Abstract
Método de producción de un fragmento Fab de inmunoglobulina que comprende las siguientes etapas: - proporcionar una célula única productora de inmunoglobulina, - obtener a partir de dicha célula el ácido nucleico que codifica los dominios variables de la cadena ligera y pesada de inmunoglobulina, que también codifica una parte del dominio constante de la cadena ligera y una parte del dominio CH1 de la cadena pesada, con un método que comprende las siguientes etapas: - realizar una primera reacción en cadena de la polimerasa con cuatro a seis cebadores 5' y un cebador 3', - realizar con el producto de la primera reacción en cadena de la polimerasa una segunda reacción en cadena de la polimerasa con trece a quince cebadores 5' y un cebador 3', por el cual en la segunda reacción en cadena de la polimerasa el cebador 3' es el mismo que en la primera reacción en cadena de la polimerasa y al menos un cebador 5' se cambia, y por el cual en la segunda reacción en cadena de la polimerasa el número de nucleótidos entre el extremo 5' de cada uno de los cebadores 5' y el extremo 3' del cebador 3' se reduce en comparación con el número de nucleótidos entre el extremo 5' de cada uno de los cebadores 5' y el extremo 3' del cebador 3' en la primera reacción en cadena de la polimerasa, cuando se une al ácido nucleico que va a amplificarse, - generar una matriz de expresión lineal que comprende el ácido nucleico obtenido, - traducir in vitro dicho ácido nucleico y así producir el fragmento Fab de inmunoglobulina.
