Abstract

Procedure for the production and constitutive superexpression of the citrate permease P of Lactococcus lactis biovar diacetilactis. Method for subcloning the citP gene of the lactococcal plasmid pCIT264 in the broad-spectrum vector plasmid of host pLS1. In order to achieve constitutive superexpression of citrate permease, the citP gene was placed below the transcription promoter of the polA gene of Streptococcus pneumoniae. By means of this procedure the plasmids pFU1, able to replicate in Escherichia coli, and the plasmid pFL3, able to replicate in S. pneumoniae and L. lactis, were obtained. The bacterial strains carrying said plasmids acquire the ability to transport citrate. L. lactis containing the plasmid pFL3 synthesizes the messenger RNA of citP from the promoter polA in a constitutive form during the exponential and stationary phases of bacterial growth.
