Abstract

L'invention concerne un procédé de pré-concentration et de détection d'au moins une molécule monocaténaire d'acides nucléiques cible, le procédé comprenant des étapes de génération d'un écoulement de liquide comprenant au moins une nanoparticule magnétique dans un micro-canal et une pluralité de molécules monocaténaires d'acides nucléiques sondes attachées à la nanoparticule, de génération d'un champ magnétique alternatif dans la partie du micro-canal au moyen de l'électroaimant,le champ magnétique présentant une intensité et une fréquence adaptées pour entraîner une hyperthermie magnétique des nanoparticules de sorte à provoquer une dénaturation du duplex formé par la molécule monocaténaire d'acides nucléiques sonde et par la molécule monocaténaire d'acides nucléiques cible, et une détection de la molécule monocaténaire cible dispersée dans le liquide.

PROCEDE DE DETECTION DE MOLECULES D'ACIDES NUCLEIQUES PAR HYPERTHERMIE MAGNETIQUE ET ENSEMBLE PERMETTANT UNE TELLE
DETECTION DOMAINE DE L' INVENTION
L'invention concerne un procédé de détection d ' une ou plusieurs molécules monocaténaires d 'acides nucléiques, tel que de l'ADN ou de l'ARN , à parti r d ' un échanti llon à tester, et la mise en œuvre de cette détection en uti lisant un biocapteur.
ETAT DE LA TECHNIQUE
L'identification de marqueurs spécifiques de certains types de cancers et de maladies infectieuses peut permettre un diagnostic précoce, peu coûteux, précis, fiable et rapide d ' une pathologie ciblée. Certains acides nucléiques, en particulier certains ARN, par exemple certains micro-ARN , sont des marqueurs connus de pathologies. Par exemple, la détection quantitative de micro-ARN- 122 dans un échantillon à tester peut permettre de diagnostiquer des maladies du foie comme l' hépatite B, C, ainsi que des maladies entraînées par des médicaments.
Pour détecter de basses concentrations en molécules d 'acides nucléiques, par exemple à des concentrations femtomolai res, il est connu de :
• mettre en œuvre une réaction d 'amplification spécifique d ' une ou plusieurs molécules d 'acides nucléiques à tester. De plus, les techniques d 'amplification de type PCR, LAMP ou RPA peuvent entraîner des erreurs de quantification limitant la précision d ' un diagnostic issu du résultat de la détection, en particulier lors de l'amplification de molécules d 'ARN comprenant sensiblement une dizaine de bases.
A cet effet, il est connu d 'utiliser des techniques de détection dites directes, ne nécessitant pas d 'étape d 'amplification, pour détecter et/ou quantifier des molécules d 'acides nucléiques cibles. En particulier, il est connu d 'utiliser des méthodes de détection électrochimique. Horny et al. (Horny, M. C , Lazerges, M. , Siaugue, J. M. , Pallandre, A. , Rose, D. , Bedioui, F. , ... & Gamby, J . (2016), Electrochemical DNA biosensors based on long- range électron transfer: investigating the efficiency of a fluidic channel microelectrode compared to an ultramicroelectrode in a two-electrode setup, Lab on a Chip, 16(22), 4373-4381 ) décrivent l'utilisation d 'un système comprenant deux électrodes agencées dans un micro-canal pour détecter un micro-ARN cible en solution dans le micro-canal. Au moins une des électrodes comprend une monocouche autoassemblée de molécules d 'acides nucléiques sondes. L'hybridation d 'un ou plusieurs brins d 'ARN permet le passage d 'un courant faradique en présence d'un couple redox dans la solution. Ce système permet de détecter des concentrations très basses d 'ARN, par exemple supérieure à un attomolaire.
Toutefois, la concentration en molécules cibles mesurées peut être très dépendante des phénomènes de diffusions en amont de la détection, des phénomènes d 'adsorption des molécules cibles, par exemple sur les parois des conduites permettant aux molécules cibles d 'être transportées jusqu'aux électrodes. Ces effets sont amplifiés lorsque les concentrations en molécules cibles sont basses, et que le rapport entre la surface des parois et le volume de liquide utilisé augmente.
RESUME DE L' INVENTION
Un but de l'invention est de proposer une solution pour détecter une quantité d 'acides nucléiques présente dans un échantillon à tester, y compris à une concentration très faible, par exemple de l'ordre d 'un attomolaire, en limitant ou en évitant les erreurs entraînées par les effets de la diffusion et/ou de l'adsorption des acides nucléiques à tester avant leur détection. Ce but est atteint dans le cadre de la présente invention grâce à un procédé de détection d 'au moins une molécule monocaténaire d 'acides nucléiques cible, le procédé comprenant des étapes de :
a) génération d 'un écoulement de liquide comprenant au moins une nanoparticule magnétique de type cœur-coquille dans un micro-canal d 'un microsystème, et une pluralité de molécules monocaténaires d 'acides nucléiques sondes attachées à la nanoparticule, chaque molécule monocaténaire d 'acides nucléiques sonde étant adaptée pour être au moins partiellement hybridée à une molécule monocaténaire d 'acides nucléiques cible en formant un duplex, au moins une partie du micro-canal étant agencée dans l'entrefer d 'un électroaimant ; b) génération d 'un champ magnétique alternatif dans la partie du micro-canal au moyen de l'électroaimant, le champ magnétique alternatif présentant une intensité et une fréquence adaptées pour entraîner une hyperthermie magnétique des nanoparticules de sorte à provoquer une dénaturation du duplex formé par la molécule monocaténaire d 'acides nucléiques sonde et par la molécule monocaténaire d 'acides nucléiques cible ;
c) détection de la molécule monocaténaire cible dispersée dans le liquide.
Comme les molécules monocaténaires d 'acides nucléiques cibles sont transportées par des nanoparticules magnétiques par l'écoulement jusqu 'à un détecteur, les effets d 'adsorption et de diffusion incontrôlées sont évités. Cela permet de détecter une concentration en molécules monocaténaires d 'acides nucléiques cibles avec une meilleure précision.
De plus, comme la dénaturation est mise en œuvre par hyperthermie magnétique, il est possible de dénaturer des duplex tout en maintenant et/ou en limitant une température moyenne du liquide permettant d 'éviter la dénaturation d 'autres composés biologiques en solution dans le liquide, en particulier en contrôlant une température physiologique du liquide.
L'invention est avantageusement complétée par les caractéristiques suivantes, prises individuellement ou en l'une quelconque de leurs combinaisons techniquement possibles : - le micro-canal présente une géométrie et l'écoulement de liquide présente une vitesse moyenne telles que la ou les nanoparticules sont transportées en moyenne pendant plus de 8 secondes dans la partie du micro-canal ;
- la largeur Le de l'entrefer est inférieure à 5 mm ;
- la nanoparticule comprend un cœur en matériau ferromagnétique ou ferrimagnétique et une coquille en silice entourant le cœur ;
- le cœur de la nanoparticule est en maghémite ;
- le cœur de la nanoparticule présente une longueur et une largeur, et la longueur moyenne du cœur de la ou des nanoparticules est supérieure à 20 nm ;
- le cœur de la ou des nanoparticules comprend un agrégat de noyaux ferromagnétiques ou ferrimagnétiques, l'agrégat comprenant en moyenne au moins deux noyaux ;
- le rapport moyen de la longueur sur la largeur des nanoparticules est supérieur à 1 ,5 ;
- la température de fusion des duplex est adaptée pour que strictement plus de la majorité des duplex soient dénaturés lors de l'étape b) du procédé ;
- l'étape c) de détection de la molécule monocaténaire cible dispersée dans l'écoulement est mise en œuvre par électrochimie en imposant une différence de potentiel électrique entre deux électrodes, les électrodes étant en partie agencées dans le micro-canal ;
- au moins un couple d 'électrodes, comprenant une électrode de travail et une contre-électrode, est agencé dans le micro-canal, en aval de la partie du micro-canal agencé dans l'entrefer de l'électroaimant, dans le sens de l'écoulement ;
- des molécules monocaténaires d 'acides nucléiques sondes sont attachées à une surface de l'une des électrodes, lesdites molécules monocaténaires étant adaptées pour être au moins partiellement hybridées à la ou aux molécules monocaténaires d 'acides nucléiques cibles ;
- l'une des électrodes comprend au moins une couche mince choisie parmi une couche mince d 'or, une couche mince de platine et une couche mince de carbone ;
- le liquide comprend un intercalant redox du duplex et un couple de composés redox ; - le procédé comprend une étape antérieure à l'étape a) dans laquelle la ou les nanoparticules munie(s) des molécules monocaténaires d 'acides nucléiques sondes sont mélangées à un échantillon à tester : comme les molécules monocaténaires d 'acides nucléiques cibles sont pré-concentrées à la surface des nanoparticules magnétiques lors du mélange avec l'échantillon à tester, il est possible détecter une concentration en molécules monocaténaires d 'acides nucléiques cibles avec une meilleure précision.
Un autre objet de l'invention est un ensemble de détection d'au moins une molécule monocaténaire d 'acides nucléiques cible, l'ensemble comprenant :
- un électroaimant présentant un entrefer,
- un micro-canal dans lequel peut s'écouler un liquide, le micro-canal comprenant au moins une partie agencée dans l'entrefer de l'électroaimant, - au moins une nanoparticule magnétique de type cœur-coquille, et une pluralité de molécules monocaténaires d 'acides nucléiques sondes attachées à la nanoparticule, chaque molécule monocaténaire d 'acides nucléiques sonde étant adaptée pour être au moins partiellement hybridée à une molécule monocaténaire d 'acides nucléiques cible en formant un duplex,
l'électroaimant étant propre à générer un champ magnétique alternatif présentant une intensité et une fréquence adaptées pour entraîner une hyperthermie magnétique de la ou des nanoparticule(s) lorsque la ou les nanoparticule(s) sont transportées par le liquide dans la partie du micro-canal agencée dans l'entrefer de l'électroaimant, de sorte à provoquer une dénaturation du duplex formé par la molécule monocaténaire d 'acides nucléiques sonde et par la molécule monocaténaire d'acides nucléiques cible.
L'invention est avantageusement complétée par les caractéristiques suivantes, prises individuellement ou en l'une quelconque de leurs combinaisons techniquement possibles :
- l'ensemble comprend en outre au moins deux électrodes agencées au moins en partie dans le micro-canal, une différence de tension entre les électrodes étant propre à être contrôlée pour détecter par électrochimie la molécule monocaténaire cible dispersée dans l'écoulement ; - l'ensemble comprend des molécules monocaténaires d 'acides nucléiques sondes attachées à une surface de l'une des électrodes, lesdites molécules monocaténaires étant adaptées pour être au moins partiellement hybridées à la ou aux molécules monocaténaires d 'acides nucléiques cibles ;
- l'une des électrodes comprend au moins une couche mince choisie parmi une couche mince d 'or, une couche mince de platine et une couche mince de carbone.
Un autre objet de l'invention est une nanoparticule magnétique de type cœur-coquille comprenant un cœur en matériau ferromagnétique ou ferrimagnétique et une coquille en silice entourant le cœur, dans lequel le cœur de la nanoparticule présente une longueur et une largeur, et dans lequel la longueur du cœur de la nanoparticule est supérieure à 20 nm.
L'invention est avantageusement complétée par les caractéristiques suivantes, prises individuellement ou en l'une quelconque de leurs combinaisons techniquement possibles :
- le cœur est en maghémite ;
- le cœur comprend un agrégat de noyaux ferromagnétiques ou ferrimagnétiques, l'agrégat comprenant au moins trois noyaux ;
- le rapport de la longueur sur la largeur de la nanoparticule est supérieur à 1 ,5 ;
- la nanoparticule est combinée avec une pluralité de molécules monocaténaires d 'acides nucléiques sondes attachées à la nanoparticule ; Un autre objet de l'invention est procédé de fabrication de nanoparticules magnétiques de type cœur-coquille comprenant un cœur en matériau ferromagnétique ou ferrimagnétique et une coquille en silice entourant le cœur, comprenant des étapes de :
a) formation d 'un agrégat d 'une pluralité de noyaux en matériau ferromagnétique ou ferrimagnétique, par dispersion des noyaux dans une solution saline, de manière à obtenir le cœur ;
b) encapsulation du cœur dans la coquille en silice.
Avantageusement, la solution saline présente une force ionique supérieure ou égale à 1 mM. PRESENTATION DES DESSINS

DESCRIPTION DETAILLEE D' UN MODE DE REALISATION DE L' INVENTION
En référence à la figure 1 , des nanoparticules magnétiques en maghémite y- Fe203 peuvent être synthétisées par le procédé dit de Massart, à partir de magnétite colloïdale. La synthèse permettant de produire les nanoparticules illustrées en figure 1 comprend une étape de polycondensation inorganique, dans laquelle on forme deux précurseu rs Fe2(OH )4 et Fe3(OH )6. Quand la concentration en précurseur atteint une valeur limite, des germes de magnétite sont formés (nucléation ) et croissent. La formation des germes est contrôlée par la concentration en précurseur, bloquant la nucléation quand elle atteint une valeur limite inférieure et entraîne la maturation et le vieillissement des germes. Ce procédé est décrit plus en détails par LaMer, V. K. , & Dinegar, R. H. dans Theory, production and mechanism of formation of monodispersed hydrosols, 1950, Journal of the American Chemical Society, 72(1 1 ), 4847-4854.
La taille et la forme des nanoparticules synthétisées peuvent être contrôlées, pour des longueurs inférieures à sensiblement 1 5 nm, par le rapport entre la quantité de Fe (I I ) et de Fe(l l l ), par la température, le pH et la nature de la base utilisée. Les nanoparticules sont ensuite oxydées dans un milieu acide, entraînant la formation de maghémite Y-Fe2C
La taille des nanoparticules et leur charge de surface peuvent être caractérisées par exemple par deux techniques : la diffusion dynamique de la lumière (DLS pour dynamic light scattering en anglais) et la microscopie électronique en transmission (TEM en anglais pour Transmission Electron Microscopy).
La figure 1 est une photographie en TEM d 'un ensemble de nanoparticules magnétiques synthétisées par la méthode précédemment décrite. Ces nanoparticules sont désignées par « noyaux » 21 . On peut introduire dans le milieu de l'acide nitrique de manière à entraîner la précipitation des noyaux 21 de plus grands diamètres, et ainsi les filtrer. La figure 1 illustre les noyaux 1 de plus grands diamètres. La barre d 'échelle de la figure 1 correspond à une longueur de 100 nm. En référence à la figure 2, les noyaux 21 , dont la synthèse est décrite précédemment, présentent un diamètre hydrodynamique moyen de 18, 1 nm (mesuré par DLS) et une longueur moyenne lo de 1 1 nm. Les colonnes (a) de l'histogramme correspondent à une mesure de la longueur (physique) des nanoparticules de l'échantillon, la courbe (b) correspond à un ajustement de la distribution P(l) de la longueur des nanoparticules selon une loi log- normale, dont les paramètres sont la longueur moyenne des nanoparticules et l'écart type σ, selon la formule (2) :
(2) En référence à la figure 3, une nanoparticule magnétique 3 conforme à l'invention peut comprendre un cœur 22 et une coquille 23. Le cœur 22 peut comprendre un ou plusieurs noyaux 21 . La coquille 23 peut être fabriquée par encapsulation du cœur 22 d'une nanoparticule magnétique 3 dans une coquille 23 en silice. Ainsi, la surface extérieure de la nanoparticule magnétique 3 en silice est fonctionnalisable par des méthodes connues, par exemple en utilisant des groupes aminés et/ou carboxyles. De plus, la coquille en silice permet d'écranter les interactions dipolaires des cœurs 22 magnétiques. La coquille peut par exemple être fabriquée en utilisant un procédé sol-gel. Ce procédé comprend d'abord une étape lors de laquelle on condense un monomère de silice (RSi(OR)3) sur la surface d'un noyau 21 . Il s'en suit des étapes d'hydrolyse et de condensation dans un mélange d'eau et d'éthanol. Pendant l'hydrolyse, des groupements silanols sont formés et produisent des groupements siloxanes par condensation intermoléculaire. Un siloxane peut être condensé sur un groupement hydroxyle à la surface du noyau 21 . Les groupements silanols sont ensuite condensés sur la silice et réticulent de manière à former un réseau de silice. Le TEOS, ajouté en premier, permet d'initier la réticulation du réseau de silice et facilite la condensation d'APTS et de PEOS. La réaction est arrêtée en introduisant de l'éther qui entraînent une précipitation des nanoparticules entre-elles, par une réduction de la constante diélectrique du milieu réactionnel. La barre d'échelle de la figure 3 correspond à une longueur de 100 nm.
Le cœur 22 d'une nanoparticule magnétique 3 peut être, de manière plus générale, fabriqué dans un matériau ferromagnétique ou ferrimagnétique. La majorité des nanoparticules magnétiques 3 illustrées dans la figure 3 ont un cœur comprenant un seul noyau 21 .
En référence à la figure 4, les nanoparticules magnétiques 3 de type cœur-coquille, dont la synthèse est décrite précédemment, présentent un cœur 22 de diamètre hydrodynamique moyen sensiblement égal à 25 nm (illustré par la colonne (c) et mesuré par DLS) et de longueur moyenne sensiblement égale à 17 nm. Les colonnes (a) de l'histogramme correspondent à une mesure de la longueur (physique) des nanoparticules de l'échantillon, la courbe (b) correspond à un ajustement de la distribution de la longueur lo des nanoparticules selon une loi log-normale. En référence à la figure 5, les nanoparticules magnétiques 3 de type cœur-coquille, dont la synthèse est décrite précédemment, présentent un diamètre hyd rodynamique moyen sensiblement égal à 45 nm (illustré par la colonne (c) et mesuré par DLS) et de longueur moyenne lo sensiblement égale à 42 nm. Les colonnes (a) de l' histogramme correspondent à une mesure de la longueur (physique) des nanoparticules de l'échantillon, la courbe (b) correspond à un aj ustement de la distribution de la longueur lo des nanoparticu les selon une loi log-normale.
En référence à la figure 6, la tai lle et l'aspect des nanoparticules magnétiques 3 peuvent être modifiés. Selon un mode de réalisation préférentiel de l'invention , l'étape d 'encapsu lation dans une coqui lle 23 de silice est précédée d ' une étape de formation d ' un agrégat 14 d ' une pluralité de noyaux 21 en matériau ferromagnétique ou ferrimagnétique, par dispersion des noyaux 21 dans une solution saline, de manière à former le cœu r 22. Préférentiellement, les nanoparticules magnétiques 3 sont dispersées dans une solution saline dont la force ionique est supérieure ou égale à 1 mM, préférentiellement comprise entre 2 mM et 1 00 mM, et préférentiellement comprise entre 4 mM et 50 mM. On disperse par exemple les nanoparticu les magnétiques 3 dans trois solutions comprenant du chlorure d 'ammonium, aux concentrations de 2 mM, 4 mM et 5 mM. La figure 6 illustre des nanoparticules magnétiques 3 fabriquées selon un procédé conforme à l'invention comprenant une étape de dispersion des noyaux 21 dans une solution de NH4CI à une concentration de 2, 5 mM. Les nanoparticu les magnétiques 3 présentent en majorité deux noyaux 21 ou plus. La barre d 'échelle de la figure 1 correspond à une longueur de 1 00 nm .
En référence à la figure 7, les nanoparticules magnétiques 3 de type cœur-coquille, dont la synthèse est décrite précédemment, présentent un cœur 22 de diamètre hyd rodynamique moyen sensiblement égal à 36 nm (illustré par la colonne (c) et mesuré par DLS) et de longueur moyenne sensiblement égale à 22 nm. En référence à la figure 9, les nanoparticules magnétiques 3 peuvent être fabriquées selon un procédé conforme à l'invention comprenant une étape de dispersion des noyaux 21 dans une solution de NH4CI à une concentration de 4 mM. Les nanoparticu les magnétiques 3 et le cœur 22 des nanoparticules magnétiques 3 présentent une forme elliptique ou ellipsoïdale. La longueu r des nanoparticules magnétiques 3 est en moyenne de 80 nm et la longueur du cœur 22 des nanoparticules magnétiques 3 est en moyenne de 49 nm. D' une manière plus générale, les nanoparticules magnétiques 3 peuvent présenter un rapport de la longueu r su r la largeur supérieu r à 1 , 5. La barre d 'échelle de la figure 1 correspond à une longueu r de 200 nm.
En référence à la figure 1 0, les nanoparticules magnétiques 3 de type cœur-coquille, dont la synthèse est décrite précédemment, présentent un cœur 22 de diamètre hyd rodynamique moyen sensiblement égal à 32 nm (illustré par la colonne (c) et mesu ré par DLS) et une longueu r moyenne lo sensiblement égale à 49 nm. En référence à la figure 12, les nanoparticules magnétiques 3 peuvent être fabriquées selon un procédé conforme à l'invention comprenant une étape de dispersion des noyaux dans une solution de NH4CI à une concentration de 5 mM. Les nanoparticules magnétiques 3 et le cœur 22 des nanoparticules magnétiques 3 présentent une forme elliptique ou ellipsoïdale. La longueur des nanoparticules magnétiques 3 est en moyenne de 86 nm et la longueur du cœur 22 des nanoparticules magnétiques 3 est en moyenne de 55 nm. La barre d 'échelle de la figure 1 correspond à une longueu r de 200 nm.
En référence à la figure 1 3, les nanoparticules magnétiques 3 de type cœur-coquille, dont la synthèse est décrite précédemment, présentent un cœur 22 de diamètre hyd rodynamique moyen sensiblement égal à 62 nm (illustré par la colonne (c) et mesu ré par DLS) et une longueu r moyenne lo sensiblement égale à 55 nm. En référence à la figure 15, le diamètre hydrodynamique d'une nanoparticule 3 peut être contrôlé par la force ionique de la solution saline dans laquelle les nanoparticules 3 sont dispersées avant l'étape de croissance de la coquille 23. Les colonnes (a), (b), (c) et (d) dans la figure 1 5 illustrent le diamètre hydrodynamique moyen de nanoparticules 3 respectivement de type A, B, C et D.
En référence à la figure 16, les nanoparticules 3 conformes à l'invention, et préférentiellement les nanoparticules 3 conformes à l'invention comprenant un cœur comprenant un agrégat 14 de noyaux 21 , peuvent présenter une magnétisation plus élevée que des nanoparticules magnétiques connues, dont le cœur comprend un seul noyau et sans coquille. La magnétisation est mesurée en phase liquide en fonction du champ magnétique appliqué. Les courbes (a), (b), (c) et (d) dans la figure 16 illustrent la magnétisation de nanoparticules 3 respectivement de type A, B, C et D. La courbe (e) illustre la magnétisation de nanoparticules dont le cœur comprend un seul noyau et sans coquille. Les mesures de magnétisation sont réalisées en utilisant un magnétomètre de type SQUI D (de l'anglais Superconducting QUantum Interférence Device).
En référence à la figure 1 7, les nanoparticules 3 conformes à l'invention, et préférentiellement les nanoparticules 3 conformes à l'invention comprenant un cœur comprenant un agrégat 14 de noyaux 21 , peuvent présenter une puissance de chauffage plus élevée que celle de nanoparticules magnétiques connues, dont le cœur comprend un seul noyau et sans coquille. La puissance de chauffage des nanoparticules 3 entraînée par un champ magnétique alternatif est mesurée en utilisant l'appareil Magnetherm de Nanotherics. Un champ magnétique de 10,56 kA. m 1 est appliqué à une fréquence de 535 kHz sur un volume de 500 μί de liquide comprenant les nanoparticules 3, désigné par volume de ferrofluide. Le volume de ferrofluide est introduit dans une bobine en cuivre, qui fait partie d 'un circuit RLC résonnant, adapté à commander un champ magnétique AC dans un gamme de fréquences comprises entre 300 et 550 kHz, et dont l'amplitude peut être par exemple de 10,56 kA. m 1. La bobine est refroidie par une circulation d 'eau de manière à éviter un effet de gradient thermique sur les mesures. La température du volume de ferrofluide est mesurée en utilisant une fibre optique non-métallique disposée au centre de l'échantillon. L'évolution de la température mesurée au cours de l'expérience dT/dt est linéaire et la puissance de chauffage SLP des nanoparticules 3 est calculée selon la formule (3) : SLP = ^ m ^i- . cv.—

Ainsi, la puissance de chauffage SLP peut être augmentée en utilisant des nanoparticules 3 conformes à l'invention, et préférentiellement des nanoparticules 3 dans lesquelles la longueur du cœur 22 de la nanoparticules 3 est supérieure à 20 nm. Ainsi, une hyperthermie magnétique des nanoparticules 3, par exemple de type A, B, C et D illustrées par les points (a), (b), (c) et (d) dans la figure 1 7, commandée par un champ magnétique alternatif permet, dans des conditions prédéterminées, d'atteindre une température du liquide en contact avec les nanoparticules 3 plus élevée qu'en utilisant des nanoparticules connues, par exemple illustrées par le point (e) dans la figure 17. En effet, l'augmentation de la SLP avec la longueur du cœur 22 d'une nanoparticule 3 peut être expliquée en considérant qu'un ordre magnétique du cœur d'une nanoparticule 3 est gardé, lors de l'agrégation de plusieurs noyaux 21 , par exemple lors de de la formation d'un cœur 22 de forme générale ellipsoïdale plutôt que sphérique.
En référence à la figure 18, les nanoparticules 3 conformes à l'invention peuvent être utilisées pour pré-concentrer, transporter et relarguer des molécules monocaténaires d'acides nucléiques cibles 1 en solution, par exemple des molécules d'ADN ou d'ARN, de manière contrôlée par hyperthermie magnétique. De manière générale, une pluralité de molécules monocaténaires d'acides nucléiques sondes 20 peuvent être attachées à la nanoparticule 3, chaque molécule monocaténaire d'acides nucléiques sonde 20 étant adaptée pour être au moins partiellement hybridée à une molécule monocaténaire d'acides nucléiques cible 1 en formant un duplex.
La capacité d'une nanoparticule 3 à relarguer des molécules monocaténaires d'acides nucléiques cible 1 en solution peut être mesurée en attachant des molécules bicaténaires d'ADN à la surface d'une nanoparticule 3 puis en entraînant la dénaturation des molécules bicaténaires d'ADN en chauffant localement la nanoparticule 3 et/ou la solution entourant la nanoparticule 3 par hyperthermie magnétique. Dans la figure 18, le rapport entre la quantité d'ADN monocaténaire cible 1 relarguée par des nanoparticules 3 par hyperthermie magnétique et la quantité d'ADN bicaténaire initial est mesuré lors de l'utilisation de nanoparticules de type A, de type C comparativement à une dénaturation par chauffage de molécules bicaténaires d'ADN en solution sans nanoparticules (contrôle positif).
L'attache de molécules bicaténaires d'ADN aux nanoparticules 3 peut être réalisée de la manière suivante. Des molécules monocaténaires d'ADN sondes 18 sont hybridées avec des molécules monocaténaires d'ADN cible 1 de manière à former un duplex, dans une solution tampon MOPS présentant une concentration en NaCl égale à 0,5 M. Ensuite, 100 équivalents de EDC et de NHS sont introduits en solution. Une attente de 20 minutes permet d'activer les fonctions carboxy des molécules monocaténaires d'ADN sondes 18. Enfin, une solution comprenant des nanoparticules 3 de type A ou C est introduite dans le milieu réactionnel. Des liaisons peptidiques de type EDC/NHS sont alors formées entre des terminaisons N H2 formées au préalable sur la surface en silice des coquilles 23 et les molécules monocaténaires d'ADN sondes 18, hybridées aux molécules monocaténaires d'ADN cibles 1 . Le nombre de duplex attachés sur une surface est maximisé. Une nanoparticule 3 de type A peut présenter sensiblement 440 molécules monocaténaires d'ADN sondes 18 attachées à sa surface et une nanoparticule 3 de type C peut présenter sensiblement 1 100 molécules monocaténaires d'ADN sondes 18 attachées à sa surface.
Avant de quantifier le nombre de molécules monocaténaires d'ADN cible 1 qui peuvent être relarguées par les nanoparticules 3, un témoin positif est réalisé en conditionnant des duplex libres en solution à une température moyenne de 95° C pendant 20 min, puis en quantifiant le nombre de molécules monocaténaires d'ADN cibles 1 en solution. Le témoin positif correspond à la colonne « 95° C » dans la figure 1 : le rapport entre le nombre de molécules monocaténaires d'ADN cible 1 quantifié en solution et le nombre de duplex introduits initialement est sensiblement égal à 100 %.
La colonne A de l'histogramme illustre un rapport sensiblement égal à
50 % entre le nombre de molécules monocaténaires d'ADN cibles 1 quantifiées en solution après un conditionnement des nanoparticules 3 de type A dans un champ magnétique alternatif entraînant une hyperthermique magnétique, et le nombre de duplex initialement présents sur les nanoparticules 3 de type A.
La colonne C de l'histogramme illustre un rapport sensiblement égal à
88 % entre le nombre de molécules monocaténaires d'ADN cibles 1 quantifiées en solution après un conditionnement des nanoparticules 3 de type C dans un champ magnétique alternatif entraînant une hyperthermique magnétique, et le nombre de duplex initialement présents sur les nanoparticules 3 de type C.
Ainsi, il est possible de relarguer de manière contrôlée par hyperthermie magnétique une proportion supérieure ou égale à 50%, et préférentiellement supérieure ou égale à 80% des molécules d'ADN monocaténaires cibles 1 initialement présentes sur des nanoparticules 3. Préférentiellement, les nanoparticules 3 de types C, et plus généralement les nanoparticules présentant un cœur 22 formé par un agrégat de noyaux 21 , sont adaptées d 'une part pour présenter une quantité plus élevée de duplex d 'acides nucléiques sur une même nanoparticule 3 que les nanoparticules 3 de type A (par exemple en moyenne 100 duplex d 'ADN par nanoparticule 3 de type A au regard de 140 duplex d 'ADN par nanoparticule de type C), et d 'autre part à relarguer une proportion d 'acides nucléiques plus importante par nanoparticule.
En référence à la figure 19, le relargage contrôlé de molécules d 'acides nucléiques cibles 1 par hyperthermie magnétique peut être réalisé en limitant la variation de température de la solution comprenant les nanoparticules 3 à 10° C, préférentiellement à 5° C. La courbe de la figure 19 illustre le maintien ou la limitation de la température de la solution comprenant des nanoparticules 3 de type C pendant la commande d 'un champ magnétique alternatif entraînant une hyperthermie magnétique des nanoparticules 3 de type C. L' hyperthermie magnétique des nanoparticules 3 peut entraîner un conditionnement à une température supérieure à la température de fusion des duplex de la couche de liquide entourant une nanoparticule 3, l'épaisseur de la couche étant par exemple inférieure à 10 nm. En revanche, la température moyenne de la solution comprenant les nanoparticules 3 varie entre 22° C et 28° C. Ainsi, le relargage de molécules d 'acides nucléiques cibles 1 par hyperthermie magnétique conforme à l'invention peut être réalisé à une température moyenne de la solution inférieure à la température de fusion des duplex, et préférentiellement à une température physiologique et/ou ambiante. I l est ainsi possible d 'éviter la dénaturation d 'autres espèces biologiques présentes dans une solution à analyser comprenant les molécules d 'acides nucléiques cibles 1 .
En référence à la figure 20, un relargage contrôlé de molécules monocaténaires d 'acides nucléiques cibles 1 peut être réalisé dans un écoulement 5 de liquide 12. La figure 20 illustre un ensemble de détection de molécules monocaténaires d 'acides nucléiques cibles 1 comprenant un électroaimant 10 présentant un entrefer 9. L'électroaimant 10 peut être paramétré tel que décrit dans Lacroix, L. M. , Carrey, J. , & Respaud, M. (2008), A frequency-adjustable electromagnet for hyperthermia measurements on magnetic nanoparticles, Review of scientific instruments, 79(9), 093909. Préférentiellement, la largeur de l'entrefer est inférieure à 5 mm. Ainsi, l'intensité du champ magnétique commandé par l'électroaimant 10, évoluant selon l'inverse de la largeur de l'entrefer 9, peut être adaptée à entraîner la fusion d 'un duplex 4 d 'acides nucléiques attaché à une nanoparticule magnétique 3 par hyperthermie magnétique. L'ensemble comprend également un microsystème 7. Le microsystème 7 comprend au moins un micro-canal 2, reliant fluidiquement au moins une entrée 24 et une sortie 25. Au moins une partie 1 1 du micro-canal 2 est agencée dans l'entrefer de l'électroaimant 10, c'est-à-dire dans le volume défini par l'entrefer 9 de l'électroaimant 10. De fait, le microsystème 7 est adapté à être inséré dans l'entrefer de l'électroaimant 10. Le champ magnétique peut par exemple être généré dans un entrefer 9 d 'une largeur de 1 ,5 mm. Le cœur de l'électroaimant 10 peut être une bague ouverte de ferrite à faible perte, sur laquelle un fil de Litz est enroulé de manière à former n = 55 tours. La bobine de magnétisation peut être couplée à un condensateur de 1 nF de manière à former un circuit LC présentant une fréquence de résonance sensiblement égale à 180 Hz. Le circuit résonant est excité à la fréquence de résonance par un générateur de signal électrique couplé à un amplificateur de puissance RF, et permet par exemple de produire un champ magnétique d 'une amplitude de 370 oe (mesuré en utilisant une bobine à tour unique).
Le micro-canal 2 peut par exemple être fabriqué par gravure humide isotrope d 'une lame en verre par une solution d 'acide fluorhydrique (HF). La lame gravée peut être superposée et scellée à une autre lame en verre, de manière à former la paroi d 'un micro-canal 2, par exemple d ' une largeur de 300 μιη et par exemple d 'une hauteur de 50 μιη. Chaque lame peut par exemple présenter une épaisseur de 300 μιη, et l'épaisseur totale du microsystème 7 une épaisseur de 600 μιη. Ainsi, le microsystème 7 est adapté à être agencé dans l'entrefer 9 d 'un électroaimant 10, la largeur de l'entrefer étant par exemple de 1 mm. Le micro-canal 2 peut également être fabriqué en utilisant les techniques de microfabrication par photolithographie molle, par exemple en utilisant des couches de PDMS microstructurées, et/ou des techniques de thermo-moulage, par exemple en utilisant des matériaux tels que le COC.
L'électroaimant 1 0 comprend un cœur magnétique et une bobine (non représentée) formant un nombre entier n de tours autour du cœur de l'électroaimant. Le champ magnétique He peut être donné par la formule (4) suivante :

Le où μ0 est la perméabilité magnétique du vide et S la surface des spires. Ainsi, l'intensité du champ magnétique peut être préférentiellement supérieure à 150 G, préférentiellement supérieure à 250 G et préférentiellement supérieure à 370 G dans l'entrefer 9. Cette intensité du champ magnétique est particulièrement adaptée à entraîner une hyperthermie magnétique des nanoparticules magnétiques 3 en solution.
En référence à la figure 21 , le micro-canal 2 présente une partie 1 1 agencée dans l'entrefer 9 ou adaptée à être agencée dans l'entrefer 9 de l'électroaimant 10. La figure 21 illustre une vue de dessus d'un micro-canal 2. Le micro-canal 2 comprend une entrée 24 et une sortie 25. Préférentiellement, la géométrie du micro-canal 2 peut être optimisée de manière à ce que le volume défini par le micro-canal 2 soit maximisé dans l'entrefer 9. Une telle optimisation peut être par exemple mise en œuvre par une partie 1 1 du micro-canal 2 en serpentins dans l'entrefer 9. La direction de l'écoulement 5 est illustrée par une flèche noire.
En référence à la figure 22, l'hyperthermie magnétique des nanoparticules magnétiques 3 peut permettre de détecter une molécule monocaténaire d'acides nucléiques cible 1 .
Lors d'une étape 101 , des nanoparticules magnétiques 3 sur lesquelles sont attachées des molécules monocaténaires d'acides nucléiques sondes 18 peuvent être introduites dans un échantillon à tester. Les molécules cibles peuvent être en particulier de molécules de micro-ARN, comme des molécules de micro-ARN 122, dont la présence peut permettre de diagnostiquer des maladies du foie comme l'hépatite B, C, ainsi que des maladies entraînées par des médicaments. Les molécules monocaténaires d'acides nucléiques cibles 1 peuvent, lors de cette étape, s'hybrider sur les molécules monocaténaires d'acides nucléiques sondes 18 attachées sur les nanoparticules magnétiques 3. Ainsi, des molécules monocaténaires d'acides nucléiques cibles 1 présentes en très faible concentration dans un échantillon à tester peuvent être attachées au même endroit, c'est-à-dire sur une ou plusieurs nanoparticules magnétiques 3.
Lors d'une étape 102 du procédé, on génère un écoulement 5 d'un liquide 12 comprenant au moins une nanoparticule magnétique 3. Selon la présence ou non d'une pathologie, ou plus généralement d'un événement physiologique à détecter, les nanoparticules magnétiques 3, peuvent présenter à leurs surfaces des molécules monocaténaires d'acides nucléiques sondes et/ou des duplex 4 formés par une molécule monocaténaire d'acides nucléiques sonde 20 et par une molécule monocaténaire d'acides nucléiques cible 1 . L'écoulement 5 de liquide 12 peut par exemple être contrôlé par un pousse-seringue ou par un contrôleur de pressions entre l'entrée 24 et la sortie 25 du micro-canal 2.
Lors d'une étape 103 du procédé, on génère un champ magnétique alternatif dans la partie 1 1 du micro-canal 2. Ce champ magnétique alternatif peut être préférentiellement généré au moyen de l'électroaimant 10 décrit précédemment. Le champ magnétique alternatif est adapté pour entraîner une hyperthermie magnétique des nanoparticules magnétiques 3 de sorte à provoquer une dénaturation du ou des duplex 4. Préférentiellement, la géométrie du micro-canal 2, en particulier dans la partie 1 1 , ainsi que la vitesse moyenne de l'écoulement des nanoparticules magnétiques 3 dans le liquide 12 sont adapté pour que les nanoparticules magnétiques 3 soient transportées, en moyenne, au moins 8 secondes, préférentiellement 9 secondes et préférentiellement 10 secondes, dans la partie 1 1 du micro-canal 2. On désigne ce temps par « temps de résidence » dans la partie 11 . En effet, la dénaturation d'un duplex présente un temps minimum, en dessous duquel, à une température permettant la dénaturation du duplex, le duplex n'est pas encore dénaturé. Il est alors nécessaire de conditionner les duplex à une température adaptée à la dénaturation plus longtemps que ce temps minimum, compris entre 8 et 12 secondes, préférentiellement compris entre 9 et 1 1 secondes. Ainsi, la géométrie du micro-canal 2 dans l'entrefer 9 et la vitesse moyenne de l'écoulement 5 permettent de dénaturer les duplex 4 et de relarguer les molécules monocaténaires d 'acides nucléiques cibles 1 en solution, dans le liquide 12 de l'écoulement 5. Le temps de résidence moyen des nanoparticules magnétiques 3 dans l'écoulement 5 peut être calculé par la formule (5) suivante :
où d est la longueur du micro-canal 2 dans la partie 1 1 dans le sens de l'écoulement 5, h est la hauteur du micro-canal 2, w est la largeur du microcanal 2 et v est la vitesse moyenne de l'écoulement.
Lors d 'une étape 104 du procédé, on détecte la ou les molécules monocaténaires d 'acides nucléiques cibles 1 relargué en solution lors de l'étape 103. On entend par « détecter » réaliser une mesure permettant de prendre connaissance de la présence ou non de molécule monocaténaire d 'acide nucléique cible 1 en solution, et en particulier mesurer la concentration en molécule monocaténaire d 'acide nucléique cible 1 en solution. Les moyens de détection des molécules monocaténaires d 'acides nucléiques cibles 1 sont décrit ultérieurement.
En référence à la figure 23, la proportion de molécules monocaténaires d 'acides nucléiques cibles 1 relarguées dans un micro-canal 2 peut évoluer avec le débit de l'écoulement 5. La figure 23 illustre la proportion entre la quantité de molécules monocaténaires d 'ADN cibles 1 relarguées et la quantité de duplex présents sur les nanoparticules magnétiques 3 avant l'étape de génération d ' un champ magnétique du procédé. La concentration d 'ADN en solution est mesurée, par exemple dans l'étape de détection de la ou les molécules monocaténaires d 'acides nucléiques cibles 1 , par fluorimétrie. La quantification d 'ADN peut par exemple être réalisée en utilisant un fluorimètre présentant une longueur d 'onde d 'excitation de 480 nm et une longueur d'onde d'émission de 520 nm, et en utilisant un marqueur de molécules monocaténaires d'acides nucléiques en solution tel que l'Oligreen (marque déposée).
La colonne (a) de l'histogramme illustre la proportion d'ADN monocaténaire libre en solution quantifiée lors d'un contrôle négatif, dans lequel l'étape de génération d'un champ magnétique n'est pas mise en œuvre. La colonne (b) de l'histogramme illustre la proportion d'ADN monocaténaire libre en solution quantifiée lors d'une dénaturation de duplex 4 entraînée par un chauffage à 95° C d'une solution en volume statique. La colonne (c) de l'histogramme illustre la proportion d'ADN monocaténaire libre en solution quantifiée lors d'une dénaturation entraînée par hyperthermie magnétique en volume statique, comme décrit précédemment. La colonne (d) de l'histogramme illustre la proportion d'ADN monocaténaire libre en solution quantifiée lors d'une dénaturation entraînée par hyperthermie magnétique selon un procédé conforme à l'invention dans un micro-canal 2 dans lequel un écoulement 5 de liquide 12 comprenant les molécules monocaténaires d'acides nucléiques cibles 1 est généré à un débit moyen de 0,1 1 μί.ε 1. La proportion d'ADN relargué correspondant à la condition (d) est sensiblement égale à la proportion mesurée lors du contrôle négatif. Le temps de résidence entre les conditions (d) et (e) est respectivement de 3 secondes et de 12 secondes. Ainsi, il est possible de détecter un relargage d'ADN sensiblement égal aux conditions (b) et (c) pour un temps de résidence suffisant, c'est-à-dire supérieur à 8 secondes, en adaptant le débit de l'écoulement 5 pour un même micro-canal 2. En référence à la figure 24, l'étape de détection peut être mise en œuvre par détection électrochimique. Horny et al. (Horny, M. C , Lazerges, M. , Siaugue, J. M. , Pallandre, A. , Rose, D. , Bedioui, F. , ... & Gamby, J. , 2016, Electrochemical DNA biosensors based on long-range électron transfer: investigating the efficiency of a fluidic channel microelectrode compared to an ultramicroelectrode in a two-electrode setup, Lab on a Chip, 16(22), 4373- 4381 ) décrivent une méthode adaptée à la détection électrochimique de molécules monocaténaires d 'acides nucléiques cibles 1 dans un micro-canal 2. Ainsi, il est possible de détecter des concentrations basses, par exemple comprises entre 1 aM et 1 μΜ de molécules monocaténaires d 'acides nucléiques cibles 1 par détection électrochimique dans un micro-canal 2. La figure 24 illustre un système dans lequel des électrodes 1 5 sont agencées dans le micro-canal 2 en aval de la partie 1 1 du micro-canal 2 par rapport au sens de l'écoulement 5.
Les électrodes peuvent par exemple être déposées sur la partie d 'une galette de verre correspondant au micro-canal 2. Des couches minces de platine et/ou de titane et/ou d 'or et/ou de carbone peuvent être déposées, par exemple par un procédé de dépôt par pulvérisation cathodique, ou plus généralement par un dépôt physique en phase vapeur. Ainsi, une paroi du micro-canal 2 peut présenter des électrodes.
Des molécules monocaténaires d 'acides nucléiques sondes 18 peuvent attachées à la surface d 'au moins une électrode 1 5, lesdites molécules monocaténaires étant adaptées pour être au moins partiellement hybridées à la ou aux molécules monocaténaires d 'acides nucléiques cibles 1 . Ces molécules monocaténaires d'acides nucléiques sondes 18 peuvent par exemple être attachées à une couche de l'électrode 1 5 en or par l'intermédiaire de groupements thiols, de manière à former une couche autoassemblée. Toutefois, ce type de couches présente un taux maximum d 'hybridation avec des molécules monocaténaires d 'acides nucléiques cibles 1 relativement bas, par exemple inférieur à 1 5 %. A cet effet, des molécules monocaténaires d'acides nucléiques sondes 18 peuvent être préférentiellement attachées à une couche mince de carbone, par exemple en carbone amorphe de type a- CNx, de manière à augmenter le taux de molécules monocaténaires d'acides nucléiques cibles 1 maximum hybridées.
Les différentes électrodes 15 peuvent être reliées à un ou plusieurs générateurs de tension, commandés par une unité de contrôle, de manière à imposer une différence de potentiel électrique entre une électrode de travail 16 et une contre-électrode 17.
En référence à la figure 25, l'impédance entre une électrode de travail 16 et une contre-électrode 17 dépend de l'état d'hybridation des molécules monocaténaires d'acides nucléiques sondes 18 attachées en surface des électrodes 15. En particulier, l'étape de détection des molécules monocaténaires d'acides nucléiques cibles 1 dispersées dans le liquide 12 peut être réalisée en introduisant un électrolyte comprenant un couple de composés redox Fe'"C N637 Fe"C N64" , du bleu de méthylène comme intercalant redox du duplex, et du NaCl à une concentration de 0,5 M. La courbe (a) correspond à une mesure de l'impédance mesurée avant un relargage de molécules monocaténaires d'acides nucléiques cibles 1 par hyperthermie magnétique, et la courbe (b) correspond à une mesure de l'impédance mesurée après un relargage de molécules monocaténaires d'acides nucléiques cibles 1 par hyperthermie magnétique, selon un procédé conforme à l'invention, lors de laquelle le transfert de charge est facilité par la formation de duplex 4.

Claims

REVENDICATIONS
1. Procédé de détection d'au moins une molécule monocaténaire d'acides nucléiques cible (1), le procédé comprenant des étapes de : a) génération d'un écoulement (5) de liquide (12) comprenant au moins une nanoparticule (3) magnétique de type cœur-coquille dans un micro-canal (2) d'un microsystème (7), et une pluralité de molécules monocaténaires d'acides nucléiques sondes (20) attachées à la nanoparticule (3), chaque molécule monocaténaire d'acides nucléiques sonde (20) étant adaptée pour être au moins partiellement hybridée à une molécule monocaténaire d'acides nucléiques cible (1) en formant un duplex, au moins une partie (11) du micro-canal (2) étant agencée dans l'entrefer d'un électroaimant (10) ; b) génération d'un champ magnétique (6) alternatif dans la partie (11) du micro-canal (2) au moyen de l'électroaimant (10), le champ magnétique (6) alternatif présentant une intensité et une fréquence adaptées pour entraîner une hyperthermie magnétique des nanoparticules (3) de sorte à provoquer une dénaturation du duplex formé par la molécule monocaténaire d'acides nucléiques sonde (20) et par la molécule monocaténaire d'acides nucléiques cible (1) ; c) détection de la molécule monocaténaire cible (1) dispersée dans le liquide (12).
2. Procédé selon la revendication 1, dans lequel le micro-canal (2) présente une géométrie et l'écoulement (5) de liquide (12) présente une vitesse moyenne telles que la ou les nanoparticules (3) soient transportées en moyenne pendant plus de 8 secondes dans la partie (11) du micro-canal (2).
3. Procédé selon la revendication 1 ou 2, dans lequel la largeur Le de l'entrefer est inférieure à 5 mm.
4. Procédé selon l'une des revendications 1 à 3, dans lequel la nanoparticule (3) comprend un cœur en matériau ferromagnétique ou ferrimagnétique et une coquille en silice entourant le cœur.
5. Procédé selon la revendication 4, dans lequel le cœur de la nanoparticule (3) est en maghémite.
6. Procédé selon la revendication 4 ou 5, dans lequel le cœur de la nanoparticule (3) présente une longueur et une largeur, et dans lequel la longueur moyenne du cœur de la ou des nanoparticules (3) est supérieure à 20 nm.
7. Procédé selon l'une des revendications 4 à 6, dans lequel le cœur de la ou des nanoparticules (3) comprend un agrégat (14) de noyaux (21 ) ferromagnétiques ou ferrimagnétiques, l'agrégat (14) comprenant en moyenne au moins deux noyaux.
8. Procédé selon l'une des revendication 4 à 7, dans lequel le rapport moyen de la longueur sur la largeur des nanoparticules (3) est supérieur à 1 ,5.
9. Procédé selon l'une des revendications 1 à 8, dans lequel la température de fusion des duplex (4) est adaptée pour que strictement plus de la majorité des duplex (4) soient dénaturés lors de l'étape b) du procédé.

Procédé selon l' une des revendications 1 à 9, dans lequel, l'étape c) de détection de la molécule monocaténaire cible (1 ) dispersée dans l'écoulement (5) est mise en œuvre par électrochimie en imposant une différence de potentiel électrique entre deux électrodes, les électrodes étant en partie agencées dans le micro-canal (2).

Procédé selon la revendication 10, dans lequel au moins un couple d 'électrodes (1 5), comprenant une électrode de travail (16) et une contre- électrode (17), est agencé dans le micro-canal (2), en aval de la partie (1 1 ) du micro-canal (2) agencé dans l'entrefer de l'électroaimant (10), dans le sens de l'écoulement (5).
12. Procédé selon la revendication 10 ou 11, dans lequel des molécules monocaténaires d'acides nucléiques sondes (18) sont attachées à une surface de l'une des électrodes (15), lesdites molécules monocaténaires étant adaptées pour être au moins partiellement hybridées à la ou aux molécules monocaténaires d'acides nucléiques cibles (1).
13. Procédé selon l'une des revendications 10 à 12, dans lequel l'une des électrodes (15) comprend au moins une couche mince (19) choisie parmi une couche mince d'or, une couche mince de platine et une couche mince de carbone.
14. Procédé selon l'une des revendications 10 à 13, dans lequel le liquide (12) comprend un intercalant redox du duplex et un couple de composés redox.
15. Procédé selon l'une des revendications 1 à 14, comprenant une étape antérieure à l'étape a) dans laquelle la ou les nanoparticules (3) munie(s) des molécules monocaténaires d'acides nucléiques sondes (20), sont mélangées à un échantillon à tester.
16. Ensemble de détection d'au moins une molécule monocaténaire d'acides nucléiques cible (1), l'ensemble comprenant :
- un électroaimant (10) présentant un entrefer (9),
- un micro-canal (2) dans lequel peut s'écouler un liquide (12), le micro-canal comprenant au moins une partie (11) agencée dans l'entrefer de l'électroaimant (10),
- au moins une nanoparticule (3) magnétique de type cœur-coquille, et une pluralité de molécules monocaténaires d'acides nucléiques sondes (20) attachées à la nanoparticule (3), chaque molécule monocaténaire d'acides nucléiques sonde (20) étant adaptée pour être au moins partiellement hybridée à une molécule monocaténaire d'acides nucléiques cible (1) en formant un duplex,
dans lequel l'électroaimant (10) étant propre à générer un champ magnétique (6) alternatif présentant une intensité et une fréquence adaptées pour entraîner une hyperthermie magnétique de la ou des nanoparticule(s) (3) lorsque la ou les nanoparticule(s) sont transportées par le liquide dans la partie (1 1 ) du micro-canal agencée dans l'entrefer de l'électroaimant (1 0), de sorte à provoquer une dénaturation du duplex formé par la molécule monocaténaire d 'acides nucléiques sonde (20) et par la molécule monocaténaire d 'acides nucléiques cible (1 ).
17. Ensemble selon la revendication 1 6, comprenant en outre au moins deux électrodes agencées au moins en partie dans le micro-canal (2), une différence de tension entre les électrodes étant propre à être contrôlée pour détecter par électrochimie la molécule monocaténaire cible (1 ) dispersée dans l'écoulement (5).
18. Ensemble selon la revendication 17, comprenant des molécules monocaténaires d 'acides nucléiques sondes (18) attachées à une surface de l'une des électrodes (1 5), lesdites molécules monocaténaires étant adaptées pour être au moins partiellement hybridées à la ou aux molécules monocaténaires d 'acides nucléiques cibles (1 ).
19. Ensemble selon l'une des revendications 17 et 18, dans lequel l'une des électrodes (1 5) comprend au moins une couche mince (19) choisie parmi une couche mince d 'or, une couche mince de platine et une couche mince de carbone.
