Abstract
Eine Zubereitung mit partikulären Polyquaternium Polymeren lässt sich in einem ölbasierenden Aerosol versprühen und zeigt eine antimikrobielle Wirksamkeit.

Zubereitungen mit partikulären Polyquaternium-6 Polymeren
Die Erfindung ist eine antimikrobiell wirksame, wasserfreie Zubereitung mit partikulären Polyquaternium Polymeren, insbesondere Polyquaternium-6 Polymeren und bevorzugt in Form von Aerosolzubereitungen.
Polyquaternium Polymere sind nach der INCI-Nomenklatur Bezeichnungen für komplexe quartäre Ammonium-Polymerverbindungen. Die verschiedenen Vertreter dieser Gruppe werden durch Ziffern unterschieden. Zum Beispiel Polyquaternium-1 , Polyquaternium-16 (PQ-16), Polyquaternium-42, usw. Als quartäre Ammoniumverbindungen besitzen
Polyquaternium Polymere quartäre Stickstoffatome, d.h. alle vier Wasserstoffatome des Ammonium-Ions sind durch organische Reste ersetzt. Die Verbindungen sind somit kationisch, besitzen also eine positive Ladung.
In der WO 2013052454 A1 ist eine Übersicht bekannter Polyquaternium Polymere und deren Handelsnamen aufgeführt.
Vertreter dieser nach INCI benannten Gruppe sind u. a. wegen ihrer filmbildenden und antistatischen Eigenschaften in vielen Körperpflegeprodukten enthalten, z. B. in Shampoos.
So ist als Polyquaternium Polymer u.a. das Merquat™ 550 Polymer bekannt, INCI Name: Polyquaternium-7.
Polyquaternium-6 Polymer (PQ-6) ist ein polymeres, quaternäres Ammoniumsalz gebildet aus der Homopolymerisation des Diallyldimethylammoniumchlorid (DADMAC) Monomers.
Als PQ-6 Polymer ist Mirapol 100 bekannt.
In der EP 2338570 A1 werden wasserfreie Formulierung zum Entfärben von Haaren beschrieben, die ein Entfärbungsmittel und ein Polymer aus quaternären
Hydroxyethylcellulosepolymeren mit einer kationischen Substitution von Trimetylammonium und Dimethyldodeceylammonium umfassen. Bevorzugt genanntes Polymer ist
POLYQUATERNIUM-67.
Polyquaternium-6 ist hingegen kein Polymer, welches aus quaternären
Hydroxyethylcellulosepolymeren gebildet wird.
In der WO 2009091794 A1 werden Zubereitungen beschrieben, die neben antitranspirant wirksamen Mitteln kationische Polyquaternium Polymere, PQ-6, PQ-7, PQ-10, PQ-1 1 , PQ-22 und insbesondere PQ-28, umfassen.
In der US 20060199899 A1 wird die Herstellung partikulärer Polyquaternium-7 Polymere beschrieben.
Die DE 102012012263 A1 offenbart ein Verfahren zur Herstellung von festem Polyquaternium-1. Das beschriebene Polyquaternium-1 kann zur Herstellung von
pharmazeutischen Zusammensetzungen oder Reinigungszusammensetzungen, wie z. B. Augentropfen, Tränenersatzflüssigkeiten und Desinfektionsmitteln, insbesondere für Kontaktlinsen, verwendet werden. Das Molekulargewicht der PQ-1 Polymere liegt dabei im Bereich von 8.000 bis 15.000 g/mol.
In der EP 1761241 A1 werden antitranspirant wirksame Stoffe auf Basis von
Diallyldimethylammoniumchloriden (DADMAC) und Diallylethylammoniumchloriden
(DADEAC) beschrieben, insbesondere PQ-6 (Merquat® 100, Flocare® C106, Floquat FL45®), Polyquaternium-33 (Floerger FO 4190 VHM®,4550 BPM®), Polyquaternium-5 (Merquat 5®, Reten®) und Polyquaternium-7 (Merquat 550 L®, Salcare Super 7®, Flocare C107®).
Im Stand der Technik sind kosmetische Zubereitungen, beispielsweise als Deodorantien oder Antitranspirantien beschrieben, die ein oder mehrere Polyquaternium Polymere umfassen. Die Polyquaternium Polymere liegen hierbei vornehmlich in gelöster Form vor.
Der Einsatz von gelösten Polyquaternium Polymeren in Aerosolzubereitungen ist jedoch problematisch. Wünschenswert ist es daher partikuläre PQ Verbindungen zur Verfügung zu haben und diese in ölbasierten Aerosolzubereitungen einsetzen zu können.
Polyquaternium Polymere sind wasserlöslich, hygroskopisch. Demzufolge ist das
Vorhandensein von Wasser einerseits problematisch, da sich Partikel als solche nicht darstellen lassen und andererseits die Einbringung in eine lipophile Umgebung nicht gelingt.
Weiter wünschenswert ist es kosmetische Rohstoffe, wie Polyquaternium Polymere, wasserfrei, partikulär und damit rieselfähig zur Verfügung zu stellen umso auch eine einfache und handhabbare Herstellung kosmetischer oder dermatologischer Zubereitungen zu gewährleisten.
Um zudem pflegende Desodorantien auf Ölbasis herzustellen, werden Partikel der Polymere benötigt, da sich beispielsweise die wässrigen PQ-6 Rohstoffe, wie Merquat® 100, eine 40% wässrige Lösung, nicht mit einer Ölbasis mischen lassen.
Wünschenswert ist es daher Polyquaternium Polymere insbesondere in ölbasierten
Aerosolzubereitungen zum Einsatz zu bringen.
Weiterhin ist die Nachfrage nach pflegenden Deodorantien auf Aerosolbasis, die keine Aluminiumchlorohydrate als Antitranspirantwirkstoffe umfassen, groß.
Die desodorierende Leistung von Aerosolzubereitungen gilt es zudem zu verbessern.
Die Erfindung ist eine Zubereitung umfassend ein oder mehrere partikuläre Polyquaternium- Polymere. Ein oder mehrere der PQ-Polymere werden aus der Gruppe der Polyquaternium-6 Polymere gewählt.
Bevorzugt handelt es sich um kosmetische oder dermatologische Zubereitungen in
Aerosolform.
Die partikulären Polyquaternium Polymere, insbesondere die PQ-6 Polymere, sind in erfindungsgemäßen Zubereitungen zu einem Anteil von 0, 1 bis 10 Gew.%, insbesondere in einem Anteil von 0, 15 bis 5 Gew.%, bezogen auf die Gesamtmasse der Wirkstofflösung, enthalten. In Bezug auf die Masse der Gesamtzubereitung eines Aerosols, umfassend auch das Treibgas, liegt der Anteil an PQ Polymeren bevorzugt im Bereich von 0,005 bis 0,1 Gew.%, insbesondere im Bereich von 0,01 bis 0,06 Gew.%.
Die Zubereitung ohne Treibgas wird auch als Wirkstofflösung bezeichnet.
Die Partikelgröße der Polyquaternium Polymer Partikel, insbesondere PQ-6 Partikel, werden bevorzugt im Bereich von 10 bis 2000 μηη, bevorzugt im Bereich von 30 μηη und mehr, insbesondere bis zu einer Partikelgröße von 1000 μηη gewählt.
Vorteilhaft werden weiterhin die Partikelgröße der Polyquaternium Polymer Partikel, insbesondere PQ 6 Partikel, so gewählt, dass mindestens 95 Vol-%, insbesondere 100% der Einzelpartikel einen Durchmesser von größer 10μηη aufweisen. Die maximale Partikelgröße liegt vorzugsweise bei 2000 μηη, besonders bevorzugt bei 1000 μηη. Ganz besonders bevorzugt ist Partikelgemisch, bei dem die 95 Vol-% der Partikel größer als 30 μηη sind und 100% der Partikel kleiner als 1000 μηι sind.
Diese erfindungsgemäßen Partikelgrößen haben den Vorteil unproblematisch einsetzbar zu sein.
Die erfindungsgemäßen Polyquaternium Polymerpartikel lassen sich wasserfrei und rieselfähig herstellen, wie nachstehend beispielhaft dargestellt. Es werden Partikel mit einer definierten Partikelgrößenverteilung bereitgestellt, die eine einfache und handhabbare Herstellung kosmetischer oder dermatologischer Zubereitungen gewährleisten.
Die Herstellung wasserfreier Partikel durch einfache Sprühtrocknung gelingt mit
Polyquaternium Polymeren nicht.
Die Definition der Begriffe partikulär, Partikel, Partikelgrößenverteilung etc. sind wie folgt zu verstehen. Partikel sind dreidimensionale Gebilde. Drei Parameter (Länge, Breite, Höhe) sind in der Regel erforderlich, um eine vollständige Beschreibung von Partikeln geben zu können. Es ist jedoch möglich, ein Partikel durch Angabe einer einzigen Zahl zu
beschreiben, die der Partikelgröße entspricht. Bei der Mehrzahl der Methoden zur
Größenbestimmung wird deshalb davon ausgegangen, dass das zu messende Material kugelförmig ist, da eine Kugel die einzige Form ist, die mittels einer einzigen Zahl (dem Durchmesser) beschrieben werden kann. Diese Vereinfachung ist für den Fachmann üblich und hat sich in verschiedensten Anwendung als ausführbar gezeigt.
Die Partikelgröße ist nach DIN 66160: 1992-09 Absatz 2.2.5„ein Feinheitsmerkmal der Dimension Länge". Gemeint ist damit zunächst der Partikeldurchmesser, was
strenggenommen nur für kugelförmige Partikel gilt bzw. bei isometrischen Partikeln zumindest sinngemäß angewandt werden kann. Im Falle nicht-isometrischer Partikel gilt als Partikelgröße ein Mittelwert der Teilchenabmessungen (Römpp Online 3.10).
Besteht das Produkt aus kugelförmigen Partikeln, so ist die Partikelgröße durch die Angabe des Kugeldurchmessers eindeutig definiert, wie erfindungsgemäß bevorzugt im Bereich von 30 μηη und mehr. Die Mehrzahl der feinkörnigen Materialien besteht zwar nicht aus Kugeln, sondern aus mehr oder weniger unregelmäßig geformten Partikeln, die in einem Extremfall nadeiförmig, im anderen plättchenförmig sein können. Die Dispersitätseigenschaft der Einzelpartikeln lässt sich hier durch das Partikelvolumen und zusätzliche Parameter wie Sphärizität (Kugelähnlichkeit) beschreiben. Anstelle der für Kugeln einparametrigen Funktion erhält man eine vielparametrige Dispersitätsfunktion, deren Bestimmung mit großem messtechnischem Aufwand verbunden ist. Ein derartiger Aufwand ist nur dann gerechtfertigt, wenn er dazu dient, wesentliche Informationen über Produkteigenschaften zu gewinnen.
Daher beschränkt man sich in der Praxis normalerweise und auch erfindungsgemäß auf die Angabe einer einparametrigen Verteilungsfunktion für die Partikelgröße. Erfindungsgemäß ist daher eine Partikelgröße der PQ-Partikel mit der 1 -dimensionalen Parameter des
Durchmessers im Bereich von 10 bis 2000 μηη, bevorzugt im Bereich von 30 μηη und mehr, insbesondere bis zu einer Partikelgröße von 1000 μηη.
Bei dem angegebenen Durchmesser handelt es sich um den volumetrischen Durchmesser, der beispielsweise mittels Laserbeugung bestimmt werden kann.
Vorteilhaft umfasst die erfindungsgemäße Zubereitung als Polyquaternium Polymere nur Polyquaternium-6 Polymere und keine anderen PQ-Polymere.
Die angeführten Partikelgrößen, Partikelgrößenverteilung und Anteilsbereiche beziehen sich dann nur auf die PQ-6 Partikel.
Die Polyquaternium-6 Polymere weisen vorteilhaft ein Molekulargewicht im Bereich von 10.000 bis 1.000.000 g/mol, insbesondere im Bereich von 100.000 bis 200.000 g/mol, wobei PQ-6 Polymere mit einem Molekulargewicht von 150.000 g/mol besonders bevorzugt sind.
Die erfindungsgemäßen Zubereitungen sind bevorzugt Sprays oder Aerosole. Die dazu jeweils notwendigen Begleitsubstanzen, wie Treibgase bei Aerosolen, kann der Fachmann aus den bekannten Mitteln wählen.
Ein Aerosol ist ein disperses System, bei dem ein Feststoff und/oder eine Flüssigkeit in einem Gas äußerst fein verteilt vorliegen. Das Aerosol wird in der Regel erst bei der Anwendung mit Hilfe eines geeigneten Sprühsystems durch Versprühen von Lösungen, Emulsionen oder Suspensionen selbst erzeugt, wozu beispielsweise Sprühdosen verwendet werden können, in denen ein verflüssigtes Druckgas als Treibgas dient. Beim Öffnen des Druckventils entweicht das Gemisch aus Treibmittel und Wirkstofflösung durch eine feine Düse, das Treibmittel verdampft und hinterlässt das fein verteilte Sprühgut als Aerosol. Im anwendungstechnischen Sprachgebrauch wird der Begriff Aerosol vom Fachmann häufig auch im Sinne von Aerosolsprays verwendet, d.h. hier wird unter„Aerosol" nicht der reine Sprühnebel, sondern eine Druckgasverpackung nebst Abgabe- bzw. Dosiervorrichtung und Füllgut verstanden. Das Füllgut umfasst Wirkstofflösung und Treibmittel. Als Wirkstofflösung wird die Summe aller Zubereitungsbestandteile ohne das Treibmittel bezeichnet. Der Begriff Wirkstofflösung wird auch dann verwendet, wenn die Zubereitung z.B. in Form einer Suspension oder Emulsion vorliegt. Als Suspension ist eine Dispersion zu verstehen, bei der die kontinuierliche Phase im flüssigen und die dispergierte Phase im festen Zustand vorliegen.
Unter Aerosolzubereitung bzw. Aerosolprodukt versteht man jeden Behälter aus Metall, Glas oder Kunststoff, einschließlich des darin enthaltenen verdichteten, verflüssigten oder unter Druck gelösten Gases mit oder ohne Flüssigkeit, Paste oder Pulver, der mit einer
Entnahmevorrichtung versehen ist, die es ermöglicht, seinen Inhalt in Form von in Gas suspendierten festen oder flüssigen Partikeln als Schaum, Paste, Pulver oder in flüssigem Zustand austreten zu lassen.
Im erfindungsgemäßen Aerosol wird als Treibgas bevorzugt Propan, Propen, n-Butan, isoButan, iso-Buten, n-Pentan, n-Penten, iso-Pentan, iso-Penten, Methan und Ethan einzeln oder in Kombination eingesetzt. Auch hydrophile Treibgase, wie z. B. Kohlendioxid, können vorteilhaft im Sinne der vorliegenden Erfindung eingesetzt werden, wenn der Anteil an hydrophilen Gasen gering gewählt wird und lipophiles Treibgas (z. B. Propan/Butan) im Überschuss vorliegt.
Der Treibgasanteil beträgt vorteilhaft 40 bis 90 Gew.%, bezogen auf die Gesamtmasse der Zubereitung, inklusive Treibgase.
Besonders bevorzugt sind Propan, Butan, iso-Butan bzw. Mischungen dieser Treibgase. Die genannten Gase können im Sinne der vorliegenden Erfindung jeweils einzeln oder in beliebigen Mischungen zueinander verwendet werden. Bevorzugt werden Mischungen mit
Druckstufen von 1 ,0 bis 3,5 bar eingesetzt. Bevorzugt 2,5 bis 3,0, ganz besonders bevorzugt ist eine Druckstufe von 2,7 bar.
Bevorzugt wird das partikuläre Polyquaternium-6 (PQ-6) mit folgender Partikelgröße gewählt:

A: Lineare PolyDADMAC (Poly(Diallyl Dimethyl Ammoniumchloride)) Partikel sind erhältlich durch Trocknung regulärer flüssiger PQ-6 Polymere (PQ-6). Es werden weiße bis gelbe Partikel erhalten (88-100%).
Die Partikelgrößenverteilung ist in Abbildung 1 dargestellt.
B: Lineare PolyDADMAC Partikel sind erhältlich durch Trocknung regulärer flüssiger PQ-6 Polymere (PQ-6). Es wird ein zermahlenes weißes Pulver erhalten (> 85%). Das Pulver wurde gesiebt mit einer Maschenweite von 100μηη. Teile größer 100μηη sind Agglomerate der Einzelpartikel.
Erfindungsgemäß lassen sich so die PQ Polymere, insbesondere die bevorzugten
Polyquaternium-6- und Polyquaternium-16-Partikel herstellen.
Die erfindungsgemäßen Zubereitungen, in denen die PQ-Partikel zum Einsatz kommen, sind öl basierend.
Eingeschlepptes Wasser kann in der Zubereitung zu geringem Anteil enthalten sein, so dass der Wasseranteil unterhalb von 3 Gew.%, insbesondere weniger als 1 Gew.%, bezogen auf die Masse an Wirkstofflösung, ohne Treibgase, beträgt. Damit ist erfindungsgemäß immer noch eine ölbasierende bzw. wasserfreie Zubereitung umfasst, da das eingeschleppte Wasser keinen Einfluss ausübt.
Öle oder auch Lipide umfassen auch Fette und fettähnliche Stoffe. Für die Kosmetik sind sie vor allem als weichmachende ("emollient") Inhaltsstoffe und als hauteigene Lipide der Hornschicht, die zwischen den Hornzellen lagern, von Bedeutung. Sie befähigen die Haut zur Speicherung von Feuchtigkeit. Neben dem pflegenden Aspekt werden Lipide, Öle den kosmetischen Zubereitungen zugesetzt, um eine bessere Verteilbarkeit auf der Haut zu gewährleisten und um die sensorischen Eigenschaften der Zubereitungen zu verbessern.
Vorteilhaft sind polare Ölkomponenten, die bevorzugt gewählt werden aus der Gruppe der Ester aus gesättigten und/oder ungesättigten, verzweigten und/oder unverzweigten
Alkancarbonsäuren mit einer Kettenlänge von 3 bis 30 C-Atomen und gesättigten und/oder
ungesättigten, verzweigten und/oder unverzweigten Alkoholen mit einer Kettenlänge von 3 bis 30 C-Atomen sowie aus der Gruppe der Ester aus aromatischen Carbonsäuren und gesättigten und/oder ungesättigten, verzweigten und/oder unverzweigten Alkoholen mit einer Kettenlänge von 3 bis 30 C-Atomen. Solche Esteröle können dann vorteilhaft gewählt werden aus der Gruppe Phenethylbenzoat, 2-Phenylethylbenzoat, Isopropyl Lauroyl Sarkosinat, Phenyl Trimethicon, Cyclomethicon, Dibutyladipat, Octylpalmitat, Octylcocoat, Octylisostearat, Octyldodeceylmyristat, Octyldodekanol, Cetearylisononanoat,
Isopropylmyristat, Isopropylpalmitat, Isopropylstearat, Isopropyloleat, n-Butylstearat, n- Hexyllaurat, n-Decyloleat, Isooctylstearat, Isononylstearat, Isononylisononanoat, 2- Ethylhexylpalmitat, 2-Ethylhexyllaurat, 2-Hexyldecylstearat, 2-Octyldodecylpalmitat,
Stearylheptanoat, Oleyloleat, Oleylerucat, Erucyloleat, Erucylerucat, Tridecylstearat, Tridecyltrimellitat, sowie synthetische, halbsynthetische und natürliche Gemische solcher Ester, wie z. B. Jojobaöl.
Weitere bevorzugte Öle sind zu wählen aus der Gruppe Caprylic / Capric Triglyceride, Tri- Isostearin, Octylstearate, Dicaprylyl Carbonate und/oder Shea Butter.
Erfindungsgemäß außerordentlich bevorzugte Ölkomponenten sind ausgewählt aus
Fettsäure- und Fettalkoholestern. In diesem Fall besonders bevorzugt ist Isopropylpalmitat.
Weiterhin bevorzugt ist, dass als Lipidbestandteil der Ölphase ein oder mehrere Pflanzenöle gewählt werden. Bevorzugte Pflanzenöle sind zu wählen aus Mandelöl, Traubenkernöl, und/oder Sonnenblumenöl.
Bevorzugt Lipide sind insbesondere Cyclomethicone, Caprylic/Capric Triglycerid, C12-15 Alkyl Benzoate und/oder Isopropyl Palmitat.
Im Stand der Technik werden leicht flüchtige Silikonöle, wie Cyclomethicone als
Hauptbestandteil in Aerosolsprays eingesetzt. Der Anteil an Cyclomethicone kann jedoch deutlich reduziert werden bzw. Cyclomethicone kann komplett ersetzt werden.
Vorteilhaft kann daher auch auf flüchtige cyclische Silikonöle verzichtet werden.
Bei Einschränkungen auf bevorzugt genannte Stoffe, seien es Lipide oder weitere bevorzugt gennannte Bestandteile, so beziehen sich deren bevorzugten Anteilsbereiche dann auch auf die dann ausgewählten Einzelbestandteile. Die anderen, die durch die Einschränkung ausgeschlossenen Bestandteile, zählen dann nicht mehr zu den aufgeführten
Anteilsbereichen hinzu.
Für eine gute Verarbeitung und Anwendung ist es wichtig, dass die Partikel stabil in der Zubereitung vorliegen. Dazu wurden Analysen zur Löslichkeit und Partikelgröße von
Polyquaternium Polymeren, insbesondere PQ-6, mittels Infrarot-Spektroskopie und Raman- Spektroskopie durchgeführt.
Es wurde dabei bestimmt, ob sich PQ-6 Partikel in der umgebenden Matrix, einer Ölmischung sowie mit und ohne Parfüm verändern, sich lösen oder verkleinern. Die
Veränderungen wurden über einen längeren Zeitraum beobachtet.
Die Ergebnisse der Untersuchungen zeigen vorteilhaft, dass keine gelösten Anteile von PQ-6 in der Ölphase (mit und ohne Parfüm) gefunden werden konnten.
Die Partikelform ändert sich nicht. Die PQ-6 Partikel bleiben partikulär kugelförmig (s.
Abbildung 2).
In weiteren Untersuchungen wurde die antibakterielle Wirksamkeit der erfindungsgemäßen Zubereitungen bestimmt (Abbildungen 3 bis 5).
Der sogenannte ATA Test dient der in-vivo Bestimmung einer antibakteriellen Wirksamkeit über 8 Stunden von kosmetischen Formulierungen nach einmaliger Applikation. Die
Anwendung des Testproduktes und die Messung der Wirksamkeit erfolgen in dem
verbraucherrelevanten Areal, der Achsel. Zielgröße ist die Keimzahl der Achselbakterien.
Die Probanden haben eine Vorkonditionierung von 5 Tagen an denen sie kein Deodorant/ Antitranspirant verwenden dürfen und die Achsel zwei Tage vor dem Teststart rasieren müssen. Nach Waschen der Achseln und Abspülen der Achseln werden 1 ,4 g Aerosol mit vorherigem Schütteln nach Vorschrift aufgetragen. Abgespült wird mit einem Teflonring, Spatel und 1 ml Abspülpuffer für 1 Minute. Die Abspüllösung wird direkt verdünnt und später mittels Spiralplater auf Agarplatten ausplattiert. Das Abspülen wird dann zu den
entsprechenden Zeitpunkten, 1 h, 4h und 8h, wiederholt. Zum Schluss wurden die Platten bei 30°C 48 Stunden bebrütet und anschließend mit Hilfe des Countermaten ausgewertet.
In der Auswertung wurde der Reduktionsfaktor bestimmt. Der Reduktionsfaktor ist der Faktor, um den die Zellzahl (CFU) der unterschiedlichen Zeitpunkte im Vergleich zur TO-Kontrolle oder untereinander reduziert wird.
Das Studiendesign ist so ausgelegt, dass eine antibakterielle Wirksamkeit wie folgt nachgewiesen werden kann:

Getestete Zubereitung ist ein silikonhaltiges Aerosol mit 1 ,2 Gew.% PQ-6 Partikel (Beispiel 2).
Die Ergebnisse sind in der nachfolgenden Tabelle und in Abbildung 3 wiedergegeben.

Die in-vivo Microflora Analyse (Abbildung 3) zeigt die Reduktionsfaktoren der PQ-6 Partikel haltigen Zubereitung in der Achsel nach 1 , 4 und 8h. Ein Reduktionsfaktor größer 10 bedeutet eine Reduktion der Bakterien um 90%.
Da die Polyquaternium Polymere in einer Ölmatrix in die Achsel appliziert wurden, muss zuerst das Öl verdampfen, anschließend muss sich der Partikel im Schweiß lösen um anschließend sein antimikrobielle Wirksamkeit zu entfalten. Außerdem werden Partikel leicht aus der Achsel ins T-Shirt übertragen. Erstaunlich ist daher, dass die Polyquaternium
Polymere, und insbesondere PQ-6, auch aus ihrer partikulären Form heraus antimikrobiell wirken. Durch die positive Ladung von Polyquaternium Polymeren bleibt der Wirkstoff nach dem Lösen im Schweiß an der leicht negativ geladenen Haut haften und kann so seine Wirkung entfalten.
Die Abbildungen 4 und 5 zeigen die synergistische Wirksamkeit von antimikrobiell und antitranspirant wirksamen Mitteln, Aluminiumchlorohydrat (ACH), in Kombination mit partikulären Polyquaternium Partikeln.
Untersucht wurde die antimikrobielle Wirksamkeit zweier Zubereitungen.
Die Zubereitung B ist ein Antitranspirant Aerosol mit 16,6 Gew.% Aluminiumchlorohydrat, bezogen auf die Gesamtmasse der Wirkstofflösung (ohne Treibgas).
Die Zubereitung A umfasst die Zubereitung B sowie zusätzlich 1 ,2 Gew.% an PQ-6 Partikeln, bezogen auf Wirkstofflösung (ohne Treibgas).
Für die Untersuchung wurde das Bakterium C. jeikeium (DSM 7171) aus einem nach EN 12353 angelegten Cryoröhrchen auf einer entsprechenden Agarplatte (C+T-Agar) ausgestrichen und bei 30°C über 48 Stunden inkubiert. Von den gewachsenen Bakterien wurde eine zweite Passage als Flüssigkultur (AC-Medium) angelegt.
Die Flüssigkultur wurde, nach einer OD-Bestimmung, auf 0, 1 eingestellt und 1 :8 mit angedicktem Medium (AC-Medium+0,2% Agar) verdünnt. Diese Bakteriensuspension wurde anschließend für den Test eingesetzt.
Der Schweinehaut-Infektionsassay wurde per Hand durchgeführt. Hierzu wurden 100,5 μΙ Bakterien auf die Haut aufgetragen (Keimzahl von 1 *105/cm2) und gleichmäßig mit einem Spatel verteilt. Nach einer Antrocknungsphase wurden die Well-Platten in eine
Sprühvorichtung gestellt und 1 ,4 g des Aerosols pro Stanze aufgesprüht. Vor jedem Sprühen wurde geschüttelt und vor und nachher gewogen. Die Platten mit der Haut werden für die
entsprechende Zeit bei 30°C und einer Luftfeuchtigkeit von ca. 90% inkubiert. Nach Ablauf der Zeit werden alle Hautstanzen mit 2,01 ml Abspülpuffer bedeckt und für 1 min im
Ultraschallbad beschallt. 200 μΙ der abgespülten Lösungen werden zu 800 μΙ
Enthemmungsmedium pipettiert. Diese Verdünnungen wurden mittels Spiralplater auf Agarplatten ausplattiert und bei 30°C über 48 Stunden bebrütet, anschließend wurde die Keimzahl mit Hilfe des Countermaten ermittelt (s. auch L.W. Bush, L. M. Benson and J. H. White "Pig Skin as Test Substrate for Evaluating Topical Antimicrobial Activity" Journal of Clinical Microbiology, Sept. 1986, p. 343-348). In der Auswertung wurde der
Reduktionsfaktor bestimmt. Der Reduktionsfaktor ist der Faktor, um den die Zellzahl (CFU) der Testsubstanz im Vergleich zur Kontrolle reduziert wird.
Abbildung 4 gibt die Keimzahlen nach 1 , 4 und 8 Stunden gegen die Kontrolle (K) wieder. Abbildung 5 zeigt die Reduktionsfaktoren nach 1 , 4 und 8 Stunden.
Das Resultat zeigt, dass durch das Hinzufügen von partikulären Polyquaternium-6 Polymere eine deutliche Steigerung der antimikrobiellen Wirksamkeit erzielt wird.
Grundsätzlich ist bei in-vitro Untersuchungen die Wirksamkeit deutlich ausgeprägter als bei in-vivo Tests.
Für den Fachmann unerwartet zeigte sich bei Aerosolzubereitungen zudem eine verbesserte antimikrobielle Wirksamkeit, wenn man neben anitranspirant und antimikrobiell wirksamen Mitteln, wie ACH, partikuläre Polyquaternium Polymeren hinzufügt.
Antitranspirantien (AT) sind schweißverhütende Mittel, die - im Gegensatz zu den
Desodorantien, die im Allgemeinen eine mikrobielle Zersetzung von bereits gebildetem Schweiß verhindern - die Absonderung von Schweiß überhaupt verhindern sollen.
Die gewünschte Minimierung der Schweißsekretion kann durch verschiedene Mechanismen realisiert werden.
Neuartige AT-Wirker basieren bspw. auf dem Prinzip der Anticholinergika, welche die Nervenreize, die zur Sekretionssteigerung der Schweißdrüsen führen, unterbrechen.
Ein weiteres Prinzip neuartiger AT-Wirker beruht auf der Beeinflussung von
Membrantransportvorgängen in der Zelle. So hemmen spezifische Aquaporin-Inhibitoren die Proteine, die Kanäle in der Zellmembran bilden, um den Durchtritt von Wasser und weiteren Molekülen zu erleichtern. Auch lonenkanal-Hemmer bewirken eine Beeinflussung von Membrantransport- bzw. Osmosevorgängen. Ein weiterer neuartiger AT-Wirkmechanismus lässt sich durch die Verwendung kurzkettiger, vicinaler Diole realisieren, welcher
möglicherweise auf deren osmotischen Aktivität zurückzuführen ist.
So durch Anwendung eines Mittels kein Einfluss auf die Schweißsekretion ausgeübt wird, also keine antitranspirante Wirkung realisiert wird, liegt erfindungsgemäß kein Antitranspirant vor.
Erfindungsgemäß sind demnach als Antitranspirantwirkstoffe solche Stoffe umfasst, die einen Einfluss auf die Schweißsekretion haben.
Im Gegensatz zu den Antitranspirantien bewirken reine Desodorantien keine aktive
Beeinflussung der Schweißsekretion, sondern lediglich die Steuerung bzw. Beeinflussung des Körper- bzw. Achselgeruches (Geruchsverbesserungsmittel). Gängige
Wirkmechanismen hierzu sind antibakterielle Effekte, wie sie auch das nicht-kolloidale Silber zeigen, Geruchsneutralisation (Maskierung), Beeinflussung von bakteriellen Metabolismen, die reine Parfümierung wie auch die Verwendung von Vorstufen bestimmter
Parfümkomponenten, welche durch enzymatische Umsetzung zu wohlriechenden Stoffen umgesetzt werden.
Zubereitungen können neben den eigentlichen schweißhemmenden Wirkstoffen (AT-Wirker) zusätzlich auch Stoffe enthalten, die den mikrobiellen Abbau des Schweißes hemmen, wie z. B. Butyloctanoic Acid. Butyloctanoic Acid zeigt jedoch nur eine desodorierende Wirkung und keine antitranspirante Wirkung, da aus der Beeinflussung der bakteriellen Hautflora keine Beeinflussung der Schweißsekretion abzuleiten ist.
Antitranspirantwirkstoffe im Sinne der vorliegenden Anmeldung sind insbesondere aus den folgenden Gruppen zu wählen.
Als Antitranspirantwirkstoffe finden insbesondere Adstringenzien Verwendung, vornehmlich Aluminiumverbindungen. Die früher eingesetzten, stark sauer wirkenden Salze
Aluminiumsulfat oder -chlorid und die Agaricinsäure sind weitgehend durch
Aluminiumhydroxychlorid und -alkoholate ersetzt worden. Die nachfolgende Auflistung vorteilhaft einzusetzender Antitranspirant-Wirker soll in keinster Weise einschränkend sein:
Aluminiumsalze:
• Aluminiumsalze wie Aluminiumchlorid AICI3, Aluminiumsulfat AI2(S04)3
• Aluminiumchloride der empirischen Summenformel [AI2(OH)mCln], wobei m+n=6
• Aluminiumchlorhydrat [AI2(OH)5CI] x H20
Standard AI-Komplexe: Locron P (Clariant), Micro-Dry 323 (Summit Reheis), Aloxicoll PF 40 (BK Giulini).
o Aktivierte AI-Komplexe: Reach 103 (Summit Reheis), AACH-7171/AACH-7172 (Summit Reheis), Aloxicoll P (BK Giulini), Aloxicoll SD100 (BK Giulini)
• Aluminiumsesquichlorhydrat [AI2(OH)4,5CI1 ,5] x H20
Standard AI-Komplexe: Aloxicoll 31 P (BK Giulini)
• Aluminiumdichlorhydrat [AI2(OH)4CI2] x H20
Die Aluminiumsalze können in gemahlener Form oder auch als Hohlkugeln oder als
Mischung dieser vorliegen. Die Dichte dieser Partikel liegt vorteilhaft im Bereich von 0,7 bis 2,0 g/cm3.
Vorteilhaft kann auch die Verwendung von AT-Salz-Suspensionen bzw. -Gelen sein, bei denen pulverförmig vorliegende Aluminiumsalze in diversen Ölen dispergiert angeboten werden.
Die Antitranspirantwirkstoffe aus den zuvor geschilderten Gruppen werden in den erfindungsgemäßen Formulierungen bevorzugt in einer Menge von 0,5 bis 10 Gew.%, bevorzugt 2 bis 6 Gew.%, bezogen auf die Gesamtmasse der Zubereitung, d.h. inklusive der ggf. vorhandenen Treibgase, eingesetzt.
Bei Einsatz von ca. 35 Gew.% AACH (aktiviertes Aluminiumchlorohydrat) in der
Wirkstofflösung für ein Aerosolspray (ohne Treibgas) und einem Abfüllverhältnis von etwa 15 : 85 (Wirkstofflösung zu Treibgas) wird ein Anteil von etwa 5,25 Gew. % AACH im
Endprodukt vorhanden sein.
Selbstverständlich ist es möglich, weitere oder andere Antitranspirantwirkstoffe und/oder Deodorantien als kosmetisch wirksamen Bestandteil zu zusetzen.
Die erfindungsgemäßen Zubereitungen sind als antimikrobiell wirksame Mittel und insbesondere als desodorierende Mittel, Desodorantien, anwendbar.
Um in den Aerosolzubereitungen ein Absinken und Absetzen der Partikel zu vermeiden wird vorteilhafterweise der Zubereitung ein oder mehrere Schichtsilikate zugesetzt.
Für den Fachmann unerwartet, zeigte sich bei den erfindungsgemäßen
Aerosolzubereitungen zudem ein besseres Absetzverhalten des partikulären Wirkstoffs im Aerosol bei gleichzeitig sehr guter Packmittelkompatibilität.
Suspendierhilfen erhöhen die Stabilität von Partikeln in Suspensionen. In wasserfreien Antitranspirantformulierungen werden daher vorzugsweise modifizierte Schichtsilikate, Tonmineralien und/oder Kieselsäuren eingesetzt, z. B. Hectorite, Bentonite und/oder Silica.
Bevorzugt sind Disteardimonium Hectorite und/oder Stearalkonium Bentonite zu wählen, bevorzugt zu einem Anteil im Bereich von 2 bis 5 Gew.%, bezogen auf die Masse der Wirkstofflösung.
Im Allgemeinen umfasst das Verfahren zur Herstellung von aus
Diallyldialkylammoniumsalzmonomeren gebildetem Homopoiymer die folgenden Schritte, wie sie auch für die Herstellung der partikulären PQ-6 Polymere angewandt werden können. Zunächst wird eine wässrige Monomerphase beispielsweise in einem Doppeimantelreaktor hergestellt.
Die Monomerphase wird dabei auf eine Temperatur unter etwa 50 ° C erwärmt und ein Radikalinitiator oder eine Mischung von Radikalinitiatoren wird zugegeben, um die
Polymerisation für einen Zeitraum von 0,5 bis 8 Stunden zu initiieren.
Die erhaltenen Homopolymere in Form eines Gels werden durch Erhitzen auf eine
Temperatur im Bereich von etwa 60 bis etwa 120 C für einen Zeitraum von 0,1 bis 4 Stunden nachbehandelt.
Das Gel, das das gewünschte Produkt enthält, wird gekühlt, aus dem Reaktor entnommen und getrocknet.
Die Copolymerisationsreaktion wird im Allgemeinen unter Verwendung von Techniken zur radikalischen Polymerisation initiiert, die dem Fachmann bekannt sind. Verbindungen, die hauptsächlich wasserlösliche Reste bilden, sind als Polymerisationsinitiatoren geeignet. Zum Beispiel können Starter wie 2,2-Azobis- (N, N-dimethylenisobutyramidin) dihydrochlorid, 2,2 mn-Azobis- (2-amidinopropan) dihydrochlorid, 2,2mn-Azobis- (N, N mn- dimethylenisobutyramidin), 4,4mn-Azobis- (4-cyanpentancarbonsäure), 2,2mn-Azobis (4- methoxy-2,4-dimethylvaleronitril), 2.2mn-Azobis (2,4-dimethylvaleronitril), (1-Phenylethyl) Azodiphenylmethan, 2,2mn-Azobisisobutyronitril, Dimethyl-2,2mn-azobisisobutyrat und 2,2mn-Azobis- (2-methylbutyronitril) verwendet werden.
Zusätzlich können Peroxidpolymerisationsinitiatoren verwendet werden, wie beispielsweise Dibenzoylperoxid, Dilaurylperoxid, Di-2-ethylhexylperoxydicarbonat,
Dicyclohexylperoxydicarbonat, Bis- (4-tert-butylcyclohexyl) peroxydicarbonat, tert.
Butylperpivalat, tert. Butylperbenzoat, tert. Butylpermaleinat, Di-tert. Butylperoxid, tert.- Butylhydroperoxid, Wasserstoffperoxid, Ammoniumpersulfat, Kaliumpersulfat,
Natriumpersulfat und Redoxkataiysatoren in Kombination mit Reduktionsmitteln wie Eisen (II) -ammoniumsulfat, Ascorbinsäure, Natriummethylsulfinat, Dinatriumdisulfit und Natrium Hydrogensulfit. Diese Initiatoren können entweder alleine oder in Kombination miteinander verwendet werden.
Die wässrige Monomerphase wird vor der Zugabe des Initiators mit einem Inertgas wie Stickstoff und Argon gespült.
Die wässrige Phase kann auch ein Chelatbildner enthalten, wie beispielsweise ein
wasserlösliches Salz von Ethylendiamintetraessigsäure und Salz von
Diethylentriaminpentaessigsäure (Chelatbildner).
Die erhaltenen Homopolymere in Form von Gelen werden gekühlt, aus dem Reaktor entnommen und getrocknet. Alternativ kann die Gelmischung vor dem Trocknen extrudiert und granuliert werden, um den abschließenden Trocknungsschritt zu unterstützen. Das so hergestellte Produkt kann unter Verwendung von bekannten Trocknungsmitteln getrocknet werden, wie beispielsweise einer Wirbelschicht, einem Trommeltrockner, einem
Sprühtrockner, einer Vakuumtrocknung oder einer Mikrowellentrocknung. Das getrocknete Produkt wird als weißes, körniges Material erhalten.
Die kosmetischen oder dermatologischen Zubereitungen gemäß der Erfindung können ferner kosmetische Hilfsstoffe und Wirkstoffe enthalten, wie sie üblicherweise in solchen Zubereitungen verwendet werden, z. B. Wirkstoffe, Konservierungsmittel, Konservierungshelfer, Bakterizide, weitere Lipide, Substanzen zum Verhindern des Schäumens, Farbstoffe und Farbpigmente, Verdickungsmittel, anfeuchtende und/oder feuchthaltende Substanzen oder andere übliche Bestandteile einer kosmetischen oder dermatologischen Formulierung wie Polyole, Polymere, Schaumstabilisatoren, organische Lösungsmittel oder Silikonderivate, sofern der Zusatz die geforderten Eigenschaften nicht beeinträchtigen oder ausgeschlossen sind.
Nachfolgende Beispiele sollen die vorliegende Erfindung verdeutlichen, ohne sie einzuschränken. Alle Mengenangaben, Anteile und Prozentanteile sind, soweit nicht anders angegeben, auf das Gewicht und die Gesamtmenge der Wirkstofflösung (ohne Treibgase) bzw. auf das Gesamtgewicht der Zubereitung bezogen.
Die Beispiele belegen zudem, dass sich die Polyquaternium Partikel unproblematisch in die Aerosolzubereitung einarbeiten lassen.

Herstellanweisung:
Die Rohstoffe sind zu vermischen und das Schichtsilikat durch hohen Energieeintrag zu aktivieren. Anschließend werden die PQ-6-Partikel hinzugefügt.
Die Formulierung wird in Aerosoldosen mit Treibgas beaufschlagt. Das Treibgas ist zu wählen aus Propan, Butan, Isobutan oder einer beliebigen Mischung dieser Komponenten.

Herstellanweisung:
Die Rohstoffe sind zu vermischen und das Schichtsilikat durch hohen Energieeintrag zu aktivieren. Anschließend werden die PQ-6-Partikel hinzugefügt.
Die Formulierung wird in Aerosoldosen mit Treibgas beaufschlagt. Das Treibgas ist zu wählen aus Propan, Butan, Isobutan oder einer beliebigen Mischung dieser Komponenten.
Claims
1 . Zubereitung umfassend ein oder mehrere partikuläre Polyquaternium Polymere dadurch gekennzeichnet, dass der Wasseranteil in der Zubereitung weniger als 3 Gew.% beträgt, insbesondere weniger als 1 Gew.%, bezogen auf die Gesamtmasse der Zubereitung und ein oder mehrere der partikulären Polyquaternium Polymere aus der Gruppe der
Polyquaternium-6 Polymere gewählt werden.
2. Zubereitung nach Anspruch 1 dadurch gekennzeichnet, dass die Polyquaternium
Polymerpartikel eine Partikelgröße im Bereich von 10 μηη und mehr, insbesondere 30 μηη und mehr, bevorzugt bis 2000 μηη, insbesondere bevorzugt bis 1000 μηη, aufweisen.
3. Zubereitung nach Anspruch 1 dadurch gekennzeichnet, dass mindestens 95 Vol-%, insbesondere 100%, der Einzelpartikel der Polyquaternium Polymerpartikel einen Durchmesser von größer 10μηη aufweisen.
4. Zubereitung nach Anspruch 1 dadurch gekennzeichnet, dass 95 Vol-% der
Polyquaternium Polymerpartikel größer als 30 μηη sind und 100% der Polyquaternium Polymerpartikel kleiner als 1000 μηη sind.
5. Zubereitung nach einem der vorstehenden Ansprüche dadurch gekennzeichnet, dass als Polyquaternium Polymere nur Polyquaternium-6 Polymere gewählt werden.
6. Zubereitung nach einem der vorstehenden Ansprüche dadurch gekennzeichnet, dass die Polyquaternium-6 Polymere ein Molekulargewicht im Bereich von 10.000 bis 1.000.000 g/mol, insbesondere im Bereich von 100.000 bis 200.000 g/mol, insbesondere 150.000 g/mol, aufweisen.
7. Zubereitung nach einem der vorstehenden Ansprüche in Form eines Sprays oder
Aerosols.
8. Zubereitung nach Anspruch 7 umfassend Treibgase dadurch gekennzeichnet, dass der Treibgasanteil im Bereich von 40 bis 90 Gew.%, bezogen auf die Gesamtmasse der Zubereitung inklusive Treibgase, gewählt wird.
9. Zubereitung nach einem der vorstehenden Ansprüche dadurch gekennzeichnet, dass die partikulären Polyquaternium Polymere zu einem Gewichtsanteil von 0, 1 bis 10 Gew.%, insbesondere zu einem Anteil von 0, 15 bis 5 Gew.%, bezogen auf die Gesamtmasse der Zubereitung ohne Treibgase, der Wirkstoff lösung, enthalten sind.
10. Zubereitung nach einem der vorstehenden Ansprüche dadurch gekennzeichnet, dass ein oder mehrere Lipide gewählt aus der Gruppe Cyclomethicone, Caprylic/Capric Triglycerid, C12-15 Alkyl Benzoate und/oder Isopropyl Palmitat enthalten sind.
1 1. Zubereitung nach einem der vorstehenden Ansprüche dadurch gekennzeichnet, dass ein oder mehrere antitranspirant wirksame Stoffe enthalten sind, insbesondere
Aluminiumchlorohydrate.
12. Zubereitung nach einem der vorstehenden Ansprüche dadurch gekennzeichnet, dass ein oder mehrere Schichtsilikate, insbesondere Disteardimonium Hectorite und/oder Stearalkonium Bentonite, enthalten sind.
13. Zubereitung nach Anspruch 12 dadurch gekennzeichnet, dass der Anteil an
Schichtsilikaten im Bereich von 2 bis 5 Gew.%, bezogen auf die Masse der Zubereitung ohne Treibgase, der Wirkstofflösung, gewählt wird.
14. Verwendung der Zubereitung nach einem der vorstehenden Ansprüche als
antimikrobiell wirksames Mittel.
15. Nicht-therapeutische Verwendung der Zubereitung nach einem der vorstehenden Ansprüche als desodorierendes Mittel.
