Abstract
PROCEDIMIENTO DE OBTENCION Y SOBREEXPRESION CONSTITUTIVA DE LA CITRATO PERMEASA P DE LACTOCOCCUS LACTIS BIOVAR DIACETILACTIS METODO PARA SUBCLONAR EL GEN CITP DEL PLASMIDO LACTOCOCCAL PCIT264 EN EL PLASMIDO VECTOR DE AMPLIO ESPECTRO DE HUESPED PLS1. PARA CONSEGUIR UNA SOBREEXPRESION CONSTITUTIVA DE LA CITRATO PERMEASA SE COLOCO EL GEN CITP BAJO EL PROMOTOR DE TRANSCRIPCION DEL GEN POLA DE STREPTOCOCCUS PNEUMONIAE. MEDIANTE ESTE PROCEDIMIENTO SE HAN OBTENIDO LOS PLASMIDOS PFU1 CAPAZ DE REPLICAR EN ESCHERICHIA COLI, Y EL PLASMIDO PFL3 CAPAZ DE REPLICAR EN S. PNEUMONIAE Y L. LACTIS. LAS ESTIRPES BACTERIANAS PORTADORAS DE DICHOS PLASMIDOS ADQUIEREN LA CAPACIDAD DE TRANSPORTAR CITRATO. L. LACTIS CONTENIENDO EL PLASMIDO PFL3 SINTETIZA EL RNA MENSAJERO DE CITP A PARTIR DEL PROMOTOR POLA DE UNA FORMA CONSTITUTIVA DURANTE LAS FASES EXPONENCIAL Y ESTACIONARIA DE CRECIMIENTO BACTERIANO.PROCEDURE FOR OBTAINING AND CONSTITUTIVE OVEREXPRESSION OF CITRATE PERMEASE P FROM LACTOCOCCUS LACTIS BIOVAR DIACETILACTIS METHOD FOR SUBCLONING THE CITP GENE OF THE LACTOCOCCAL PLASMID PCIT264 IN THE VAST PLASMID OF HUESPED SPECTRUM OF HUESP. TO OBTAIN A CONSTITUTIVE OVEREXPRESSION OF CITRATE PERMEASE, THE CITP GENE WAS PLACED UNDER THE TRANSCRIPTION PROMOTER OF THE POLA GENE OF STREPTOCOCCUS PNEUMONIAE. THROUGH THIS PROCEDURE, PLASMIDS PFU1 CAPABLE OF REPLICATION IN ESCHERICHIA COLI, AND PLASMID PFL3 CAPABLE OF REPLICATION IN S. PNEUMONIAE AND L. LACTIS. BACTERIAL STRAINS CARRYING SUCH PLASMIDS ACQUIRE THE ABILITY TO TRANSPORT CITRATE. L. LACTIS CONTAINING PLASMID PFL3 SYNTHESIZES CITP MESSENGER RNA FROM THE POLA PROMOTER IN A CONSTITUTIVE FORM DURING THE EXPONENTIAL AND STATIONARY PHASES OF BACTERIAL GROWTH.
