These aromatic groups are preferably monocyclic (e.g., furan or benzene), bicyclic, or tricyclic groups containing from 5 to 14 atoms in the ring portion.

- aryl or “Ar” as used herein alone or as part of another group denote optionally substituted homocyclic aromatic groups, preferably monocyclic or bicyclic groups containing from 6 to 12 carbons in the ring portion, such as phenyl, biphenyl, naphthyl, substituted phenyl, substituted biphenyl, or substituted naphthyl.

- heteroatom refers to atoms other than carbon and hydrogen.
- heteroaromatic as used herein alone or as part of another group denotes optionally substituted aromatic groups having at least one heteroatom in at least one ring, and having in the range of 2 up to 12 carbon atoms, or preferably 5 or 6 atoms in each ring.
- the heteroaromatic group preferably has 1 or 2 oxygen atoms and/or 1 to 4 nitrogen atoms in the ring, and is bonded to the remainder of the molecule through a carbon.

- the heterocycio group preferably has 1 or 2 oxygen atoms and/or 1 to 4 nitrogen atoms in the ring, and is bonded to the remainder of the molecule through a carbon or heteroatom.
- Exemplary heterocycio groups include heteroaromatics as described above.

- hydrocarbon and “hydrocarbyl” as used herein describe organic compounds or radicals consisting exclusively of the elements carbon and hydrogen. These moieties include alkyl, alkenyl, alkynyl, and aryl moieties. These moieties also include alkyl, alkenyl, alkynyl, and aryl moieties substituted with other aliphatic or cyclic hydrocarbon groups, such as alkaryl, alkenaryl and alkynaryl. Unless otherwise indicated, these moieties preferably comprise 1 to 20 carbon atoms.

- the five compounds selected for the full dose response studies have very effective GI 5 o and TGI (Total Growth Inhibition) values against a variety of human tumor cell lines. All the compounds have LD 50 values >100 ⁇ against most of the human cancer cell lines implying the compounds are anti-proliferative.
- Example 5 had impressive GI 5 o values of less than 10nM against almost all the cell lines except melanoma cancer cell line UACC-62, colon cancer cell lines COLO 205 and HCC-2998.
- This compound also showed potent TGI values of ⁇ 10nM against CNS cancer cell line SF-539, melanoma cell MDA-MB-435, ovarian cancer cell line OVCAR-3 and renal cancer cell line A498.
- Example 4 and Example 6 have ⁇ 10nM GI 5 o values against Leukemia cancer cell lines K-562, SR, non-small lung cancer cell line HOP-92, colon cancer cell lines HCT-1 16, HCT-15, melanoma cancer lines M14, UACC-62, ovarian cancer cell line NCI/ADR-RES and renal cancer cell line A498.
- Melanoma cancer cell line MDA-MB-435 appeared to be sensitive to the growth inhibitory effects of Example 4; and Example 5 exhibited a TGI value of ⁇ 10 nM against ovarian cancer cell line OVCAR-3.

- Calcein-AM for 30 minutes at 25°C. Fluorescence was measured using a 490 nm excitation filter and a 520 nm emission filter. The fluorescence intensity is proportional to the number of viable cells. The fluorescence intensity of control (untreated) cells was taken as 100% viability. The relative cell viability compared to control was calculated by using the following formula:
