Abstract

Procedimiento para identificar un gen cuya activación o inactivación evita una dependencia de una célula por un oncogén inducible para tumorigenicidad, comprendiendo el procedimiento: (a) proporcionar células tumorales ex vivo, siendo dichas células tumorales células tumorales de ratón, comprendiendo dichas células dicho oncogén unido operativamente a un promotor inducible seleccionado del grupo que consiste en un sistema promotor basado en el promotor de tetraciclina, un sistema promotor basado en el promotor de metalotioneína y un sistema promotor basado en el receptor de estrógenos, confiriendo dicho oncogén tumorigenicidad cuando se activa la expresión; (b) provocar la inactivación de dicho oncogén, haciendo que dichas células estén a una mutación de distancia de la tumorigenicidad; (c) introducir en las células un vector retroviral que se integre en los genomas de dichas células; (d) introducir dichas células en un sitio de inyección en un ratón inmunocomprometido, y monitorizar la formación de un tumor en el sitio de inyección en el ratón; y (e) identificar como dicho gen cuya activación o inactivación evita la dependencia de una célula por dicho oncogén para tumorigenicidad, un gen que presenta una expresión aumentada o disminuida en dicho tumor como resultado de la introducción del vector retroviral.
