Abstract

A procedure for the preparation of new substituted perhaloalkylamidines of formula: where R is a perhalogenated lower alkyl group; R2 is hydrogen, lower alkyl, or lower alkanoyl; is lower alkyl, lower phenylalkyl, lower alkenyl, lower alkynyl, cycloalkyl or cycloalkenyl, the groups of which are substituted with one or more hydroxy, lower hydroxyalkyl, lower alkoxy, lower alkylthio, lower alkanoyloxy, lower alkanoylamino, phenyl, hydroxyphenyl, lower alkoxyphenyl, phenoxy, phenylsulfinyl, naphthylsuffinyl, cycloalkyl, lower alkylphenyl, lower alkenylphenyl, phenylcycloalkyl, or halogen; R3 is also lower phenylalkyl substituted with perhaloalkylamidino-lower alkyl; R3 may also be a 5- to 7-membered heterocyclic group containing one or two amidine nitrogen-linked heteroatoms through a lower alkyl group selected from oxygen and nitrogen or a benzoheterocyclic group optionally substituted by lower alkyl, lower alkoxy, or benzoyl, where the benzo radical is fused to a 5- to 7-membered heterocyclic ring with one or two heteroatoms selected from oxygen and nitrogen; and where each amino or hydroxy group with a labile proton is optionally converted to a glycoside; and its non-toxic and pharmaceutically acceptable salts; whose procedure consists of: 1) reacting a perhalogenated lower alkylnitrile with an amine of formula: H-NR2R3 where R2 and R3 defined above. 2) Optionally, when R is hydrogen, react the compound obtained in the previous step with an alkylating agent or an acylating agent.
