Abstract

Stable lipase prepd. by culturing a strain of Mucoraceae under aerobic conditions until the enzyme-activity of the medium has become constant; the mycelium is then separated from the liquid and the lipase removed from the filtrate. Lipase in general is an important enzyme in fat-metabolism by hydrolysis of triglycerides. The new lipase may be used in the therapy of gastrointestinal ailments caused by pancreas-insufficiency, insufficiency of secretion caused by age, atheromatoses and psoriasis, which is often accompanied by disturbances in the fat-metabolism. Specially purified prepns. may be used in the treatment of all forms of pancreas insufficiency, possibly accompanied by steatorrhea, dyspepsia, difficulties of fat-metabolism, fat intolerance, dyslepemias and difficulties in fat-absorption after operations of the metabolic system. The very pure form may be injected whereby the lipase is present in the form of a lyophilic-dried powder which is dissolved in situ in physiological salt soln. or suitable buffer. Therapeutic indications: Xanthomatoses, arteriosclerosis, severe pancreas-insufficiency, essential hyperlipidemia and inherent dyslipidemia. The known lipase is based on dried and powdered pancreas which has limited activity, is only active at pH 7.5-9.5, has great instability together with unpleasant odour and taste. The new lipase is very active, stable and free from unpleasant odour.
