Abstract

The invention comprises thiocolchicine derivatives of the general formula:- <FORM:0935187/IV(a)/1> wherein R is alkyl containing from 1 to 12 carbon atoms, alkenyl containing from 2 to 10 carbon atoms, cycloalkyl containing from 3 to 8 carbon atoms inclusive, or a mono-or di-cyclic aromatic radical, and X is an oxygen or sulphur atom, and processes for their preparation (a) by reacting a primary amine R1NH2 with an isothiocyanate or isocyanate RNCX where R1 represents: <FORM:0935187/IV(a)/2> (b) by reacting RNH2 with R1NCX, the latter compound preferably being prepared from R1NH2 by a known non-oxidative method for converting a primary amine into an isothiocyanate or isocyanate, especially reaction with phosgene, or with thiophosgene, or with carbon disulphide in the presence of ammonia followed by treatment of the product with phosgene, ethyl chloroformate or a heavy metal salt. Twelve detailed examples are given. The invention also comprises anti-mitotic compositions containing one or more of the novel compounds and an inert carrier.
