- SC samples were then immersed in the test compositions diluted 4-fold (25%) with the Evian water for 2h with gentle stirring on shaker and rinsed by three successive washes of Evian water. The samples were left at room temperature for at least 4h to ensure drying. The SC samples were then placed in the temperature and humidity control chamber for equilibrium at 30°C and 30% RH. After overnight equilibrium, TEWL measurements “Tl” were recorded.
- the harshness of surfactants in the compositions were evaluated by employing the zein solubility test, wherein, zein, a yellow corn protein, similar to keratin present in the skin, with limited solubility in water is denatured (solubilized) by the surfactant.

Description
A CLEANSING COMPOSITION FOR KERATIN SURFACE AND METHODS THEREOF
FIELD OF INVENTION
[0001] The present disclosure relates to the field of cosmetic compositions, in particular, relates to transparent serum-like cleansing compositions. The present disclosure also relates to a process of preparing the composition and a method for treating keratin surface.
BACKGROUND OF INVENTION
[0002] An optimized cleansing regimen is the first key step to achieve an acne free skin. An acne prone skin is highly sensitive to soaps, harsh surfactants, and high pH, which are common in basic cleansing products in the market. Therefore, usage of such products worsens the existing acne and is an inappropriate strategy for cleansing acne prone skin.
[0003] Several cleansers have encroached the consumer market claiming to be mild, and with an ability to moisturize the skin. In addition, these cleansers comprise certain anti-acne agents. However, the cocktail of mild cleansing agents and anti-acne agents leads to several shortcomings with respect to product stability and ultimately falls short on consumer’s expectations. Moreover, such cleansers suffer stability issues due to increased temperatures thereby affecting storage stability and shelf-life. It is also necessary for such cleansers to remain homogenous throughout their shelf life.
[0004] Therefore, there is a need to develop a cleansing composition with optimized combination of mild cleansing agents and act as effective anti-acne agents, with desired product texture and stability.

Example 1
Cleansing compositions
[0069] Cleansing compositions C(l) to C(6) in accordance with the present disclosure as provided in Table 1 were prepared as described below.
[0070] The process involved blending ingredients of phase A with water at a temperature of about 75 to 80°C to obtain the first mixture. The ingredients of phase B were added at about 75 to 80°C to the main vessel along with the first mixture and was homogenized to obtain the second mixture. The ingredients of phase C were then added to the second mixture at about 75 to 80°C and allowed for complete mixing and cooling below 65°C to obtain a third mixture. Phase D ingredients are premixed and was then added to the third mixture, followed by addition of phase E ingredients, at about 45 to 50°C and were allowed to blend under continuous stirring, resulting in the clear homogenous composition.
[0071] Similarly, the comparative compositions CP(1) to CP(10) were prepared with the set of ingredients in their corresponding weight % as indicated in Table 2 below.
Table 1
Table 2
Example 2
Evaluation of the compositions
Viscosity measurements [0072] The compositions as prepared in Example 1 were subjected to viscosity measurements using LR Lamy Rheology instrument, Rheometer RM200 Touch with M3 spindle. Each of the test composition was filled in the cup attached to the
rim of the instrument and the viscosity was measured. The final reading at 600 seconds was recorded.
Determination of foam volume
[0073] 1% sample solution comprising the test composition with hard water was prepared. The sample solution was mixed under constant speed for about a time period of 60 seconds and was then transferred to a measuring cylinder. The foam was levelled and the foam volume for each of the sample solution was noted. pH measurement
[0074] pH of the test compositions were measured using pH instrument of Mettlor Toledo, calibrated with buffer solution. The pH of the compositions were measured at room temperature, at about 25°C.
Measurement of trans epidermal water loss (TEWL)
[0075] The surface of the Stratum Corneum (SC) samples were cleaned with isooctane and placed in sample holders of TEWL. The sample holders were then immersed in Evian water to wet the SC and was then dried in ambient atmosphere for at least Ih. The surface of the SC sample was pre-wetted with 10 pl per solution of a mixture of water and n-propanol (8:2) before being conditioned for 8h in a glove box in controlled atmosphere (30°C, 30%RH). Control TEWL measurement "TO" were recorded.
[0076] SC samples were then immersed in the test compositions diluted 4-fold (25%) with the Evian water for 2h with gentle stirring on shaker and rinsed by three successive washes of Evian water. The samples were left at room temperature for at least 4h to ensure drying. The SC samples were then placed in the temperature and humidity control chamber for equilibrium at 30°C and 30% RH. After overnight equilibrium, TEWL measurements “Tl” were recorded.
Zein score
[0077] The harshness of surfactants in the compositions were evaluated by employing the zein solubility test, wherein, zein, a yellow corn protein, similar to
keratin present in the skin, with limited solubility in water is denatured (solubilized) by the surfactant. The ability of surfactants to denature and solubilize zein had been linked to their skin-irritation potential. The soluble zein protein was measured by modified Lowry method using spectrophotometer. The absorption was correlated with the solubility of the zein protein which in turn was considered as a measure of the irritation potential or the harshness of the product. The harshness of the analyzed samples were graded and compared using a 1.5% solution of sodium laureate sulfate (SLS) as a positive control. The harshness data for the test compositions were expressed in terms of percentage of the SLS score.
Appearance and Textural analysis
[0078] The compositions were analysed for their appearance and texture using Datacolor light box. The test samples were placed in transparent glass beaker, against Black background in the datacolor light box. The light source was adjusted, and each of the test compositions were observed for any particles or haziness or translucency.
Consumer feedback
[0079] Consumer study was conducted to understand the cosmetic potential of (Dry or wet face leave on) the compositions. Study was conducted on 16 consumers, aged 18-24 years. The consumers were asked to provide feedback on the texture, radiance, and glow.
Instrumental Test
[0080] Antimicrobial activity of the composition was evaluated by standard protocol ASTM 2783 using test organism Cutibacterium acnes ATCC 11827 at a temperature of 25°C±1°C. The test compositions were NEAT C(7) and C(7) with 75% dilution in water. The % germ kill activity was measured for a time period of 1 min.
Example 3
Results
Viscosity measurements
[0081] The viscosities of the compositions of the present disclosure and the comparative compositions were determined for various time period and at varying temperatures. The viscosity values obtained are as shown in Table 3 below.
Table 3
ND-not determined
[0082] It could be observed that the viscosities of the compositions of the present disclosure are within the desired range of 400 to 3000 cps, whereas the comparative compositions exhibited very low viscosity, hence was free flowing and not suitable for easy spread and retention on the skin. The favourable viscosity of the composition of the present disclosure, could be attributed to the combination of the anionic and non-ionic surfactants. It could be noted that CP(1) and CP(2) were devoid of non-ionic surfactants and the CP(3) was devoid of anionic surfactant such as sulphonate surfactant unlike the compositions of the present disclosure. The compositions of the present disclosure was found to exhibit serum like appearance with desired viscosity for easy spreading on wet as well as dry skin. Also, the composition of the present disclosure provided the possibility of leaving the
composition on the skin for a time period of 1 to 5 minutes, providing ample time for skin nourishment.
Determination of foam volume
[0083] Foam volume of the compositions of the present disclosure and the comparative compositions were measured and are tabulated below. It was observed that the combination of surfactants such anionic, amphoteric and non-ionic surfactants as disclosed herein provided better foam volume in comparison to the composition without the combination of these surfactants.
Table 4
pH measurement
[0084] pH of the compositions measured are shown in Table 5 below, wherein the compositions were all within the favorable pH in a range of 5 to 5.5 even at increased temperatures.
Table 5
TEWL and Zein measurement
[0085] The composition of the present disclosure and the comparative composition were subjected to TEWL analysis and zein measurement as per protocol explained above and the corresponding values are indicated in Table 6 below.
[0086] The composition of the present disclosure with said surfactant combination did not hamper the integrity of SC barrier and hence showed less TEWL. Further the composition of the present disclosure was milder on skin proteins as it showed lower Zein scores in comparison with the composition without the desired the combination of the surfactants.
Table 6
Appearance and Textural analysis
[0087] Table 7 indicate the observations made for the appearance and the texture of the compositions. It could be clearly understood that the compositions of the present disclosure were transparent and stable, even at higher temperatures for a longer duration of time, thereby providing increased storage stability and shelf-life.
Table 7
Consumer feedback
[0088] 15 out of 16 consumers preferred the composition of the present disclosure against their regular cleansers for benefits on radiance, glow, acne management. Consumers perceived incremental benefits on acne and radiance with the use of the composition of the present disclosure.
Instrumental Test
[0089] The antimicrobial activity study of the composition of the present disclosure, confirmed that the composition was able to efficiently kill the microbes, as a NEAT as well as in its diluted form as shown in Table 8 below. Thus the cleansing composition even when diluted was found to effectively killing microbes, thereby confirming its enhanced cosmetic behaviour.
Table 8
[0090] Accordingly, it was observed that the use of specific ingredients in the disclosed weight ranges as disclosed in various embodiments herein facilitated in achieving the cosmetic composition with desired sensorial properties and stability.
