg++ or Mn++ ions is added, and (g) the quantity of light emitted is determined.
