Abstract
Die Erfindung betrifft ein Verfahren und eine Reagenzzusammenstellung zur Anfärbung und damit Sichtbarmachung von Aminosäuren, Peptiden und ähnlichen Verbindungen, insbesondere nach Auftrennung mittels Dünnschichtchromatographie. Die Anfärbung erfolgt unter Verwendung von NINHYDRIN zum Nachweis von Aminosäuren, Peptiden oder Proteinen in Kombination mit mindestens einer ionischen Flüssigkeit.
Description
Verfahren und Reagenzzusammenstellung zur Sichtbarmachung von
Aminosäuren und Peptiden
Die Erfindung betrifft ein Verfahren und eine Reagenzzusammenstellung zur Anfärbung und damit Sichtbarmachung von Aminosäuren, Peptiden und ähnlichen Verbindungen insbesondere nach Auftrennung mittels Dünnschichtchromatographie. Die Anfärbung erfolgt unter Verwendung von mindestens einem Reagenz zum Nachweis von Aminosäuren, Peptiden in Kombination mit mindestens einer ionischen Flüssigkeit.
Die Analytik von Proteinen spielt eine wichtige Rolle bei der Untersuchung von Zellstrukturen oder Zellstoffwechseln bzw. Zellfunktionen. Die Analytik von Proteinen oder Peptiden umfasst dabei z.B. die Strukturanalyse, Bestimmung des Molekulargewichts, Bestimmung eines Verteilungsmusters (Peptide Mapping) oder auch die Identifikation bestimmter Peptide oder Proteine.
In der Regel muss vor der Analyse der Proteine oder Peptide zunächst eine Auftrennung durchgeführt werden. Bekannte Methoden hierzu sind beispielsweise die Flüssigkeitschromatographie, die lonenaustausch- chromatographie, die Kapillarelektrophorese oder die Gelelektrophorese. Anschließend erfolgt die Analyse z.B. mittels Anfärben oder mittels Massenspektrometrie.
Auch die Dünnschichtchromatographie ist zur Auftrennung von Proteinen, Peptiden oder Aminosäuren geeignet. Ein großer Vorteil der Dünnschichtchromatographie besteht darin, dass sie kostengünstig und einfach in der Durchführung ist. Weiterhin kann die Auftrennung ein- oder mehrdimensional erfolgen, so dass die Auflösung der Trennung an das Trennproblem angepasst werden kann.
Für Aminogruppen-haltige Verbindungen wie Proteine, Peptide oder Aminosäuren stehen zur Auswertung einer dünnschichtchromatographischen Trennung (DC-Trennung) verschiedene Färbemethoden zur Verfügung. Die bekanntesten sind die Anfärbung mit Ninhydrin (2,2-Dihydroxyindan-1 ,3-dion), mit Fluorescamin oder lsatin-5-
Sulfonsäure. Fluorescamin bietet im Vergleich zu Ninhydrin eine höhere Empfindlichkeit. Sowohl Ninhydrin als auch Fluorescamin geben jedoch keine zusätzliche Information über die angefärbte Verbindung, da alle Verbindungen in der gleichen Farbe angefärbt werden. lsatin-5-Sulfonsäure färbt Aminosäuren selektiv an, ist aber für Verbindungen, die aus mehreren Aminosäuren bestehen nicht geeignet, da die Empfindlichkeit sehr stark mit zunehmender Anzahl der Aminosäuren im zu untersuchenden Molekül abnimmt.
Aufgabe der vorliegenden Erfindung ist es daher, ein Verfahren bzw. ein
Reagenz zur selektiven Sichtbarmachung, insbesondere zur Anfärbung von Aminosäuren, Peptiden und Proteinen zur Verfügung zu stellen, das insbesondere auch nach einer DC-Trennung eingesetzt werden kann.
Es wurde gefunden, dass ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, insbesondere Ninhydrin oder eines seiner Derivate, ganz besonders bevorzugt Ninhydrin, in Kombination mit mindestens einer ionischen Flüssigkeit Aminosäuren und Verbindungen, die aus mehreren Aminosäuren bestehen, selektiv anfärbt. Man erhält Färbungen in einem breiten Farbspektrum zwischen gelb, grün, blau bis hin zu rot bis rosa. Die Empfindlichkeit der Anfärbung entspricht dabei zumindest der einer herkömmlichen Anfärbung von Aminosäuren mit beispielsweise Ninhydrin.
Gegenstand der vorliegenden Erfindung ist daher eine zumindest enthaltend mindestens ein Reagenz zum Nachweis von Aminosäuren,
Peptiden oder Proteinen, insbesondere Ninhydrin oder eines seiner Derivate, und eine ionische Flüssigkeit.
In der einfachsten Ausführungsform der vorliegenden Erfindung besteht die Reagenzzusammenstellung nur aus dem Reagenz zum Nachweis von
Aminosäuren, Peptiden oder Proteinen, insbesondere Ninhydrin oder eines seiner Derivate, und einer ionischen Flüssigkeit. In diesem liegt beispielsweise das Ninhydrin in der ionischen Flüssigkeit gelöst vor, so dass die ionische Flüssigkeit zusätzlich als Lösungsmittel fungiert.
In einer bevorzugten Ausführungsform enthält die Reagenzzusammenstellung zumindest a) ein Reagenz zumindest enthaltend mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen in mindestens einem Lösungsmittel und b) ein Reagenz zumindest enthaltend mindestens eine ionische Flüssigkeit.
In einer bevorzugten Ausführungsform enthält die Reagenzzusammenstellung zumindest a) ein Reagenz zumindest enthaltend 0.01-10 Massen-%, vorzugsweise 0.1-1 Massen-% Ninhydrin in mindestens einem Lösungsmittel b) ein Reagenz zumindest enthaltend mindestens eine ionische Flüssigkeit, vorzugsweise in mindestens einem Lösungsmittel.
Ionische Flüssigkeiten oder flüssige Salze sind ionische Spezies, die aus einem organischen Kation (K+) und einem in der Regel anorganischen Anion (A") bestehen. Sie enthalten keine neutralen Moleküle und weisen meistens Schmelzpunkte kleiner 373 K auf.
Das Gebiet der ionischen Flüssigkeiten wird zurzeit intensiv erforscht, da die Anwendungsmöglichkeiten vielfältig sind. Übersichtsartikel zu ionischen

Wesentlich für das erfindungsgemäße Verfahren zur Sichtbarmachung sind die Ionischen Flüssigkeiten der allgemeinen Formel K+A". Vorzugsweise ist das Anion A" ausgewählt aus der Gruppe umfassend Halogenide, insbesondere Chlorid und Bromid, Tetrafluoroborat, Sulfonate, insbesondere Trifluormethylsulfonat.
In Bezug auf die Wahl des Kations K+ der Ionischen Flüssigkeit der ionischen Flüssigkeit gibt es per se keine Einschränkungen. Vorzugsweise handelt es sich aber um organische Kationen, wobei es sich insbesondere bevorzugt um Ammonium-, Phosphonium, Thiouronium-,
Guanidiniumkationen oder um heterocyclische Kationen handelt.
Ammoniumkationen können beispielsweise durch die Formel (1 )

Ausgeschlossen sind jedoch Kationen der Formeln (1 ) und (2), in denen alle vier oder drei Substituenten R und R2 vollständig mit Halogenen substituiert sind, beispielsweise das Tris(trifluormethyl)methylammonium- kation, das Tetra(trifluormethyl)ammoniumkation oder das Tetra(nonafluor- butyl)ammoniumkation.
Geeignete Thiouroniumkationen können durch die Formel (3),

Ringsystem bilden können, wobei ein oder mehrere Substituenten R1' bis R4' teilweise oder vollständig mit Halogenen, insbesondere -F und/oder -Cl, oder -OR', -CN, -C(O)OH, -
C(O)NR'2, -SO2NR'2, -C(O)X, -SO2OH, -SO2X, -NO2, substituiert sein können, wobei jedoch nicht gleichzeitig R1 und R4 vollständig mit
Halogenen substituiert sein dürfen, und wobei ein oder zwei nicht benachbarte und nicht am Heteroatom gebundene Kohlenstoffatome der
Substituenten R1' bis R4', durch Atome und/oder Atomgruppierungen ausgewählt aus der -O-, -S-, -S(O)-, -SO2-, -N+R'2-, -C(O)NR1-, -SO2NR'-, oder -P(O)R'- ersetzt sein können mit R' = H, nicht, teilweise oder perfluoriertes Cr bis C6-Alkyl, C3- bis C7-Cycloalkyl, unsubstituiertes oder substituiertes Phenyl und X =Halogen.
Als Substituent R2 eignen sich insbesondere auch Atomgruppierungen ausgewählt aus -OR',-NR'2, -C(O)OH, -C(O)NR'2, -SO2NR'2, -SO2OH, - SO2X oder -NO2.
Unter vollständig ungesättigten Substituenten werden im Sinne der vorliegenden Erfindung auch aromatische Substituenten verstanden.
Als Substituenten R und R2 bis R13 der Verbindungen der Formeln (1 ) bis (4) kommen erfindungsgemäß dabei neben Wasserstoff bevorzugt in
Frage: d- bis C2o-, insbesondere d- bis Cu-Alkylgruppen, und gesättigte oder ungesättigte, d.h. auch aromatische, C3- bis C7-Cycloalkylgruppen, die mit Cr bis C6-Alkylgruppen substituiert sein können, insbesondere Phenyl.
Die Substituenten R und R2 in den Verbindungen der Formel (1 ) oder (2) können dabei gleich oder verschieden sein. Bevorzugt sind die Substituenten R und R2 verschieden.
Die Substituenten R und R2 sind insbesondere bevorzugt Methyl, Ethyl, Isopropyl, Propyl, Butyl, sek.-Butyl, Pentyl, Hexyl, Octyl, Decyl oder Tetradecyl.
Bis zu vier Substituenten des Guanidinium-Kations [C(NR8R9)(NR10R11)(NR12R13)]+ können auch paarweise derart verbunden sein, dass mono-, bi- oder polycyclische Kationen entstehen.
Ohne Einschränkung der Allgemeinheit sind Beispiele für solche Guanidinium-Kationen:
wobei die Substituenten R8 bis R10 und R13 eine zuvor angegebene oder besonders bevorzugte Bedeutung haben können. Gegebenenfalls können die Carbocyclen oder Heterocyclen der zuvor angegebenen Guanidinium-Kationen noch durch Cr bis Cβ-Alkyl, Cr bis Ce-Alkenyl, NO2, CN, NR'2, F, Cl, Br, I, C1-C6-AIkOXy, SCF3, SO2CF3, COOH, SO2NR'2> SO2X' oder SO3H substituiert sein, wobei X und R' eine zuvor angegebene Bedeutung haben, substituiertes oder unsubstituiertes Phenyl oder unsubstituierter oder substituierter Heterocyclus substituiert sein.
Bis zu vier Substituenten des Thiouroniumkations [(R3R4N)- C(=SR5)(NR6R7)]+ können auch paarweise derart verbunden sein, dass mono-, bi- oder polycyclische Kationen entstehen.
Ohne Einschränkung der Allgemeinheit sind Beispiele für solche Kationen im Folgenden angegeben, wobei Y = S bedeutet:
wobei die Substituenten R3, R5 und R6 eine zuvor angegebene oder besonders bevorzugte Bedeutung haben können. Die Substituenten R3 bis R13 sind jeweils unabhängig voneinander bevorzugt eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 10 C- Atomen. Die Substituenten R3 und R4, R6 und R7, R8 und R9, R10 und R11 und R12 und R13 in Verbindungen der Formeln (3) bis (4) können dabei gleich oder verschieden sein. Besonders bevorzugt sind R3 bis R13 jeweils unabhängig voneinander Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, tert.-
Butyl, sek.-Butyl, Phenyl oder Cyclohexyl, ganz besonders bevorzugt Methyl, Ethyl, n-Propyl, Isopropyl oder n-Butyl.
Als Substituenten R1 bis R4 von Verbindungen der Formel (5) kommen erfindungsgemäß dabei neben Wasserstoff bevorzugt in Frage: d- bis C2o, insbesondere C1- bis Ci2-Alkylgruppen, und gesättigte oder ungesättigte, d.h. auch aromatische, C3- bis C7-Cycloalkylgruppen, die mit d- bis C6- Alkylgruppen substituiert sein können, insbesondere Phenyl.
Die Substituenten R1 und R4 sind jeweils unabhängig voneinander insbesondere bevorzugt Methyl, Ethyl, Isopropyl, Propyl, Butyl, sek.-Butyl, Pentyl, Hexyl, Octyl, Decyl, Cyclohexyl, Phenyl oder Benzyl. Sie sind ganz besonders bevorzugt Methyl, Ethyl, n-Butyl oder Hexyl. In Pyrrolidinium-, Piperidinium- oder Indolinium-Verbindungen sind die beiden Substituenten R1' und R4 bevorzugt unterschiedlich.
Der Substituent R2 oder R3 ist jeweils unabhängig voneinander insbesondere Wasserstoff, Methyl, Ethyl, Isopropyl, Propyl, Butyl, sek.- Butyl, tert.-Butyl, Cyclohexyl, Phenyl oder Benzyl. Besonders bevorzugt ist R2' Wasserstoff, Methyl, Ethyl, Isopropyl, Propyl, Butyl oder sek.-Butyl. Ganz besonders bevorzugt sind R2 und R3 Wasserstoff. Ein geradkettiges oder verzweigtes Alkenyl mit 2 bis 20 C-Atomen, wobei auch mehrere Doppelbindungen vorhanden sein können, ist beispielsweise AIIyI, 2- oder 3-Butenyl, Isobutenyl, sek.-Butenyl, ferner 4-Pentenyl, iso- Pentenyl, Hexenyl, Heptenyl, Octenyl, -C9Hi7, -Ci0H19 bis -C2oH39;

HetN+ ist bevorzugt

Vorzugsweise handelt es sich bei den Kationen der erfindungsgemäßen ionischen Flüssigkeit um Ammonium-, Imidazolium-, Pyridinium-, Pyrrolidinium-, Piperidinium-, Phosphonium-, Morpholinium- oder Piperidinium-Kationen.
Besonders bevorzugte Ionische Flüssigkeiten sind 1-Pentyl-3-methyl- imidazoliumchlorid, 1 -Hexyl-3-methyl-imidazoliumchlorid, 1 -Heptyl-3- methyl-imidazoliumchlorid, 1-Octyl-3-methyl-imidazoliumchlorid, 1-Nonyl-3- methyl-imidazoliumchlorid, 1-Decyl-3-methyl-imidazoliumchlorid, Trihexyl- tetradecyl-phosphoniumchlorid, Ethyldimetylpentylammoniumbromid, Ethyldimethylpropylammoniumchlorid, 1-Methyl-1-octylpyrrolidiniumchlorid, n-Butylpyridiniumchlorid, n-Octylpyridiniumchlorid, 4-Methyl-4- pentylmorpholiniumbromid oder 1-Methyl-1-pentylpiperidiniumbromid.
Ganz besonders bevorzugte Ionische Flüssigkeiten sind 1-Pentyl-3-methyl- imidazoliumchlorid, i-Hexyl-3-methyl-imidazoliumchlorid, 1-Heptyl-3- methyl-imidazoliumchlorid, i-Octyl-3-methyl-imidazoliumchlorid, 1-Nonyl-3- methyl-imidazoliumchlorid, 1 -Decyl-3-methyl imidazoliumchlorid, Ethyldimetylpentylammoniumbromid und 1-Methyl-1- octylpyrrolidiniumchlorid.
In einer anderen bevorzugten Ausführungsform enthält die Reagenzzusammenstellung zusätzlich zumindest eine DC-Platte.
Gegenstand der vorliegenden Erfindung ist weiterhin ein Verfahren zur Analyse einer Probe mit Aminosäuren, Peptiden und/oder Proteinen durch a) chromatographische Auftrennung der Probe auf einer dünnschichtchromatographischen Platte (DC-Platte) b) Anfärben der aufgetrennten Probe auf der DC-Platte mit einer
Reagenzzusammenstellung zumindest enthaltend mindestens ein
Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen und mindestens eine ionische Flüssigkeit.
Ein weiterer Gegenstand der vorliegenden Erfindung betrifft ein Verfahren zur Analyse einer Probe mit Aminosäuren, Peptiden und/oder Proteinen durch a') chromatographische Auftrennung der Probe auf einer dünnschichtchromatographischen Platte (DC-Platte), wobei das Fließmittel mindestens eine ionische Flüssigkeit enthält, und b') Anfärben der aufgetrennten Probe auf der DC-Platte mit einer Reagenzzusammenstellung zumindest enthaltend mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen.
Ebenfalls Gegenstand der vorliegenden Erfindung ist ein Verfahren zur
Analyse einer Probe mit Aminosäuren, Peptiden und/oder Proteinen durch chromatographische Auftrennung der Probe auf einer
dünnschichtchromatographischen Platte (DC-Platte), wobei das Fließmittel mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin oder eines seiner Derivate, und mindestens eine ionische Flüssigkeit enthält.
In einer bevorzugten Ausführungsform wird als DC-Platte in Schritt a) oder a') eine Platte mit einer unpolaren Phase oder einer Cellulose-Phase eingesetzt.
In einer anderen bevorzugten Ausführungsform erfolgt die
Sichtbarmachung in Schritt b), indem die DC-Platte in einem Schritt b1 ) zunächst mit einem Reagenz behandelt wird, das zumindest eine ionische Flüssigkeit enthält und anschließend in einem Schritt b2) mit einem Reagenz, das zumindest mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin oder eines seiner Derivate, enthält. Es ist auch möglich die Sichtbarmachung in Schritt b) durchzuführen, indem zunächst eine Behandlung mit mindestens einem Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin (b2), und dann mit der ionischen Flüssigkeit (b1 ) erfolgt. Vorzugsweise jedoch erfolgt die Sichtbarmachung in Schritt b), indem die DC-Platte in einem Schritt b1 ) zunächst mit einem Reagenz behandelt wird, das zumindest eine ionische Flüssigkeit enthält und anschließend in einem Schritt b2) mit einem Reagenz, das zumindest Ninhydrin oder eines seiner Derivate enthält, behandelt wird.
In einer bevorzugten Ausführungsform wird in Schritt b) nach der Behandlung der DC-Platte mit der Reagenzzusammenstellung zumindest enthaltend mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin, und mindestens eine ionische Flüssigkeit oder nach Schritt b') für 0.5 bis 10 Minuten auf eine Temperatur zwischen 50 und 2000C erhitzt.
In einer bevorzugten Ausführungsform wird eine Reagenzzusammenstellung verwendet, die ein Reagenz enthält, das zumindest 0.01-10 Massen-%, vorzugsweise 0.1-1 Massen-% Ninhydrin in mindestens einem Lösungsmittel enthält, und ein Reagenz, das zumindest 0.1 bis 10% mindestens einer ionischen Flüssigkeit in mindestens einem Lösungsmittel enthält.
Gegenstand der vorliegenden Erfindung ist zudem ein Verfahren zur Sichtbarmachung, insbesondere zur Anfärbung von Aminosäuren, Peptiden und/oder Proteinen, dadurch gekennzeichnet, dass die Aminosäuren, Peptide und/oder Proteine mit einer Reagenzkombination behandelt werden, die zumindest eine ionische Flüssigkeit und mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin oder eines seiner Derivate, enthält.
In einer bevorzugten Ausführungsform werden die Aminosäuren, Peptide und/oder Proteine zunächst mit einem Reagenz behandelt, das zumindest eine ionische Flüssigkeit enthält und dann mit einem Reagenz, das zumindest Ninhydrin enthält.
Weitere Kombinationen bzw. bevorzugte Ausführungsformen sind in den Patentansprüchen offenbart.
Eine Probe mit Aminosäuren, Peptiden und/oder Proteinen ist erfindungsgemäß eine Probe, von der angenommen wird, dass sie zumindest eine Aminosäure und/oder ein Peptid enthält. Die Probe kann natürlichen oder synthetischen Ursprungs sein, z.B. ein Zellextrakt, eine Körperflüssigkeit, ein Proteinverdau oder das Reaktionsgemisch einer Peptidsynthese. In der Regel handelt es sich um eine flüssige Probe, wobei die Analyten typischerweise in einem organischen Lösungsmittel oder
Wasser oder Gemischen organischer Lösungsmittel oder Mischungen von organischen Lösungsmitteln und Wasser vorliegen. Die Probe kann
beliebige weitere feste, emulgierte oder gelöste Bestandteile enthalten, die aber weder die DC-Auftrennung noch die spätere Anfärbung der Analyten (Aminosäuren und/oder Peptide) stören sollten. Feste Proben werden in der Regel zunächst in einem der oben genannten Lösungsmittel aufgenommen, damit sie auf die DC-Platte aufgetragen werden können. Bei konzentrierten Proben kann es notwendig sein, diese zunächst zu verdünnen. Dem Fachmann im Bereich der Dünnschichtchromatographie ist bekannt, wie groß die Probenmenge und Probenkonzentration in Abhängigkeit von der eingesetzten DC-Platte und dem jeweiligen Trennproblem sein darf bzw. sein muss, um möglichst ideal auswertbare Banden zu erhalten.
Aminosäuren, Peptide und/oder Proteinen sind erfindungsgemäß alle natürlichen oder synthetischen Aminosäuren, Verbindungen, die zwei oder mehrere natürliche oder synthetische Aminosäuren enthalten sowie Aminogruppen-haltige Verbindungen, die beispielsweise mit einer herkömmlichen Ninhydrin-Färbung angefärbt werden können. Verbindungen, die zwei oder mehrere natürliche oder synthetische Aminosäuren enthalten, sind bevorzugt Oligoaminosäuren oder Peptide, insbesondere Peptide mit einer Kettenlänge bis zu 150 Aminosäuren, bevorzugt mit einer Kettenlänge zwischen 2 und 100, besonders bevorzugt zwischen 2 und 25 Aminosäuren. Beispiele für Aminogruppen-haltige Verbindungen, die mit einer herkömmlichen Ninhydrin-Färbung angefärbt werden können, sind Aminosäuren, Amine oder Aminozucker. Bei den Aminosäuren kann es sich um synthetische oder natürliche Aminosäuren handeln. Vorzugsweise handelt es sich um natürliche Aminosäuren wie AIa, Arg, Asn, Asp, Cys, GIn, GIu, GIy, His, He, Leu, Lys, Met, Phe, Pro, Ser, Thr, Typ, Tyr oder VaI.
Möchte man mit dem erfindungsgemäßen Verfahren Proteine oder Peptide mit einer Kettenlänge von mehr als 150 Aminosäuren analysieren, so können diese in einer weiteren Ausführungsform zunächst in Peptide mit
einer Kettenlänge von unter 150 Aminosäuren gespalten werden - z.B. durch einen tryptischen Verdau.
Eine DC-Platte ist erfindungsgemäß jedes Medium, auf dem eine dünnschichtchromatographische Auftrennung durchgeführt werden kann. In der Regel besteht eine DC-Platte aus einem Träger z.B. in Form einer Glasplatte, einer Metallplatte oder -folie oder einer Kunststofffolie, die mit einer Sorbens-Phase belegt oder beschichtet ist. Als Sorbentien werden typischerweise die für chromatographische Zwecke bekannten Basis- Sorbentien verwendet. Dies sind z.B. Kieselgel, Aluminiumoxid, Cellulose, Kieselgur oder andere organische oder anorganische Polymere oder organisch/anorganische Hybridpolymere. Weiterhin können die Basis- Sorbentien mit funktionellen Gruppen derivatisiert sein, die ihre Trenneigenschaften verändern. Beispiele hierfür sind RP-Phasen, bei denen z.B. Kieselgel mit Liganden derivatisiert ist, die C8 oder C18 Ketten aufweisen (Reversed Phase Material). Andere Bespiele sind CN oder Diol- modifizierte Phasen. Einen Überblick über gängige Sorbens-Phasen für die DC findet man in Klaus K. Unger, Packings and Stationary Phases in Chromatographie Techniques, M. Decker, New York 1990. Entsprechend der Polarität ihrer Oberfläche kann man die Sorbens-Phasen in polare, mittelpolare und unpolare Phasen aufteilen. Es wurde gefunden, dass die erfindungsgemäße Reagenzzusammenstellung besonders geeignet ist zur Anfärbung von Aminosäuren und/oder Peptiden auf unpolaren Phasen, wie Kieselgur oder Reversed Phase Materialien. Auch Cellulose-Phasen lassen sich mit hervorragender Farb-Qualität und Intensität anfärben.
Eine erfindungsgemäße Reagenzzusammenstellung enthält zumindest mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin, und mindestens eine ionische Flüssigkeit. Die Reagenzzusammenstellung kann in Form eines
Reagenzes, typischerweise eines flüssigen Reagenzes, vorliegen, oder mehrere unterschiedliche Reagenzien bzw. Komponenten enthalten. Dabei
können das mindestens eine Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin, und zumindest eine ionische Flüssigkeit zusammen in einem Reagenz vorliegen oder in zwei unterschiedlichen Reagenzien bzw. Komponenten der Reagenzzusammenstellung.
In einer Ausführungsform besteht die Reagenzzusammenstellung zumindest aus einem typischerweise flüssigen Reagenz, das zumindest mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin, und mindestens eine ionische Flüssigkeit enthält.
In einer bevorzugten Ausführungsform besteht die Reagenzzusammenstellung zumindest aus a) einem Reagenz, das zumindest Ninhydrin oder eines seiner Derivate enthält b) einem Reagenz, das zumindest eine ionische Flüssigkeit enthält. Beide Reagenzien sind typischerweise flüssig.
Als Lösungsmittel können ein oder mehrere organische Lösungsmittel oder Mischungen von einem oder mehreren organischen Lösungsmitteln mit Wasser oder Wasser eingesetzt werden. Es ist auch möglich, dass die ionische Flüssigkeit selbst als Lösungsmittel dient und beispielsweise gemäß der oben beschriebenen Ausführungsform als Lösungsmittel für das Reagenz zum Nachweisen von Aminosäuren, Peptiden und/oder Proteinen fungiert. Es können mehrere LM sein, bevorzugt wird nur ein LM verwendet. Für in organischen LM schwer lösliche ILs kann Wasser oder eine Mischung aus Wasser + Alkohol sinnvoll sein.
Bei einem Reagenz, das beispielsweise Ninhydrin und zumindest eine ionische Flüssigkeit enthält, dient als Lösungsmittel bevorzugt Aceton oder Propanol. Bei einem Reagenz, das Ninhydrin aber keine ionische
Flüssigkeit enthält, dient bevorzugt ein organisches Lösungsmittel wie Propanol oder Aceton als Lösungsmittel. Bei einem Reagenz, das mindestens eine ionische Flüssigkeit aber kein Ninhydrin enthält, dient als Lösungsmittel bevorzugt Aceton, Propanol, Wasser und Wasser/Alkohol- Mischungen.
Die Reagenzien können zudem optional weitere Stoffe enthalten, z.B. eine oder mehrere zusätzliche ionische Flüssigkeiten, Lösungsmittel, Stabilisatoren, Puffer, weitere Farbstoffe etc.
Ninhydrin liegt in den erfindungsgemäßen Reagenzien typischerweise in Konzentrationen zwischen 0.01 und 10, bevorzugt in Konzentrationen zwischen 0.1 und 1 Massen-% vor.
Die ionische Flüssigkeit kann, je nach Art der Reagenzienzusammenstellung, in Konzentrationen zwischen 0.1 und 100 Massen-% vorliegen. Dient die ionische Flüssigkeit zugleich als Lösungsmittel, bzw. umfasst die Reagenzienzusammenstellung getrennte Reagenzien von beispielsweise Ninhydrin und ionischer Flüssigkeit, so liegt der Anteil der ionischen Flüssigkeit typischerweise zwischen 80 und 100%. Es versteht sich von selbst, dass im Falle der Ausführungsform, bei der beispielsweise Ninhydrin und die ionische Flüssigkeit zusammen vorliegen, der Anteil der ionischen Flüssigkeit nicht 100% betragen kann, sondern entsprechend um den Anteil an Ninhydrin verringert werden muss. Wird zusätzlich ein anderes Lösungsmittel verwendet, liegt die ionische Flüssigkeit typischerweise in Konzentrationen zwischen 0.1 und 15, bevorzugt in Konzentrationen zwischen 0.1 und 10, besonders bevorzugt in Konzentrationen zwischen 1 und 5 Massen-% vor. Enthält ein Reagenz zwei oder mehrere verschiedene ionische Flüssigkeiten, dann gelten die obigen Konzentrationsangaben für die Gesamtmenge an ionischer Flüssigkeit.
Weitere optionale Bestandteile der erfindungsgemäßen Reagenzkombination sind beispielsweise ein oder mehrere weitere Reagenzien, die zumindest mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen, vorzugsweise Ninhydrin, und/oder eine oder mehrere ionische Flüssigkeit enthalten (z.B. Reagenzien, die diese Bestandteile in anderen Konzentrationen enthalten oder Reagenzien, die andere ionische Flüssigkeit enthalten), eine oder mehrere DC-Platten, alternative Färbereagenzien (z.B. Reagenzien zur Fluorescamin-Färbung) oder Farbtabellen bzw. Farbmuster zur Auswertung der Färbeergebnisse.
Beispiele für erfindungsgemäße Reagenzkombinationen sind:
1. Eine Reagenzkombination bestehend aus einem Reagenz, das zumindest zwischen 0.01 und 10% Ninhydrin und zwischen 0.1 und 10% einer oder mehrerer ionischer Flüssigkeiten in einem Lösungsmittel enthält.
2. Eine Reagenzkombination enthaltend a) ein Reagenz das zumindest zwischen 0.01 und 10% Ninhydrin in einem
Lösungsmittel enthält b) ein Reagenz, das zumindest zwischen 0.1 und 10% einer oder mehrerer ionischer Flüssigkeiten in einem Lösungsmittel enthält
3. Eine Reagenzkombination entsprechend Variante 2, die zusätzlich ein weiteres Reagenz enthält, das zumindest eine oder mehrere ionische Flüssigkeiten in einem Lösungsmittel enthält, wobei sich die Art,
Konzentration und/oder Zusammensetzung der ionischen Flüssigkeiten dieses zusätzlichen Reagenzes von der des anderen Reagenzes, das zumindest eine oder mehrere ionische Flüssigkeiten in einem Lösungsmittel enthält, unterscheidet.
4. Eine Reagenzkombination entsprechend Variante 1 , 2 oder 3, zusätzlich enthaltend eine oder mehrere DC-Platten und/oder alternative
Färbereagenzien (z.B. Reagenzien zur Fluorescamin-Färbung) und/oder Farbtabellen bzw. Farbmuster zur Auswertung der Färbeergebnisse.
Bevorzugte Reagenzkombinationen enthalten zwei oder mehrere Reagenzien, wobei Ninhydrin einerseits und ein oder mehrere ionische
Flüssigkeiten andererseits getrennt in unterschiedlichen Reagenzien vorliegen.
Bei dem erfindungsgemäßen Verfahren zur Analyse einer Probe mit Aminosäuren, Peptiden und/oder Proteinen wird die Probe zunächst auf eine DC-Platte aufgetragen und dünnschichtchromatographisch aufgetrennt. Die Durchführung einer dünnschichtchromatographischen Trennung ist dem Fachmann bekannt, z.B. aus 0b1 s12v1 P Dünnschichtchromatographie, Praktische Durchführung und Fehlervermeidung von Elke Hahn-Deinstrop, Wiley-VCH (1998). Die Auftrennung kann eindimensional oder mehrdimensional durchgeführt werden.
Nach erfolgter dünnschichtchromatographischer Auftrennung erfolgt die Anfärbung mit einer Reagenzzusammenstellung zumindest enthaltend
Ninhydrin und mindestens eine ionische Flüssigkeit zur Sichtbarmachung der in der Probe enthaltenen Aminosäuren, Peptide und/oder Proteine.
Dazu wird die DC-Platte bevorzugt zunächst leicht, vorzugsweise vollständig, getrocknet, damit das Fließmittel, das bei der DC-T rennung verwendet wurde, ganz oder teilweise entfernt wird. Die erfindungsgemäße
Anfärbung kann jedoch auch auf noch vom Fließmittel angefeuchteten DC-
Platten durchgeführt werden.
Der Auftrag der erfindungsgemäßen Reagenzkombination, d.h. in der Regel das Benetzen der DC-Platte mit den Reagenzien der
Reagenzzusammenstellung, erfolgt typischerweise durch Eintauchen der
DC-Platte in die entsprechenden flüssigen Reagenzien oder bevorzugt durch Aufsprühen der Reagenzien auf die DC-Platte.
In einer Ausführungsform wird die DC-Platte mit einem Reagenz benetzt, das zumindest Ninhydrin und eine oder mehrere ionische Flüssigkeiten in einem Lösungsmittel enthält. Dieses Reagenz kann bereits längere Zeit vor seiner Verwendung zusammengestellt worden sein oder bevorzugt maximal 24 Stunden vor seiner Verwendung zusammengemischt worden sein.
In einer bevorzugten Ausführungsform wird die DC-Platte zunächst mit einem Reagenz benetzt, das zumindest eine ionische Flüssigkeit in mindestens einem Lösungsmittel enthält. Anschließend, bevorzugt nach ganz oder teilweisem Trocknen der DC-Platte, wird die Platte mit einem Reagenz benetzt, das zumindest Ninhydrin enthält.
Je nach Art der anzufärbenden Aminosäuren, Peptide und/oder Proteine kann die Farbreaktion spontan oder nach mehrstündiger Inkubationszeit auftreten oder, wie in den häufigsten Fällen, erst nach Erhitzen. Aus diesem Grund wird die DC-Platte bevorzugt nach dem Auftrag der Reagenzien der erfindungsgemäßen Reagenzkombination auf Temperaturen über 35°C, bevorzugt über 50°C erhitzt. In einer besonders bevorzugten Ausführungsform wird auf Temperaturen zwischen 50 und 2000C erhitzt. Die Dauer der Temperaturbehandlung beträgt in der Regel zwischen 0,5 und 10 Minuten.
Die entstehende Färbung kann visuell oder mit entsprechenden optischen Geräten ausgewertet werden.
Die erfindungsgemäße Reagenzkombination kann auch zum Anfärben von Aminosäuren, Peptiden und/oder Proteinen in anderen Medien verwendet werden, beispielsweise in flüssiger Phase oder auf anderen Trägern, wie
Papier, Gelen oder Membranen. Genauso kann die Reagenzkombination für einen Schnelltest eingesetzt werden, in dem zunächst nur generell festgestellt wird, ob Aminosäuren, Peptide und/oder Proteine in einer Probe vorhanden sind. Dazu muss keine chromatographische oder andere Trennung durchgeführt werden. Es ist ausreichend, die Probe - bevorzugt auf einem Träger, z.B. einer DC-Platte - mit der Reagenzkombination in Kontakt zu bringen. Die Durchführung erfolgt dabei entsprechend Schritt b) des oben beschriebenen Verfahrens. Anhand der Farbentwicklung lässt sich dann feststellen ob und gegebenenfalls welche Aminosäuren, Peptide und/oder Proteine in der Probe vorhanden sind.
Der besondere Vorteil der erfindungsgemäßen Reagenzzusammenstellung beziehungsweise des erfindungsgemäßen Verfahrens ist, dass bei verschiedenen Aminosäuren, Peptiden und/oder Proteinen eine unterschiedliche Färbung entsteht. Während bei der herkömmlichen Ninhydrin-Färbung, bei der keine ionischen Flüssigkeiten zugesetzt werden, alle Aminosäuren und/oder Peptide außer Prolin eine blau bis violette Färbung zeigen, variiert die Farbe bei der erfindungsgemäßen Färbung über das gesamte Farbspektrum. Hinzu kommt, dass durch die Wahl der eingesetzten ionischen Flüssigkeit bzw. Mischung zweier oder mehrerer ionischer Flüssigkeiten das entstehende Farbspektrum verändert werden kann. So kann man, falls nötig, anhand weniger Versuche mit verschiedenen ionischen Flüssigkeiten herausfinden, welche ionische Flüssigkeit oder welche Mischung für das vorliegende Analyseproblem besonders vorteilhaft ist und somit die Zielanalyte besonders gut und unterscheidbar von anderen in der Probe vorhandenen Aminosäuren und/oder Peptiden anfärbt.
Sowohl Aminosäuren wie auch Peptide zeigen je nach ihrer chemischen Struktur bzw. ihrer Zusammensetzung nach Behandlung mit der erfindungsgemäßen Reagenzkombination verschiedene Färbungen. Dadurch wird sowohl die Identifikation bestimmter Aminosäuren in einer
Probe erheblich erleichtert, aber auch die Identifikation von Peptiden, z.B. für das Peptide Mapping nach einem tryptischen Verdau.
In den angefügten Tabellen 1 und 2 werden beispielhaft für erfindungsgemäße Reagenzkombinationen mit unterschiedlichen ionischen Flüssigkeiten die bei der Färbung einiger Aminosäuren entstehenden Farbmuster aufgelistet.

Üblicherweise kann die Sichtbarmachung bereits bei Mengen von 10 bis 50 ng an Aminosäure bzw. Peptid erfolgen.
Neben Ninhydrin können selbstverständlich auch entsprechende Derivate von Ninhydrin oder Komplexe des Ninhydrins, z.B. jene in Kombination mit z.B. Cu, Cd, Sn, eingesetzt werden.
Auch ohne weitere Ausführungen wird davon ausgegangen, daß ein Fachmann die obige Beschreibung im weitesten Umfang nutzen kann. Die bevorzugten Ausführungsformen und Beispiele sind deswegen lediglich als beschreibende, keineswegs als in irgendeiner Weise limitierende Offenbarung aufzufassen.
Die vollständige Offenbarung aller vor- und nachstehend aufgeführten Anmeldungen, Patente und Veröffentlichungen ist durch Bezugnahme in diese Anmeldung eingeführt.

Verfahrensbeschreibung:
Probenauftragung: Die Proben werden mit einer Kapillare oder mit einem automatischen DC Auftragegerät (z.B. Linomat V, Camag, Muttenz, Schweiz) auf die DC-Platte aufgetragen. Bei einer typischen Probekonzentration von 1-2 mg/ml werden 1-1 Oμl aufgetragen.
Entwicklung: Die Entwicklung erfolgt in einer DC-Kammer. Die Platte wird nach der Auftragung kurz getrocknet und dann in eine mit Fließmittel (Mobile Phase) befüllte DC- Kammer (ohne Kammersättigung) gestellt und entwickelt. Die Entwicklungsstrecke ist 5 cm und die Entwicklungszeit für 5 cm liegt bei ca. 1 h.
Trocknung: Die Platte wird für mindestens 30 min unter Druckluft getrocknet, um das Fließmittel möglichst vollständig zu entfernen.
Anfärbung: Die Anfärbung erfolgt durch Besprühen mit einem DC-
Sprühgerät. Dazu wird eine 3,5%ige IL Lösung und eine 0,5%ige Ninhydrin
Lösung vorbereitet. Die Platte wird erst mit der IL Lösung besprüht . Dann erfolgt eine Zwischentrocknung von 5 min. Dann wird die Platte mit Ninhydrin Lösung besprüht und gleich danach für zwei Minuten auf 1100C erhitzt. Die Platte wird kurz abgekühlt und dokumentiert.
Alternativ kann auch ein vollautomatisches Sprühgerät, z.B. das Gerät ChromaJet DS 20 von DESAGA SARSTEDT-Gruppe (Deutschland), eingesetzt werden.
Claims
1. Reagenzzusammenstellung zumindest enthaltend mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen und mindestens eine ionische Flüssigkeit.
2. Reagenzzusammenstellung nach Anspruch 1 , dadurch gekennzeichnet , dass das Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen Ninhydrin oder eines seiner Derivate enthält.
3. Reagenzzusammenstellung nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass die Reagenzzusammenstellung zumindest a) ein Reagenz zumindest enthaltend Ninhydrin oder eines seiner Derivate in mindestens einem Lösungsmittel und b) ein Reagenz zumindest enthaltend mindestens eine ionische Flüssigkeit enthält.
4. Reagenzzusammenstellung nach einem oder mehreren der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass die Reagenzzusammenstellung zumindest a) ein Reagenz zumindest enthaltend 0.01-10 Massen-% Ninhydrin in mindestens einem Lösungsmittel b) ein Reagenz zumindest enthaltend 0.1 bis 10 Massen-% mindestens einer ionischen Flüssigkeit in mindestens einem Lösungsmittel enthält.
5. Reagenzzusammenstellung nach einem oder mehreren der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die Reagenzzusammenstellung ein Reagenz enthält, das als ionische Flüssigkeit 1-Pentyl-3-methyl- imidazoliumchlorid, i-Hexyl-3-methyl-imidazoliumchloπd, i-Heptyl-3- methyl-imidazoliumchlorid, 1 -Octyl-3-methyl-imidazoliumchlorid, 1 -Nonyl-3- methyl-imidazoliumchlorid, 1-Decyl-3-methyl-imidazoliumchlorid, Trihexyl- tetradecyl-phosphoniumchlorid, Ethyldimetylpentylammoniumbromid, Ethyldimethylpropylammoniumchlorid, 1 -Methyl-1 -octylpyrrolidiniumchlorid, n-Butylpyridiniumchlorid, n-Octylpyridiniumchlorid, 4-Methyl-4- pentylmorpholiniumbromid oder 1 -Methyl-1 -pentylpiperidiniumbromid enthält.
6. Reagenzzusammenstellung nach einem oder mehreren der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass die Reagenzzusammenstellung zusätzlich zumindest eine DC-Platte enthält.
7. Verfahren zur Analyse einer Probe mit Aminosäuren, Peptiden und/oder Proteinen durch a) chromatographische Auftrennung der Probe auf einer dünnschichtchromatographischen Platte (DC-Platte) b) Anfärben der aufgetrennten Probe auf der DC-Platte mit einer
Reagenzzusammenstellung zumindest enthaltend mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen und mindestens eine ionische Flüssigkeit.
8. Verfahren zur Analyse einer Probe mit Aminosäuren, Peptiden und/oder Proteinen durch a') chromatographische Auftrennung der Probe auf einer dünnschichtchromatographischen Platte (DC-Platte), wobei das Fließmittel mindestens eine ionische Flüssigkeit enthält, und b') Anfärben der aufgetrennten Probe auf der DC-Platte mit einer
Reagenzzusammenstellung zumindest enthaltend mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen.
9. Verfahren zur Analyse einer Probe mit Aminosäuren, Peptiden und/oder Proteinen durch chromatographische Auftrennung der Probe auf einer dünnschichtchromatographischen Platte (DC-Platte), wobei das Fließmittel mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen und mindestens eine ionische Flüssigkeit enthält.
10. Verfahren nach einem oder mehreren der Ansprüche 7 bis 9, dadurch gekennzeichnet, dass das Reagenz zum Nachweis von Aminosäuren,
Peptiden oder Proteinen Ninhydrin oder eines seiner Derivate ist.
11. Verfahren nach einem oder mehreren der Ansprüche 7 bis 10, dadurch gekennzeichnet, dass als DC-Platte in Schritt a) oder a') eine Platte mit einer unpolaren Phase oder einer Cellulose-Phase eingesetzt wird.
12. Verfahren nach einem oder mehreren der Ansprüche 7 bis 11 , dadurch gekennzeichnet, dass die Anfärbung in Schritt b) erfolgt, indem die DC- Platte in einem Schritt b1 ) zunächst mit einem Reagenz behandelt wird, das zumindest eine ionische Flüssigkeit enthält und anschließend in einem Schritt b2) mit einem Reagenz, das zumindest mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen enthält.
13. Verfahren nach einem oder mehreren der Ansprüche 7 bis 12, dadurch gekennzeichnet, dass in Schritt b) nach der Behandlung der DC-Platte mit der Reagenzzusammenstellung zumindest enthaltend mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen und eine ionische Flüssigkeit für 0.5 bis 10 Minuten auf eine Temperatur zwischen 50 und 2000C erhitzt wird.

Verfahren nach einem oder mehreren der Ansprüche 7 bis 13, dadurch gekennzeichnet, dass eine Reagenzzusammenstellung verwendet wird, die ein Reagenz enthält, das zumindest 0.01-10 Massen-% Ninhydrin in mindestens einem Lösungsmittel enthält, und ein Reagenz, das zumindest 0.1 bis 10 Massen-% mindestens einer ionischen Flüssigkeit in mindestens einem Lösungsmittel enthält.

Verfahren zur Sichtbarmachung von Aminosäuren, Peptiden und/oder Proteinen, dadurch gekennzeichnet, dass die Aminosäuren, Peptide und/oder Proteine mit einer Reagenzkombination behandelt werden, die zumindest eine ionische Flüssigkeit und mindestens ein Reagenz zum Nachweis von Aminosäuren, Peptiden oder Proteinen enthält.
16. Verfahren nach Anspruch 15, dadurch gekennzeichnet, dass die Aminosäuren, Peptide und/oder Proteine zunächst mit einem Reagenz behandelt werden, das zumindest eine ionische Flüssigkeit enthält und anschließend mit einem Reagenz, das zumindest Ninhydrin oder eines seiner Derivate enthält.
17. Verfahren nach einem oder mehreren der Ansprüche 7 bis 16, dadurch gekennzeichnet, dass die ionische Flüssigkeit ausgewählt ist aus 1-Pentyl- 3-methyl-imidazoliumchlorid, 1-Hexyl-3-methyl-imidazoliumchlorid, 1-
Heptyl-3-methyl-imidazoliumchlorid, 1-Octyl-3-methyl-imidazoliumchlorid, 1- Nonyl-3-methyl-imidazoliumchlorid, 1-Decyl-3-methyl-imidazoliumchlorid, Trihexyl-tetradecyl-phosphoniumchlorid, Ethyldimetylpentylammoniumbromid, Ethyldimethylpropylammoniumchlorid, 1-Methyl-i-octylpyrrolidiniumchlorid, n-Butylpyridiniumchlorid, n-
Octylpyridiniumchlorid, 4-Methyl-4-pentylmorpholiniumbromid oder 1- Methyl-1-pentylpiperidiniumbromid.
