Abstract
Medicamento para la prevención de angiogénesis tumoral que comprende una proteína de fusión del receptor de la linfotoxina Beta (LTBR) y de inmunoglobulina.
Description
Composición para la prevención de crecimiento
tumoral.
La presente invención se refiere a un
medicamento para la prevención de crecimiento tumoral, suprimiendo
la activación, que se transmite a través del receptor de linfotoxina
\beta (LTBR).
La proteína de membrana linfotoxina \beta
(Browing y col., Cell, Vol. 72, 847-856, 1993) se
liga en forma homómera y heterómera en un receptor específico de
linfotoxina \beta (LTBR) (Crowe y col., Science Vol. 264, 29 de
Abril de 1994, 707-709).
Browing y col. (J. Exp. Med. Vol. 183, Marzo de
1996, 867-878) describen que la transmisión de
señales a través del sistema LTBR provoca la muerte celular para
algunas células tumorales de adenocarcinoma. Por lo tanto,
proponen la activación de LTBR como posible terapia
anticancerosa.
Degli-Esposti y col. (Journal of
Immunology, 1997, 158: 1756-1762) informan que se
puede inhibir el crecimiento de células de melanoma, cuando ser
activa el sistema LTBR. Por lo tanto, proponen el tratamiento de
tumores sólidos con linfotoxina \beta.
En cambio, nuestros hallazgos muestran que se
puede reducir la velocidad o impedir el crecimiento tumoral,
suprimiendo la activación de LTBR.
Por lo tanto, el objeto de la invención es un
medicamento para impedir el crecimiento tumoral, suprimiendo la
actividad transmitida por LTBR o al menos reduciendo su
actividad.
El medicamento es especialmente adecuado en
tumores sólidos.
La supresión del sistema LTBR se puede realizar
en diferentes planos.
Por ejemplo, se puede suprimir la expresión del
gen LTBR, utilizando moléculas antisentido. Otra posibilidad para
suprimir específicamente la expresión del gen consiste en eliminar
el ARNm del LTBR, por ejemplo a través de hidrólisis específica por
medio de ribozomas adecuados.
Otra posibilidad para suprimir la expresión del
gen LTBR consiste en el bloqueo de las secuencias de señales de
trascripción correspondientes, por ejemplo de la zona del promotor
del gen LTBR.
Otra posibilidad para suprimir la activación del
sistema LTBR consiste en la utilización de antagonistas de
linfotoxina \beta, es decir, de moléculas que se pueden ligar en
el LTBR, pero no pueden provocar ninguna activación del sistema
receptor. Se pueden hallar tales sustancias verificando en las
sustancias a ensayar en un sistema de ensayo celular su capacidad
para reducir la activación del sistema LTBR en presencia de
linfotoxina \beta añadida.
Para la presente invención se utiliza una
proteína de fusión de LTBR e inmunoglobulina.
Otro objeto de la invención es la utilización de
sustancias, que suprimir la actividad transmitida por LTBR para la
producción de medicamentos citostáticos.
Se ha observado que la supresión de la
activación de LTBR provoca una inhibición de la angiogénesis del
tumor y, por lo tanto, tiene una influencia inhibidora sobre el
crecimiento de tumores sólidos.
La prevención de la activación de LTBR con los
medios y procedimientos indicados anteriormente es adecuada, por lo
tanto, especialmente para la inhibición de angiogénesis de
tumores.
Los siguientes experimentos siguientes explican
la invención.
Experimento
1
Se transfirieron células tumorales
(BFS-1) con una proteína de fusión a partir de LTBR
e inmunoglobulina (BFS-LTD). Esta proteína de
fusión a partir de LTBR e inmonoglobulina actúa como inhibidor de
LTBR. Estas células tumorales transferidas mostraron, en
comparación con células tumorales de tipo no cultivado un
crecimiento tumoral inhibido, cuando se introdujeron en ratón
syn-gen (figura).

Experimento
2
Si se introducen en un segundo experimento estas
células en ratones deficientes de ligandos (es decir, ratones
noqueados dobles, que no producen ni linfotoxina alfa ni linfotoxina
beta), no resultan diferencias significativas en el crecimiento
tumoral de la célula tumoral transferida frente a la célula tumoral
no cultivada (figura).
Experimento
3
Se aplicaron a ratones (C57BL/6) células
tumorales normales o bien células tumorales, a las que había sido
transferido el inhibidor del receptor LT\beta
(BSF-1-LT\betaR-Ig:Fc)
o un plásmido de control
(p55TNF-receptor-Ig-Fc),
subcutáneamente en una cámara cutánea y se siguió el crecimiento de
la célula tumoral o bien la angioneogénesis a través de microscopio
intravital. La formación de edema el día 2 después de la aplicación
de célula tumoral en los animales, que recibieron células tumorales
transferidas con inhibidor de receptor LT\beta, era claramente
menor que en los controles. Mientras que las células tumorales
normales y las células tumorales transferidas a una estructura de
control al cabo de 5 a 9 días indujeron angiogénesis clara y
crecieron, por lo tanto, entonces como tumores, sin embargo en los
tumores, que son transferidos con el inhibidor del receptor
LT\beta, no se mostró angioneogénesis de ningún tipo en la zona
tumoral ni en el entorno de la piel. Entonces estos tumores tampoco
continuaron creciendo.
En las figuras se pueden ver tomas de los
tumores de control (arriba) y de los tumores transferidos con
inhibidor del receptor LT\beta (abajo) el día 2 (figura 2) y el
día 9 (figura 3) después de la aplicación de célula tumoral.
