Abstract
A composition for improving hair growth, anti-hair loss and/or hair health, comprising a yeast extract, a nutrition mixture, and optionally a xanthine compound is provided. A method of improving hair growth, anti-hair loss and/or hair health, comprising: applying such a composition to the hair is further provided.
Description
- The present invention relates to a composition for improving hair growth, anti-hair loss and/or hair health. In particular, the present invention relates to a composition for improving hair growth, anti-hair loss and/or hair health, comprising a yeast extract, a nutrition mixture, and optionally a xanthine compound.
- Alopecia or hair loss is suffered by many people around the world. In recent years, not only men suffer from hair loss, but also more and more women suffer from it. In China, almost half of the consumers are concerned about hair loss and hair health. Currently, minoxidil and finasteride are the most effective solutions for treating hair loss. However, due to regulations in some countries, such as China, and unwanted side-effects such as irritation, minoxidil and finasteride cannot be used in personal care products.
- As is known to all, hair loss is caused by various factors, including internal factors such as genetic constitution, hormones, aging and deficiency of vitamin, or extrinsic factors such as stress in daily life; and to increase hair growth and anti-hair loss, the key is to regulate hair follicle cycle by promoting anagen phase and inhibiting catagen phase, and keratin is also a key for improving hair growth, anti-hair loss and hair health.
- In view of the above, it would be desirable to provide a composition for improving hair growth, anti-hair loss and/or hair health, and a method of improving hair growth, anti-hair loss and/or hair health, which exhibit improved effects in promoting anagen gene expression and inhibiting catagen gene expression, and/or in promoting keratin gene expression.
- In a first aspect, the present invention provides a composition for improving hair growth, anti-hair loss and/or hair health, comprising a yeast extract, a nutrition mixture, and optionally a xanthine compound.
- In a second aspect, the present invention provides a method of improving hair growth, anti-hair loss and/or hair health, comprising: applying a composition according to the present invention to the hair. All of the composition and the method according to the present invention are based on the following discoveries of the inventors: a composition comprising a yeast extract and a nutrition mixture according to the present invention has improved efficacies in both promoting anagen gene expression and inhibiting catagen gene expression; and a composition comprising a yeast extract, a nutrition mixture and a xanthine compound further has synergetic efficacies in promoting keratin gene expression.
- It is to be understood by one of ordinary skill in the art that the present discussion is a description of exemplary embodiments only, and is not intended as limiting the broader aspects of the present invention. Each aspect so described may be combined with any other aspect or aspects unless clearly indicated to the contrary. In particular, any feature indicated as being preferred or advantageous may be combined with any other feature or features indicated as being preferred or advantageous.
- Unless specified otherwise, as used herein, the terms “a” , “an” and “the” include both singular and plural referents.
- The terms “comprising” and “comprises” as used herein are synonymous with “including” , “includes” or “containing” , “contains” , and are inclusive or open-ended and do not exclude additional, non-recited members, elements or process steps.
- Unless specified otherwise, the recitation of numerical end points includes all numbers and fractions subsumed within the respective ranges, as well as the recited end points.
- Unless otherwise defined, all terms used in the disclosing the invention, including technical and scientific terms, have the meaning as commonly understood by one of the ordinary skill in the art to which this invention belongs.
- According to the present invention, surprisingly, the inventors of the present invention found that a composition comprising a yeast extract and a nutrition mixture according to the present invention has improved efficacies in both promoting anagen gene expression and inhibiting catagen gene expression; and a composition comprising a yeast extract, a nutrition mixture and a xanthine compound further has synergetic efficacies in promoting keratin gene expression.
- As to anagen phase, HGF (Hematopoietic Growth Factor) is a biomarker of anagen phase. In particular, HGF is an important factor released by dermal papilla to steer different follicular processes; it is maximum expressed during anagen phase, stimulates hair follicle growth, and plays an important role in hair cycle control; and additionally, HGF can retard the anagen-catagen transformation, thus prolonging the active growth phase and counteracting premature hair loss. KGF (Keratinocyte Growth Factor) is also expressed during the anagen phase and plays a role in cell division rate and differentiation of keratinocytes. To improve hair growth and anti-hair loss, the anagen phase and thus HGF gene expression and KGF gene expression should be improved. The inventors of the present invention found that merely using either of a yeast extract and a nutrition mixture has almost no impact on HGF gene expression, but the combination of a yeast extract and a nutrition mixture greatly improves HGF over-expression; and the combination of a yeast extract and a nutrition mixture can significantly improve KGF over-expression.
- As to catagen gene, TGFβ1 (Transforming Growth Factor beta 1) is a biomarker of catagen phase. In particular, TGFβ1 is an important marker of inducing catagen by most likely inhibition of keratinocyte proliferation and induction of apoptosis. To increase hair growth and anti-hair loss, the catagen phase and thus TGFβ1 gene over-expression should be inhibited. The inventors of the present invention found that merely using either of a yeast extract and a nutrition mixture increases TGFβ1 gene over-expression significantly, which is not desirable; however, the combination of a yeast extract and a nutrition mixture does not significantly increase TGFβ1 gene expression.
- As to keratin, KRT33a, KRT34 and KRT86 are biomarkers for keratin production. In particular, hair structure depends from the composition of hair-specific-structure related proteins, hair keratins, and with an impact of the composition of hair-specific proteins, a biological influence on hair structure is produced. To obtain a better hair structure and improve hair health, keratin gene expression, and thus KRT33a, KRT34 and KRT86 gene expressions should be promoted. The inventors of the present invention found that the combination of a yeast extract, a nutrition mixture and a xanthine compound induces synergic increases in the expressions of KRT33a, KRT34 and KRT86, as compared with the sum of merely using any one of a yeast extract, a nutrition mixture and a xanthine compound.
- In a first aspect, the present disclosure is generally directed to a composition for improving hair growth, anti-hair loss and/or hair health, comprising a yeast extract, a nutrition mixture, and optionally a xanthine compound.

- As used herein, the term “yeast extract” refers to any yeast extract which is conventionally used in a hair care product, and preferably a substance having CAS No. 8013-01-2.
- Preferably, in the present invention, the yeast extract is used in the form of a mixture comprising a yeast extract, a maltodextrin and water, preferably in the form of a mixture comprising from 25.0%to 75.0%by weight of a yeast extract, from 25.0%to 75.0%by weight of maltodextrin, and 0.1%to 10.0%by weight of water, and more preferably in the form of a mixture comprising from 35.0%to 65.0%by weight of a yeast extract, from 35.0%to 65.0%by weight of maltodextrin, and 2%to 8%by weight of water, each based on the total weight of the mixture.
- Commercially available examples of the yeast extract which can be used in the present invention include but not limited to a product under the trade name of Hairgenyl from Silab Company and a product under the trade name of HYDROGLYCOLIC EXTRACT (HG) from AQiA Química Inovativa.
- In a preferred embodiment of the present invention, the content of the yeast extract is from 0.01%by weight to 10.0%by weight, preferably from 0.02%by weight to 5.0%by weight, more preferably from 0.025%by weight to 1.0%by weight, and most preferably from 0.035%by weight to 0.070%by weight, based on the total weight of the composition. If the amount of the yeast extract is higher than 10.0%by weight, based on the total weight of the composition, the composition obtained will have a problem of incompatibility. If the amount of the yeast extract is lower than 0.01%by weight, based on the total weight of the composition, it will not achieve the advantageous technical effects of the present invention.
- Nutrition mixture
- According to the present invention, the composition comprises a nutrition mixture.
- As used herein, the term “nutrition mixture” refers to any nutrition mixture which is conventionally used in a hair care product, and preferably is selected from the group consisting of products under the trade name of Trichogen available from BASF.
- Preferably, the nutrition mixture which can be used in the present invention is preferably under the trade name of Trichogen VEG BC10164, Trichogen VEG LS 9812 and/or Trichogen VEG LS 9922 from BASF, and more preferably is under the trade name of Trichogen VEG BC10164 from BASF.
- In a preferred embodiment of the present invention, the content of the nutrition mixture is from 0.01%by weight to 10.0%by weight, preferably from 0.02%by weight to 5.0%by weight, more preferably from 0.03%by weight to 1.0%by weight, and most preferably from 0.04%by weight to 0.5%by weight, based on the total weight of the composition. If the amount of the nutrition mixture is higher than 10.0%by weight, based on the total weight of the composition, the composition obtained will have a problem of incompatibility. If the amount of the nutrition mixture is lower than 0.01%by weight, based on the total weight of the composition, it will not achieve the advantageous technical effects of the present invention.
- Xanthine compound
- According to the present invention, the composition may comprise a xanthine compound. When a xanthine compound is used in the composition according to the present invention, the combination of a yeast extract, a nutrition mixture and a xanthine compound significantly improves hair growth, anti-hair loss and hair health, especially in terms of keratin.
- As used herein, the term “xanthine compound” refers to any xanthine compound which is conventionally used in a hair care product, and preferably is selected from the group consisting of caffeine, xanthine, 1-methyl xanthine, theophylline, theobromine, and derivatives thereof.
- Preferably, the xanthine compound which can be used in the present invention is caffeine. In a preferred embodiment of the present invention, the caffeine is water free.
- Commercially available examples of the xanthine compound which can be used in the present invention include but not limited to caffeine available from Siegfried CMS AG Switzerland, BASF, Merck KGaA, Biocosmethic, etc.
- In a preferred embodiment of the present invention, the content of the xanthine compound is from 0.01%by weight to 10.0%by weight, preferably from 0.02%by weight to 5.0%by weight, more preferably from 0.03%by weight to 1.0%by weight, and most preferably from 0.04%by weight to 0.1%by weight, based on the total weight of the composition. If the amount of the xanthine compound is higher than 10.0%by weight, based on the total weight of the composition, the composition obtained will have a problem of incompatibility. If the amount of the xanthine compound is lower than 0.01%by weight, based on the total weight of the composition, it will not achieve the advantageous technical effects of the present invention.
- In a more preferred embodiment, the present invention provides a composition for improving hair growth and anti-hair loss, comprising: from 0.025%by weight to 1.0%by weight, preferably 0.0485%by weight of a yeast extract, preferably one under the trade name of Hairgenyl; and from 0.03%by weight to 1.0%by weight, preferably 0.1%by weight of a nutrition mixture, preferably one under the trade name of Trichogen VEG BC10164, each based on the total weight of the composition.
- In another more preferred embodiment, the present invention provides a composition for improving hair growth, anti-hair loss and hair health, comprising: from 0.025%by weight to 1.0%by weight, preferably 0.0485%by weight of a yeast extract, preferably one under the trade name of Hairgenyl; from 0.03%by weight to 1.0%by weight, preferably 0.1%by weight of a nutrition mixture, preferably one under the trade name of Trichogen VEG BC10164; and from 0.01%by weight to 10.0%by weight, preferably 0.05%by weight of a xanthine compound, preferably caffeine , each based on the total weight of the composition.
- Additive
- The composition according to the present invention may further comprise an additive which is conventionally used in a hair care product, including but not limited to a surfactant, a hair humectant, a thickening agent, a preservative, a perfume, a cationic hair conditioner, a fatting agent, a skin conditioner, a skin humectant, and ethanol.
- The presence, type and amount of the additive may be determined by a specialist in the art according to actual needs, as long as they do not negatively affect the purpose of the present invention. For example, when the composition is a shampoo, it may further comprise a surfactant, a hair humectant, a thickening agent, a preservative and a perfume; when the composition is a hair conditioner, it may further comprise a surfactant, a cationic hair conditioner, a fatting agent, a preservative and a perfume; and when the composition is a scalp care product, such as a scalp serum or a scalp spray, it may further comprise a skin conditioner, a skin humectant, ethanol, a preservative and a perfume.
- The composition according to the present invention may be a hair care, styling and/or color product, and/or a scalp care product, such as shampoo, a hair conditioner, a hair styling product, a hair color product, a scalp serum, and/or a scalp spray. Preferably, the composition according to the present invention is a shampoo or a scalp serum.
- The composition according to the present invention may be prepared according to conventional methods in the art, such as by mixing all the components in the composition at room temperature.
- In a second aspect, the present disclosure is generally directed to a method of improving hair growth, anti-hair loss and/or hair health, comprising: applying a composition according to the present invention to the hair.
- With regard to the further preferred embodiments of the method of improving hair growth, anti-hair loss and/or hair health, and the hair care, styling and/or color product, what has been said about the composition according to the present invention applies mutatis mutandis.
- The invention will be illustrated in more details by way of the following examples which are not to be understood as limiting the concept of the invention.
- Examples
- 1. Gene expression analysis with Real-Time PCR
- In order to evaluate improvements in hair growth, anti-hair loss and hair health, a hanging drop model was used as an in-vitro model to test the gene expression results of HGF, KGF, TGFβ1, KRT33a, KRT34, and KRT86.
- In particular, hanging drops consisted of different human cells which are important for cosmetic claims. They were cultured in AkuraTM PLUS Hanging Drop System (InSphero) . Different cell types were seeded in appropriate count in the plates. All cells were primary cells obtained from plastic surgery. In the plates, the drops were matured for an evaluated period of time. Then they were harvested and treated for an evaluated time. All samples were performed with 7 or 8 replicates and taken for the following gene expression analyses.
- The molecular regulation of keratins and growth factors were evaluated by analyzing the gene expression profiles of the used organotypic model using PCR techniques. RNA was isolated from organotypic models using the RNeasy MiniKit (Qiagen) according to the manufacturer’s instructions with slight optimization steps. Spectrometric quantification of the isolated RNA was performed using Spectramax (Molecular Devices) . The cDNA Synthesis was performed according to the protocol for M-MLV reverse Transcriptase from Promega Corporation, USA. with slight optimization steps.
- Following standard reverse transcription of each RNA sample, comparative quantitative PCR analysis was performed using a MX3000P Real Time PCR System (Stratagene, La Jolla, California) with gene specific primer and taqman probes. After polymerase activation for 10 min at 95℃, iterative cycles were set up as follows: 30 s at 95℃, 1 min at 60℃. The analysis was performed in a multiplex panel with different gene sets:
- 1. set: Housekeeper, HGF, and KGF,
- 2. set: Housekeeper, TGFβ1,
- 3. set: Housekeeper, KRT86, KRT33a, and KRT34.
- In the PCR reaction, the formation of the PCR product can be made visible via a fluorescene signal. The fluorescene signal is proportional to the formed product. The stronger the expression of a gene is, the more product has been formed, and the higher the fluorescene signal is.
- Comparative Quantitation A QPCR analysis method enables determination of relative gene expression compared to a calibrator (asingle standard) .
- This method is used to establish relative fold-increase in expression by assuming unchanging reaction efficiency. This method eliminates the requirement to include a standard curve with each reaction. Comparative quantitation can be applied to DNA and cDNA targets, and the most common application is the comparison of mRNA expression levels in treated versus untreated or normal versus diseased cells or tissue.
- All samples tested were obtained by dissolving the components listed in the first column of the following tables 1-6 in culture media, and the untreated samples were treated with the same culture media but without any actives. The culture media was a mixture of three standard media provided by Gibco and Fujifilm/Irvine scientific with addition of different growth supplements and 10%sera. In the following tables 1-6, “Trichogen” refers to a nutrition mixture obtained under the trade name of Trichogen VEG BC10164 from BASF; “Hairgenyl” refers to a yeast extract product obtained under the trade name of Hairgenyl from Silab Company, containing 48.50%by weight of yeast extract (CAS No. 8013-01-2) , 46.50%by weight of maltodextrin (CAS No. 9050-36-6) and 5.00%by weight of water; and “Caffeine” refers to a pure caffeine (100%, and water-free) obtained from Siegfried CMS AG Switzerland. The weight percentage following the component refers to the amount of the corresponding component based on the total weight of the sample. “Hairgenyl 0.1 wt.%” refers to the content of the yeast extract product under the trade name of Hairgenyl, rather than the content of the yeast extract; and the content of the yeast extract in the sample was 0.04850%by weight based on the weight of the sample.
- Statistical difference was determined by use of two-tailed student T-test, and a p-value less than 0.05 was considered as significant difference.
- Gene expression results of HGF, KGF, TGFβ1, KRT33a, KRT34, and KRT86 thus obtained were as follows:

- From the above table 1, it can be seen that merely using either of a yeast extract and a nutrition mixture has almost no significant impact on HGF gene expression (p values of 0.1803 and 0.8162 respectively) , but the combination of a yeast extract and a nutrition mixture greatly and significantly improves HGF over-expression (121%, and p value of 0.0006) . Thus, the combination of a yeast extract and a nutrition mixture according to the present invention can synergistically increase hair growth and anti-hair loss.

- From the above table 2, it can be seen that the combination of a yeast extract and a nutrition mixture can significantly improve KGF over-expression. Thus, the combination of a yeast extract and a nutrition mixture according to the present invention can significantly increase hair growth and anti-hair loss.
- Table 3: Gene Expression results of TGFβ1
- From the above table 3, it can be seen that merely using either of a yeast extract and a nutrition mixture significantly increases TGFβ1 gene over-expression (49%and 45%respectively, and p values of 0.0175 and 0.0071 respectively) , but the combination of a yeast extract and a nutrition mixture does not significantly increase TGFβ1 gene expression (20%and p value of 0.1014) . Thus, the combination of a yeast extract and a nutrition mixture according to the present invention has synergistical effect in hair growth and anti-hair loss.

- From the above Table 4, it can be seen that the increase in the over-expression of KRT33a obtained from the combination of a yeast extract, a nutrition mixture and a xanthine compound (822%) is much higher than those obtained from any one of a yeast extract, a nutrition mixture and a xanthine compound (119%, 84%and 184%respectively) , and even is much higher than the sum of the increases obtained from any one of a yeast extract, a nutrition mixture and a xanthine compound. Thus, the combination of a yeast extract, a nutrition mixture and a xanthine compound according to the present invention can synergistically increase hair growth, anti-hair loss and hair health.

- From the above Table 5, it can be seen that the increase in the over-expression of KRT34 obtained from the combination of a yeast extract, a nutrition mixture and a xanthine compound (624%) is much higher than those obtained from any one of a yeast extract, a nutrition mixture and a xanthine compound (106%, 75%and 169%respectively) , and even is much higher than the sum of the increases obtained from any one of a yeast extract, a nutrition mixture and a xanthine compound. Thus, the combination of a yeast extract, a nutrition mixture and a xanthine compound according to the present invention can synergistically increase hair growth, anti-hair loss and hair health.

- From the above Table 6, it can be seen that the increase in the over-expression of KRT34 obtained from the combination of a yeast extract, a nutrition mixture and a xanthine compound (172%) is much higher than those obtained from any one of a yeast extract, a nutrition mixture and a xanthine compound (43%, 54%and 71%respectively) , and even is a little higher than the sum of the increases obtained from any one of a yeast extract, a nutrition mixture and a xanthine compound. Thus, the combination of a yeast extract, a nutrition mixture and a xanthine compound according to the present invention can synergistically increase hair growth, anti-hair loss and hair health.
- 2. Examples of Compositions for improving hair growth, anti-hair loss and/or hair health

- Shampoos 1 &2, and Scalp Serums 1 &2 as shown above in Tables 7-10, all of which are compositions for improving hair growth, anti-hair loss and/or hair health according to the present invention, were prepared as follows: at room temperature, dispersing surfactants or carbomer in water by stirring; adding other substances including humectants, preservatives and actives one by one when stirring; and then adding sodium hydroxide and fragrance into the mixture.
- Shampoo 1 and Scalp Serum 1 according to the present invention exhibited improved hair growth and anti-hair loss. Shampoo 2 and Scalp Serum 2 according to the present invention exhibited improved hair growth, anti-hair loss and hair health.
- Although some preferred embodiments have been described, many modifications and variations may be made thereto in light of the above teachings. It is therefore to be understood that the invention may be practiced otherwise than as specifically described without departing from the scope of the appended claims.
Claims (10)
- A composition for improving hair growth, anti-hair loss and/or hair health, comprising a yeast extract, a nutrition mixture, and optionally a xanthine compound.
- The composition according to claim 1, wherein the yeast extract is a substance having CAS No.8013-01-2, which is preferably used in the form of a mixture comprising a yeast extract, a maltodextrin and water, more preferably under the trade name of Hairgenyl from Silab Company, and is preferably used under the trade name of HYDROGLYCOLIC EXTRACT from AQiA Química Inovativa; and most preferably, the yeast extract is used under the trade name of Hairgenyl from from Silab Company.
- The composition according to claim 1 or 2, wherein the nutrition mixture is selected from the group consisting of products under the trade name of Trichogen, preferably Trichogen VEG BC10164, Trichogen VEG LS 9812 and/or Trichogen VEG LS 9922, and more preferably Trichogen VEG BC10164.
- The composition according to any one of claims 1 to 3, wherein the xanthine compound is selected from the group consisting of caffeine, xanthine, 1-methyl xanthine, theophylline, theobromine, and derivatives thereof, and preferably is caffeine.
- The composition according to any one of claims 1 to 4, wherein the content of the yeast extract is from 0.01%by weight to 10.0%by weight, preferably from 0.02%by weight to 5.0%by weight, and more preferably from 0.025%by weight to 1.0%by weight, based on the total weight of the composition.
- The composition according to any one of claims 1 to 5, wherein the content of the nutrition mixture is from 0.01%by weight to 10.0%by weight, preferably from 0.02%by weight to 5.0%by weight, and more preferably from 0.03%by weight to 1.0%by weight, based on the total weight of the composition.
- The composition according to any one of claims 1 to 6, wherein the content of the xanthine compound is from 0.01%by weight to 10.0%by weight, preferably from 0.02%by weight to 5.0%by weight, and more preferably from 0.03%by weight to 1.0%by weight, based on the total weight of the composition.
- The composition according to any one of claims 1 to 7, further comprising an additive selected from the group consisting of a surfactant, a hair humectant, a thickening agent, a preservative, a perfume, a cationic hair conditioner, a fatting agent, a skin conditioner, skin humectant, and ethanol.
- The composition according to any one of claims 1 to 8, which is a hair care, styling and/or color product, and/or a scalp care product, such as shampoo, a hair conditioner, a hair styling product, a hair color product, a scalp serum, and/or a scalp spray.
- A method of improving hair growth, anti-hair loss and/or hair health, comprising: applying a composition according to any one of claims 1 to 9 to the hair.
