Abstract

A method of preparing a 1-substituted 1,4-benzodiazepine compound of the formula **(See formula)** wherein R1 is nitro or chlorine, R2 and R3 are each phenyl which may be substituted with chlorine and R4 is methylene or ethylidene, and an acid addition salt thereof, which comprises reacting a compound of the formula : **(See formula)** where R1 is nitro, when R5 and R6, taken together, represent nitrogen, or R1 is chlorine, when R5 and R6, taken together, represent nitrogen or R5 is phthalimido and R6 is oxygen, and R2 is as defined above, with an N-substituting agent of the formula: Y-R4-O-CH2-R3 wherein Y is an acidic moiety and R3 and R4 are each as defined above, to obtain an N-substituted compound of the formula: **(See formula)** wherein R1, R2, R3, R4, R5 and R6 are each as defined above, and react the resulting compound where R1 is chlorine and R5 is phthalimido and R6 is oxygen, with hydrazine hydrate, and further converting the 1-substituted 1,4-benzodiazepine compound looking for it into an acid addition salt thereof.
