Abstract
New cpds. of formula (I) having exo configuration with respect to the -C(OR5)R4-C(OR5)R2R3 group:- (where R1 = H, C1-8 alkyl, C3-10 cycloalkyl, C7-12 aralkyl, phenyl opt. subst. by 1-3 Cl atoms or C1-4 alkyl or ethyl beta-subst. by 3 Cl, 2 or 3 Br or 1-3 I atoms; R2 = H or C1-8 alkyl subst. by 0-3 F atoms. R3, R4 = H or C1-4 alkyl, R5 = H or C1-5 alkylsulphonyl R1 being H only when R5 is H , -CnH2n- = C1-8 alkylene subst. by 0-2 F atoms), can be converted into PGE1 and analogues (formula VII), PGF1 and analogues (formula VIII) and PGA1 and analogues (formula Ix). Use:- PGE1, PGF1 alpha, PGF1 beta and PGA (covered by formulae VII, VIII and IX) and their esters and pharmaceutically acceptable salts have biological activity e.g. they lower arterial blood pressure (PGE1, PGF1 beta and PGA1), raise blood pressure (PGF1 alpha,), stimulate smooth muscle, potentiate other smooth-muscle stimulants, have antilipolytic activity, inhibit gastric secretion (PGE1 and PGA1), have CNS activity, reduce blood platelet adhesion, and inhibit blood platelet aggregation and thrombosis formation.
